Meningiomas are the second most common intracranial tumours. Most meningiomas grow slowly; however, atypical and anaplastic meningiomas show an aggressive biological behaviour. Overexpression of growth factors is considered to be a cause of carcinogenesis. Midkine and pleiotrophin are heparin-binding growth factors that promote growth, survival, migration and differentiation of various target cells. Both molecules are highly expressed during human embryogenesis but are rarely seen in the adult. We show that in relation to normal dura and arachnoid tissues, midkine was overexpressed in meningiomas on the mRNA and protein level, whereas pleiotrophin was not. Thereby, not only the intact but also the truncated form of midkine could be observed. The expression of midkine receptors was variable in different samples. Midkine stimulation of cultured meningioma cells induced phosphorylation of Akt, whereas no increase in phosphorylation of p42/44 MAPK or p38 MAPK could be detected. Midkine did not influence the proliferation of meningioma cells in vitro, but it did protect meningioma cells from camptothecin-mediated apoptotic cell death through reduction in the amounts of active caspase-3. These findings provide evidence for the overexpression of midkine in meningiomas which contributes to protection from cell death in these second most common intracranial tumours.  相似文献

Many of the risk factors for cerebrovascular disease and atherosclerosis also increase the risk of Alzheimer's disease, characterized by the cerebral deposition of beta-amyloid plaques resulting from the abnormal processing of the transmembrane amyloid precursor protein (APP). The initiating event of cholesterol-induced atherosclerosis is the retention and accumulation of atherogenic apolipoprotein B (apoB) together with low-density lipoproteins in the vascular intima. Biglycan, a member of the small leucine-rich protein family, was suspected of contributing to this process. The individual and combined overexpressions of biglycan and apoB-100 were therefore examined on the cortical APP mRNA levels of transgenic mice by means of semiquantitative PCR. As compared with the control littermates, transgenic biglycan mice had significantly increased cortical APP695 (122%) and APP770 (157%) mRNA levels, while the double transgenic (apoB(+/-)xbiglycan(+/-)) mice did not exhibit any changes. These results provide the first experimental evidence that the atherogenic risk factor biglycan alters APP splicing and may participate in the pathogenesis of both Alzheimer and vascular dementias.  相似文献

The aim of our study was to investigate the effect of maternal obesity on the quality and developmental capabilities of in vivo-derived preimplantation embryos. A two-generation dietary model, based on mice overfeeding during intrauterine and early postnatal development, was used to produce four types of female animals: with physiological (7%–8%), slightly elevated (8%–11%), highly elevated (>11%), and low (<7%) amounts of body fat. Spontaneously ovulating females (5–6 weeks old) were mated with male animals and subjected to embryo isolation at Day 4. Stereomicroscopical evaluation of collected embryos showed that the amount of maternal body fat did not affect the average number of collected embryos per dam. However, significant differences were found in the stage-distribution of isolated embryos: dams with highly elevated body fat and dams with low fat delivered decreased numbers of blastocysts and increased numbers of lower-stage or degenerated embryos compared with dams with physiological or slightly elevated fat value. Fluorescence staining showed that blastocysts isolated from dams with high and low percentage of body fat contained significantly higher numbers of dead cells. Most of such dead cells were of apoptotic origin. In contrast, the amount of maternal body fat did not affect blastocyst growth—the average numbers of cells per blastocyst were comparable in all groups. In conclusion, highly elevated or decreased amount of maternal body fat slowed down the development and negatively affected the quality of naturally in vivo-derived preimplantation embryos. No negative effect of slightly elevated fat was observed.  相似文献

Phenolic compounds from plants are known for their antioxidant properties and have been proposed as therapeutic agents to counteract oxidative stress. However, under normal circumstances, the body only receives a very small amount of these substances in the diet. We have investigated the effect of extracts from known and frequently used plants as part of diet, food seasoning, medicinal tea, and sweetener at different concentrations on the ability to scavenge free radicals, to affect antioxidant enzymes, and finally in the survival of cancer cell lines. We found extract concentrations of about 100 μg.ml?1 more indicative in the assessment of all parameters investigated. Ginseng possessed a very good ability to scavenge superoxide and hydroxyl radicals, while stevia also manifested significant effects against hydroxyl radicals. Both extracts also showed NO decomposition ability. The antioxidant defense system against the excessive production of radicals in mitochondria was sufficient. In contrast, the range of operating concentrations for sage and oregano mainly presented no significant effects against reactive oxygen and nitrogen species. Taken together with the significantly reduced activity of glutathione peroxidase, this led to the depletion of glutathione. The demonstrated modulation of redox state capability was sufficient to affect the viability of all tested cancer cell lines, but especially A-549, CEM and HeLa by oregano extract. Results support the promising role of the tested extracts as a source of compounds for further in vivo studies with the ability to powerfully interfere with or modify the redox state of cells according to the type of disease, which is expected to be associated with oxidative stress.  相似文献

