Basidiomycete mushrooms are a rich source of unique substances, including lectins, that could potentially be useful in biotechnology
or biomedical applications. Lectins are a group of carbohydrate-binding proteins with diverse biological activities and functions.
Here, we demonstrate the presence of a number of lectins in the basidiomycete mushroom Clitocybe nebularis. Glucose-, galactose-, sucrose-, lactose-, and Sepharose-binding lectins were isolated from fruiting bodies using affinity
chromatography on Sepharose-immobilized sugars or on Sepharose. The lectins were characterized biochemically and their binding
specificities examined by agglutination and agglutination inhibition assays. In addition, insecticidal and anti-nutritional
properties of the lectins were studied against a model organism, fruit fly (Drosophila melanogaster), and Colorado potato beetle (Leptinotarsa decemlineata). Of the several basidiomycete mushrooms screened, C. nebularis extract showed the most potent insecticidal activity. Sucrose-binding lectin showed the strongest activity against D. melanogaster, followed by lactose- and galactose-binding lectins. Feeding bioassays with Colorado potato beetle revealed that C. nebularis extract exhibited high anti-nutritional activity against the insect; and of those tested, only lactose-binding lectin, named

Background
Lectins are a diverse group of carbohydrate-binding proteins exhibiting numerous biological activities and functions.Methods
Two-step serial carbohydrate affinity chromatography was used to isolate a lectin from the edible mushroom clouded agaric (Clitocybe nebularis). It was characterized biochemically, its gene and cDNA cloned and the deduced amino acid sequence analyzed. Its activity was tested by hemagglutination assay and carbohydrate-binding specificity determined by glycan microarray analysis. Its effect on proliferation of several human cell lines was determined by MTS assay.Results
A homodimeric lectin with 15.9-kDa subunits agglutinates human group A, followed by B, O, and bovine erythrocytes. Hemagglutination was inhibited by glycoprotein asialofetuin and lactose. Glycan microarray analysis revealed that the lectin recognizes human blood group A determinant GalNAcα1–3(Fucα1–2)Galβ-containing carbohydrates, and GalNAcβ1–4GlcNAc (N,N'-diacetyllactosediamine). The lectin exerts antiproliferative activity specific to human leukemic T cells.Conclusions
The protein belongs to the ricin B-like lectin superfamily, and has been designated as C. nebularis lectin (CNL). Its antiproliferative effect appears to be elicited by binding to carbohydrate receptors on human leukemic T cells.General significance

The Eurasian lynx Lynx lynx (Linnaeus 1758) is an opportunistic predator that usually selects the smallest ungulate available. Its diet varies considerably
among different regions; therefore it is important to study lynx diet in different parts of the species’ range. We studied
the diet of lynx from the endangered Dinaric population in Slovenia and Croatia by analyzing lynx scats, prey remains, and
stomach contents. Dinaric lynx mainly killed European roe deer Capreolus capreolus (0.64 frequency of occurrence, 79% of all consumed biomass), which were used more frequently during winter and spring. Ungulates
were killed more often by adult males than by lynx of other age and sex groups. In contrast to studies from other regions,
lynx in the northern Dinaric Mountains also frequently fed on the edible dormice Glis glis (0.18 frequency of occurrence, 7% of all consumed biomass). This large rodent appears to be an important alternative prey,
especially for females and young lynx, and was the reason for the highest use of rodents reported so far for the Eurasian
lynx. Edible dormice in Dinaric forests have highly variable numbers of active animals. Seasonal and possibly annual variation
in dormouse availability obviously affects lynx diet. This is a rare example where variability in the availability of the
alternative prey and not the preferred prey leads to the dietary shift. This study confirms the opportunistic nature of Eurasian

