Summary This study provides epidemiological data on acute infantile (ASMA) and chronic childhood spinal (CSMA) muscular atrophy in Warsaw for the period 1976–1985. All calculations are based on the assumption that ASMA and CSMA result from mutations at two different gene loci. The incidence of ASMA and CSMA was 1 in 19474 live births with a corresponding gene and carrier frequency of 714 × 10–5 and 1 in 70, respectively. The prevalance of CSMA for the year 1985 was 1.26 × 10–5. These figures are higher than in similar studies in other countries. This fact might be connected with the careful ascertainment in this study.  相似文献

Hydrolase activities of extracts from different parts of the bodies of parasitic (Theromyzon tessulatum, Piscicola geometra) and predatory (Erpobdella octoculata, Glossiphonia complanata) leeches were examined. The highest activity was detected in the extracts from sections containing the intestine. Hydrolase activities in the crop and intestine of parasitic leeches were higher than in predatory leeches. The high activity of most of hydrolases in those segments may indicate the intensity of digestion and absorption processes in leeches. A lack of trypsin activity and low chymotrypsin activity are likely to result from the presence of inhibitors of these enzymes. The high activity of the majority of the analyzed hydrolases in extracts derived from the head segment of predatory leeches enables, through digestion of tissues, their easy access to physiological fluids of a host. In turn, in extracts from the head segment of predatory leeches, only four hydrolases were shown to be active. Lipase activity was not found in any of the samples, while α-galactosidase activity was found only in extracts from the head segment of T. tessulatum and P. geometra. Trypsin activity was detected in the extract from the intestine contents of H. sanguisuga and in the extract from the head segment of P. geometra. The results demonstrate the presence of majority of hydrolases occurring in other animals in the alimentary tract of leeches. The study also shows that the crop of leeches is not only a food reservoir, but also the site where digestion and absorption processes take place.  相似文献

The immune response triggers a complicated sequence of events, one of which is release of the cytokine tumor necrosis factor-α (TNF-α) from stromal cells, for example monocytes and macrophages. In this work we investigated the biophysical effects of TNF-α on endothelial cells (ECs), including changes in cell morphology, biomechanics, migration, and cytoskeletal dynamics. We found that TNF-α induces a wide distribution of cell area and aspect ratio, with these properties increasing on average during treatment. Interestingly, aspect ratio peaks after approximately 10?h of exposure to TNF-α, corresponding also to a peak in exerted traction forces. Meanwhile, ECs treated with TNF-α soften, and we associate this with significant increases in estimated cellular volume. In addition, our evaluation of migratory dynamics revealed an inverse correlation between cell aspect ratio and migration speed after TNF-α treatment, suggesting that cell shape may be an important functional regulator of EC migration during an inflammatory response. Finally, we addressed the basic mechanics of how the reorganization of F-actin filaments occurs during TNF-α treatment, and observed a dynamic shift of existing actin filaments. Together, our results suggest a functional link between EC morphology, biomechanics, migration, and cytoskeletal dynamics during an inflammatory response.  相似文献

Phylogenetic relationships of (19) serpulid taxa (including Spirorbinae) were reconstructed based on 18S rRNA gene sequence data. Maximum likelihood, Bayesian inference, and maximum parsimony methods were used in phylogenetic reconstruction. Regardless of the method used, monophyly of Serpulidae is confirmed and four monophyletic, well-supported major clades are recovered: the Spirorbinae and three groups hitherto referred to as the Protula-, Serpula-, and Pomatoceros-group. Contrary to the taxonomic literature and the hypothesis of opercular evolution, the Protula-clade contains non-operculate (Protula, Salmacina) and operculate taxa both with pinnulate and non-pinnulate peduncle (Filograna vs. Vermiliopsis), and most likely is the sister group to Spirorbinae. Operculate Serpulinae and poorly or non-operculate Filograninae are paraphyletic. It is likely that lack of opercula in some serpulid genera is not a plesiomorphic character state, but reflects a special adaptation.  相似文献

