SYNOPSIS. Protostelium mycophaga Olive and Stoianovitch was isolated from 7 of 19 plant sources collected in a 15-mile radius of Syracuse, New York. Two of these isolations were made from up-right, nondecomposed plants. Spore diameter measurements of 4 of the isolates gave values that agree with those published for this species. The optimum temperature for the growth of P. mycophaga on enriched corn meal agar and Rhodotorula mucilaginosa was 20 C. Limited growth occurred at 10 C and no growth was observed above 30 C.

Due to their ability for direct electron transfer to electrodes, the utilization of rare earth metals as cofactor, and their periplasmic localization, pyrroloquinoline quinone‐dependent alcohol dehydrogenases (PQQ‐ADHs) represent an interesting class of biocatalysts for various biotechnological applications. For most biocatalysts protein stability is crucial, either to increase the performance of the protein under a given process condition or to maximize robustness of the protein towards mutational manipulations, which are often needed to enhance or introduce a functionality of interest. In this study, we describe a whole‐cell screening assay, suitable for probing PQQ‐ADH activities in Escherichia coli BL21(DE3) cells, and use this assay to screen smart mutant libraries for increased thermal stability of the PQQ‐ADH PedE (PP_2674) from Pseudomonas putida KT2440. Upon three consecutive rounds of screening, we identified three different amino acid positions, which significantly improve enzyme stability. The subsequent combination of the beneficial mutations finally results in the triple mutant R91D/E408P/N410K, which not only exhibits a 7°C increase in thermal stability but also a twofold increase in residual activity upon incubation with up to 50% dimethyl sulfoxide (DMSO), while showing no significant difference in enzymatic efficiency (kcat/KM).  相似文献

1. Locomotor performance of limbless vertebrates depends on the substrate through which individuals move and may result in selection on vertebral number in different habitats. To evaluate the effect of push-point density on snake locomotion, the density of vegetation and other potential push-points was quantified at two sites in California (coastal and inland), where conspecific snakes differed greatly in vertebral number (230 and 256 average total vertebrae, respectively; Arnold 1988). The coastal site had significantly higher push-point densities than the inland site.
2. Five experimental push-point densities that fell within the natural range of push-point densities were employed in laboratory trials of juvenile snake locomotion. Density of push-points significantly affected both crawling speed and head-to-tail distance (HTD), an indirect measure of lateral bending. The fastest speed was achieved at an intermediate push-point density. The shortest HTD occurred when snakes moved through the lowest push-point density.
3. Sex, total number of vertebrae and total length significantly affected HTD, regardless of push-point density. Snakes with relatively more vertebrae had a shorter HTD, suggesting they were able to achieve greater lateral bending than snakes with fewer vertebrae. Coastal and inland populations did not differ in HTD during locomotion.
4. Numbers of body and tail vertebrae significantly influenced speed at different push-point densities. In general, snakes with more body vertebrae were slower than those with fewer, while snakes with more tail vertebrae were faster than those with fewer. Snakes of greater total length were faster at all densities. Coastal snakes crawled faster than inland snakes at all push-point densities.  相似文献

Practicing phylogenetic systematics as a sophisticated falsification research program provides a basis for claiming increased knowledge of sister species relationships and synapomorphies as evidence for those cladistic propositions. Research in phylogenetic systematics is necessarily cyclic, and the place where the positive shift in understanding occurs is subsequent to discovering the most parsimonious cladogram(s). A priori differential character weighting is inconsistent with seeking the maximally corroborated cladogram (sensu Popper), because weighting adds to background knowledge, the evidence being then less improbable than it would be otherwise. Also, estimating weights from character state frequencies on a cladogram is inconsistent with the view that history is unique. Sophisticated falsification provides the place in the cycle of phylogenetic systematic research where weight of evidence can be evaluated and these inconsistencies do not apply. On balance, phylogenetic systematics appears to achieve greater coherence and generality as a result of focusing on the foundations for claiming increased knowledge and avoiding efforts to differentially weight characters.  相似文献

Abstract: We used a 60-yr forest simulation of the Cherokee National Forest, Tennessee, USA, to model the effects of timber harvest and natural disturbance upon habitat availability for 6 songbird species: Acadian flycatcher (Epidonax virescens), blue-headed vireo (Vireo solitarius), chestnut-sided warbler (Dendroica pensylvanica), tufted titmouse (Parus bicolor), yellow-billed cuckoo (Coccyzus americanus), and yellow-throated warbler (Dendroica dominica). Forest simulations, based on expected harvest intensities and historic levels of natural disturbance, were used to update a stand inventory database at 10-yr intervals between 1993 and 2053. Habitat models for the 6 bird species were applied to the updated stand inventory and available habitat quantified for each decade. Late-successional species showed substantial increases in habitat availability over the 60-yr period at most harvest intensities, whereas habitat for early-successional species was stable or declined at most harvest intensities. Acadian flycatcher, yellow-throated warbler, and blue-headed vireo habitat increased by 200%, 213%, and 40%, respectively, whereas tufted titmouse habitat remained relatively constant at expected harvest levels. Chestnut-sided warbler habitat was stable at expected harvest levels but declined at lower harvest intensities, and yellow-billed cuckoo habitat declined by 37% at expected harvest levels. Natural disturbance had little effect on habitat availability for any bird species compared to the effects of timber harvests and increasing forest age. Our models suggest that anthropogenic disturbance, and lack thereof, can play a definitive role determining habitat availability and population viability for forest songbirds.  相似文献

ABSTRACT Brood:pair ratios could provide an economical method for assessing spatial or temporal variation in waterfowl productivity, but such estimators are severely biased by incomplete detection of broods. We conducted 3 sequential counts of 1,357 waterfowl broods in northeastern North Dakota, USA, and used closed-population mark-recapture models to estimate total brood abundance while controlling for variation in detection probabilities (p). Blue-winged teal (Anas discors) broods had the lowest average detection probability (p = 0.305), whereas diving-duck broods had the highest average detectability (p = 0.571). Detection was generally highest in morning or evening, but temporal patterns varied among species and there was no survey window that maximized detection probabilities for all species. Detection probabilities averaged 0.108 (SD = 0.056) higher for an experienced observer versus an inexperienced observer. Detection probabilities were 0.044 higher for roadside versus walk-up surveys and increased with increasing brood size, total brood abundance, survey date, wind speed, temperature, cloud cover, and amount of time spent surveying each wetland. Detection probabilities declined with increasing wetland size and amount of tall peripheral vegetation. Our mark-recapture results indicated that a traditional unreplicated brood survey would have missed 67.5% of estimated broods, summed over all species. Use of closed-population mark-recapture techniques provided an effective method for reducing this bias and identifying and quantifying factors that reduce detection probabilities of waterfowl broods. We recommend that future brood surveys incorporate 2 or 3 temporally segregated replicate counts to allow for formal estimation of detection probabilities.  相似文献