Severe malaria occurs predominantly in young children and immunity to clinical disease is associated with cumulative exposure in holoendemic settings. The relative contribution of immunity against various stages of the parasite life cycle that results in controlling infection and limiting disease is not well understood. Here we analyse the dynamics of Plasmodium falciparum malaria infection after treatment in a cohort of 197 healthy study participants of different ages in order to model naturally acquired immunity. We find that both delayed time-to-infection and reductions in asymptomatic parasitaemias in older age groups can be explained by immunity that reduces the growth of blood stage as opposed to liver stage parasites. We found that this mechanism would require at least two components – a rapidly acting strain-specific component, as well as a slowly acquired cross-reactive or general immunity to all strains. Analysis and modelling of malaria infection dynamics and naturally acquired immunity with age provides important insights into what mechanisms of immune control may be harnessed by malaria vaccine strategists.  相似文献

During an experiment carried out in 2009–2010 we observed different population densities of Typhlodromus pyri in three monitored pear cultivars in Organic Pest Management (OPM) as well as Integrated Pest Management (IPM) orchards. In both years the population density of T. pyri was the highest in the cultivar Conference (organic orchard). The lowest population density was found in 2009 on the cultivar Dicolor (IPM orchard) and in 2010 on the cultivar Bohemica. Factors involved are discussed.  相似文献

The Src family kinase/Focal Adhesion Kinase (FAK) complex is a signaling platform playing a crucial role in transformation downstream of oncogenic growth factor receptors. In the case of melanoma in Xiphophorus fish, the oncogenic EGF receptor orthologue Xiphophorus melanoma receptor kinase (Xmrk) effects continuous activation of the Src family kinase Fyn, but not of the other family members Src or Yes. Here, Fyn is strongly involved in promoting many tumorigenic events. Although Fyn is expressed in most mammalian tissues, there are only few reports of its involvement in the development of solid tumors. To find out whether the prominent role of Xiphophorus Fyn is based on an altered binding to its important binding partner FAK when compared to its mammalian Fyn counterparts, we performed yeast-two-hybrid analyses. We compared Xiphophorus and murine Fyn with respect to their binding to full-length and truncated FAK constructs. We found that interaction with FAK occurs similarly for Xiphophorus and mouse Fyn. Both phosphorylated FAK residue Y397 and FAK proline-rich domain are involved in Fyn binding. We also found interaction of FAK and Fyn in human melanoma cell lines. These data suggest a possible, yet unrecognized role of Fyn in the tumorigenesis of human melanoma, too.  相似文献

A long-living (of up to several years) bipartite system was constructed between the unicellular green alga Chlamydomonas reinhardtii and the ascomycetous fungus Alternaria infectoria. The metabolic cooperation between the two organisms was tested with infecting A. infectoria hyphae into nitrogen starving yellow C. reinhardtii culture. After the infection, a slow greening process of the algal cells was observed, which was studied by measuring the increasing chlorophyll content, the appearance of chlorophyll-protein complexes – using 77 K fluorescence spectroscopy, and the measurement of photosynthetic oxygen production. Transmission electron microscopy and laser scanning microscopy images showed that no direct physical contacts were formed between the algal cells and the hyphae in the long-living symbiosis but they were joint in a mucilaginous bed allowing diffusion processes for metabolic cooperation. The increased free amino acid content of the medium of the long-living bipartite cultures’ indicated possible nitrogen supply of hyphal origin, which allowed the re-greening of the algal cells. The results of this work underline the importance of symbiosis-like stable metabolic coexistence, which ensures survival under extreme environmental conditions.  相似文献

Green tea extract (EFLA85942) is able to induce specifically the neutral endopeptidase (NEP) activity and to inhibit the proliferation of SK-N-SH cells; the angiotensin-converting enzyme (ACE) activity is not influenced under the same conditions. The treatment of the cells with arabinosylcytosine and green tea extract results in a strong enhancement of cellular NEP activity whereas cellular ACE activity was not changed significantly, indicating a green tea extract-specific regulation of NEP expression. Because of its role in the degradation of amyloid beta peptides this enzyme induction of NEP by long term treatment with green tea extract may have a beneficial effect regarding the prevention of forming amyloid plaques.  相似文献
