Goal of the work was to evaluate the differences level among H. influenzae strains of b serotype isolated from children attending day-care centres and orphanages and among strains isolated from invasive infections. In the work PFGE in Hib strains transmission examination and for epidemiological studies among three sources of invasive infection was applied. Among 35 Hib strains tested and control strain 8 different pulsotyped were found. Among 21 strains colonising the nasopharynx of healthy children, and among 13 Hib strains isolated from cerebrospinal fluid, 6 and 1 pulsotypes were found, respectively. Results obtained show that healthy children might be colonizing with genotypes characteristic for Hib strains isolated from invasive infections. In this view wider Hib vaccination seems be expected, as Hib circulation is common. 相似文献

Levels of indolic and phenylpropanoid secondary metabolites in Arabidopsis (Arabidopsis thaliana) leaves undergo rapid and drastic changes during pathogen defense, yet little is known about this process in roots. Using Arabidopsis wild-type and mutant root cultures as an experimental system, and the root-pathogenic oomycete, Pythium sylvaticum, for infections, we analyzed the aromatic metabolite profiles in soluble extracts from uninfected and infected roots, as well as from the surrounding medium. A total of 16 indolic, one heterocyclic, and three phenylpropanoid compounds were structurally identified by mass spectrometry and nuclear magnetic resonance analyses. Most of the indolics increased strongly upon infection, whereas the three phenylpropanoids decreased. Concomitant increases in both indolic and phenylpropanoid biosynthetic mRNAs suggested that phenylpropanoids other than those examined here in "soluble extracts" were coinduced with the indolics. These and previous results indicate that roots differ greatly from leaves with regard to the nature and relative abundance of all major soluble phenylpropanoid constituents. For indolics, by contrast, our data reveal far-reaching similarities between roots and leaves and, beyond this comparative aspect, provide an insight into this highly diversified yet under-explored metabolic realm. The data point to metabolic interconnections among the compounds identified and suggest a partial revision of the previously proposed camalexin pathway. 相似文献

The new enantiopure complexes [LnL](NO3)3 · nH2O (Ln = Dy+3, Ho+3, Er+3, Lu+3) and [LnL]Cl3 · nH2O (Ln = Nd+3, Sm+3, Gd+3, Tb+3, Dy+3, Ho+3, Er+3, Tm+3, Lu+3) of the chiral macrocycle L derived from (1R,2R)-1,2-diaminocyclohexane and 2,6-diformylpyridine have been synthesised. The preference of macrocycle L for the heavier lanthanide(III) ions has been established on the basis of competition reaction. The complexes have been characterised by NMR spectroscopy and mass spectrometry. 1H NMR signals of deuterated water solutions of the Ce+3, Nd+3 and Eu+3 complexes have been assigned on the basis of the COSY and HMQC spectra, and for the remaining lanthanide complexes the signals were assigned on the basis of linewidths analysis. The paramagnetic shifts of the series of lanthanide complexes [LnL](NO3)3 · nH2O and [LnL]Cl3 · nH2O have been analysed using both crystal-field dependent and independent methods in order to separate contact and dipolar contributions and establish isostructurality along the series of lanthanide complexes in solution. The data obtained for nitrate derivatives in organic solvent indicate rather irregular deviations from the plots based on those methods, while the plots obtained for water solutions show the characteristic brake in the middle of the lanthanide series, that is interpreted as a result of change of the number of axially coordinated water molecules. The apparent inconsistencies of results obtained on the basis of crystal-field independent method are discussed. 相似文献