In recent years, the red fox (Vulpes vulpes) has increasingly occupied urban areas in central Europe. Meanwhile, prevalence of infection in foxes with the small fox tapeworm (Echinococcus multilocularis) has increased, thereby increasing the human risk of infection with the parasite, which causes alveolar echinococcosis. Baiting strategies to counteract E. multilocularis have been implemented in cities and the open countryside, but there are few data on the situation in villages and small towns (<10,000 inhabitants). We recorded disappearance rates of praziquantel-containing bait within villages and small towns and on settlement edges for 7 days after distribution. Disappearance rates were 89.2% in villages, 88.8% in small towns, and 91.8% in settlement edges. More than 75% of the bait was consistently taken within the first three nights. There were no significant differences in disappearance rates between years or among seasons (Cox proportional hazard model). The survival time of the bait in small towns (P=0.021) and villages (P=0.026) depended on the zone (zone 1, first row of houses bordering on open countryside; zone 2, second to fourth rows, zone 3; beyond the fifth row) in which bait was distributed. In villages, the probability of bait being eaten in zone 1 was 119% higher than it was in zone 3 (P=0.007). In small towns, the probability was 60% higher (P=0.006). 相似文献

In the context of the paradoxical ubiquity of sex, we tested whether stable coexistence of sexual and asexual fish of the genus Cobitis is mediated by parasites, as asexual fish suffer more from parasitic infections because of their lower genetic variability [the Red Queen hypothesis (RQH)], or by partial niche shift of the two strains differing in mode of reproduction. We did not find a clear correlation between infection risk with a helminth parasite and the proportion of sexuals, and we found similar infection rates among sexual females and co‐occurring asexuals in general, including the most frequent clone in particular. These results suggest that the mechanisms of the RQH are not directly engaged in stabilizing this asexual complex. On the other hand, the temporally stable gradient in sexual/asexual proportions along the river correlated with gradients in environmental parameters (physicochemical water parameters, velocity, and shading of the habitat) and turnover in the fish assemblage structure. Sexual and asexual forms thus appear to prefer different habitats. Theoretical probabilistic kinetic model has been applied to describe the measurements of several oxidation markers as a function of time, during slow oxidation of low-density lipoprotein (LDL). It has been demonstrated that such a process could be described as tocopherol-mediated peroxidation (TMP), initiated and sustained by the action of copper ions, present in LDL in trace amounts. In that process concentration of alpha-tocopherol remains essentially unaltered. Tocopherol and copper ions act as catalysts, oscillating between the oxidized and reduced states. The fitting of the theoretical model to the experimental data resulted in determination of the numerical values for the kinetic parameters. It has been found that the parameter values used for the fitting of the data collected for a number of samples from various donors differ rather little. The kinetic chain length of 1.3 (in presence of co-antioxidants) and 2.9 (in the absence of co-antioxidants) is shorter than found by others. The difference probably comes from the much lower concentration of copper ions in our systems (about 0.1 ion per LDL particle). 相似文献

It has been shown that adult human Leydig cells express a number of neuroendocrine markers, and, therefore, could be considered as a part of the neuroendocrine system in the adult. A limited number of studies have dealt with the dynamics of development of human foetal Leydig cells, whereas studies on their neuroendocrine nature are still extremely rare. Therefore, the aim of our study was to investigate the development of human foetal Leydig cells in different weeks of gestation (wg) and to check if these cells express certain markers characteristic of the diffuse neuroendocrine system (DNS). Qualitative, quantitative histological studies and immunohistochemical analyses of human foetal testicular tissue have been performed. According to our data, Leydig cells formed a dynamic population of cells within the interstitum of testes in the period between 15 and 36 wg. The total number of Leydig cells of human foetal testes changed through different stages of gestation by means of 'pulsatile' dynamics (most likely, by following the variable level of gonadotropins). At early stages of development (15-17 wg) immunohistochemical reactions for the expression of neuron specific enolase (NSE) were positive within sex cords and between them, in the interstitum. Pro-spermatogonia in the sex cords were positive, as well as elongated spindle-shaped cells of the interstitum (very likely, precursors of Leydig cells). During the later stages of development (28-36 wg), excluding the pro-spermatogonia, the interstitial Leydig cells were also positive. The results of the immunohistochemical analyses (the expression of NSE) confirmed the hypothesis that human foetal Leydig cells were of neuroendocrine nature. 相似文献