The shells of Lymnaea stagnalis show great morphological variability. This phenomenon has been described as the result of an environmental influence. The main object of the present study was to compare some biometric data from shells of naturally infected and uninfected snails from 25 different lakes in the central part of Poland. The height of the shell, the height of the spiral, and the width of the shell were measured. Some inter- and intrapopulation differences among individuals were found. Greater variability of shell shape was observed among snails parasitized with digenean larvae than in nonparasitized ones. Snails infected with Echinoparyphium aconiatum, Echinostoma revolutum, Diplostomum pseudospathaceum, and Opisthioglyphe ranae differed in shell shape compared with uninfected individuals. Snails infected with Plagiorchis elegans did not differ from uninfected individuals. The same was true of snails in which the commensal oligochaete, Chaetogaster limnei, was found. The results of the present study support the assumption that the deformation of shells of the snails under study was in some way influenced by the presence of certain species of digenetic trematodes. 相似文献

The aliphatic amino diacid alpha-aminosuberic acid can function as an effective, stable mimic of the hydrolysis-susceptible Tyr(SO3H) group in sulfakinin neuropeptide analogs for both hindgut contractile activity in cockroach and food intake-inhibition activity in the desert locust. In the analog, the acidic sulfate group is replaced with an acidic carboxyl group. The degree of activity of sulfakinin analogs is correlated with the carboxyl/alpha-carbon distance in the cockroach hindgut contractile assay. The results represent an important step in the design and synthesis of biostable, sulfakinin analogs that could potentially suppress the feeding behavior of destructive insect pests of agricultural importance. 相似文献

Expression of the noradrenaline transporter (NAT) was examined in normal human adrenal medulla and phaeochromocytoma by using
immunohistochemistry and confocal microscopy. The enzymes tyrosine hydroxylase (TH) and phenylethanolamine N-methyltransferase
(PNMT) were used as catecholamine biosynthetic markers and chromogranin A (CGA) as a marker for secretory granules. Catecholamine
content was measured by using high performance liquid chromatography (HPLC). In normal human adrenal medulla (n=5), all chromaffin cells demonstrated strong TH, PNMT and NAT immunoreactivity. NAT was co-localized with PNMT and was located
within the cytoplasm with a punctate appearance. Human phaeochromocytomas demonstrated strong TH expression (n=20 samples tested) but variable NAT and PNMT expression (n=24). NAT immunoreactivity ranged from absent (n=3) to weak (n=10) and strong (n=11) and, in some cases, occupied an apparent nuclear location. Unlike the expression seen in normal human adrenal medullary
tissue, NAT expression was not consistently co-localized with PNMT. PNMT also showed highly variable expression that was poorly
correlated with tumour adrenaline content. Immunoreactivity for CGA was colocalized with NAT within the cytoplasm of normal
human chromaffin cells (n=4). This co-localization was not consistent in phaeochromocytoma tumour cells (n=7). The altered pattern of expression for both NAT and PNMT in phaeochromocytoma indicates a significant disruption in the

Thus far, identification of functionally important residues in Type II restriction endonucleases (REases) has been difficult using conventional methods. Even though known REase structures share a fold and marginally recognizable active site, the overall sequence similarities are statistically insignificant, unless compared among proteins that recognize identical or very similar sequences. Bsp6I is a Type II REase, which recognizes the palindromic DNA sequence 5′GCNGC and cleaves between the cytosine and the unspecified nucleotide in both strands, generating a double-strand break with 5′-protruding single nucleotides. There are no solved structures of REases that recognize similar DNA targets or generate cleavage products with similar characteristics. In straightforward comparisons, the Bsp6I sequence shows no significant similarity to REases with known structures. However, using a fold-recognition approach, we have identified a remote relationship between Bsp6I and the structure of PvuII. Starting from the sequence–structure alignment between Bsp6I and PvuII, we constructed a homology model of Bsp6I and used it to predict functionally significant regions in Bsp6I. The homology model was supported by site-directed mutagenesis of residues predicted to be important for dimerization, DNA binding and catalysis. Completing the picture of sequence–structure–function relationships in protein superfamilies becomes an essential task in the age of structural genomics and our study may serve as a paradigm for future analyses of superfamilies comprising strongly diverged members with little or no sequence similarity. 相似文献