Sulfate-reducing bacteria (SRB) in anoxic waters and sediments are the major producers of methylmercury in aquatic systems. Although a considerable amount of work has addressed the environmental factors that control methylmercury formation and the conditions that control bioavailability of inorganic mercury to SRB, little work has been undertaken analyzing the biochemical mechanism of methylmercury production. The acetyl-coenzyme A (CoA) pathway has been implicated as being key to mercury methylation in one SRB strain, Desulfovibrio desulfuricans LS, but this result has not been extended to other SRB species. To probe whether the acetyl-CoA pathway is the controlling biochemical process for methylmercury production in SRB, five incomplete-oxidizing SRB strains and two Desulfobacter strains that do not use the acetyl-CoA pathway for major carbon metabolism were assayed for methylmercury formation and acetyl-CoA pathway enzyme activities. Three of the SRB strains were also incubated with chloroform to inhibit the acetyl-CoA pathway. So far, all species that have been found to have acetyl-CoA activity are complete oxidizers that require the acetyl-CoA pathway for basic metabolism, as well as methylate mercury. Chloroform inhibits Hg methylation in these species either by blocking the methylating enzyme or by indirect effects on metabolism and growth. However, we have identified four incomplete-oxidizing strains that clearly do not utilize the acetyl-CoA pathway either for metabolism or mercury methylation (as confirmed by the absence of chloroform inhibition). Hg methylation is thus independent of the acetyl-CoA pathway and may not require vitamin B12 in some and perhaps many incomplete-oxidizing SRB strains.  相似文献

Background

Primary immunodeficiencies are inborn errors of immunity that lead to life threatening conditions. These predispositions describe human immunity in natura and highlight the important function of components of the Toll-IL-1- receptor-nuclear factor kappa B (TIR-NF-κB) pathway. Since the TIR-NF-κB circuit is a conserved component of the host defence in higher animals, genetically tractable models may contribute ideas for clinical interventions.

Methodology/Principal Findings

We used immunodeficient fruit flies (Drosophila melanogaster) to address questions pertaining to survival following bacterial infection. We describe here that flies lacking the NF-κB protein Relish, indispensable for countering Gram-negative bacteria, had a greatly improved survival to such infections when subject to dietary short-term starvation (STS) prior to immune challenge. STS induced the release of Nitric Oxide (NO), a potent molecule against pathogens in flies, mice and humans. Administering the NO Synthase-inhibitory arginine analog N-Nitro-L-Arginine-Methyl-Ester (L-NAME) but not its inactive enantiomer D-NAME increased once again sensitivity to infection to levels expected for relish mutants. Surprisingly, NO signalling required the NF-κB protein Dif, usually needed for responses against Gram-positive bacteria.

Conclusions/Significance

Our results show that NO release through STS may reflect an evolutionary conserved process. Moreover, STS could be explored to address immune phenotypes related to infection and may offer ways to boost natural immunity.  相似文献

Laccase, an enzyme responsible for aerobic transformations of natural phenolics, in industrial applications requires the presence of low-molecular substances known as mediators, which accelerate oxidation processes. However, the use of mediators is limited by their toxicity and the high costs of exploitation. The activation of extracellular laccase in growing fungal culture with highly diluted mediators, ABTS and HBT is described. Two high laccase-producing fungal strains, Trametes versicolor and Cerrena unicolor, were used in this study as a source of enzyme. Selected dilutions of the mediators significantly increased the activity of extracellular laccase during 14 days of cultivation what was distinctly visible in PAGE technique and in colorimetric tests. The same mediator dilutions increased demethylation properties of laccase, which was demonstrated during incubation of enzyme with veratric acid. It was established that the activation effect was assigned to specific dilutions of mediators. Our dose-response dilution process smoothly passes into the range of action of homeopathic dilutions and is of interest for homeopaths.  相似文献

The process of silicification in plants and the biochemical effects of silica in plant tissues are largely unknown. To study the molecular changes occurring in growing cells that are exposed to higher than normal concentration of silicic acid, Raman spectra of germinating pollen grains of three species (Pinus nigra, Picea omorika, and Camellia japonica) were analyzed in a multivariate classification approach that takes into account the variation of biochemical composition due to species, plant tissue structure, and germination condition. The results of principal component analyses of the Raman spectra indicate differences in the utilization of stored lipids, a changed mobilization of storage carbohydrates in the pollen grain bodies, and altered composition and/or structure of cellulose of the developing pollen tube cell walls. These biochemical changes vary in the different species.

The aim of this experiment, conducted under greenhouse conditions, was to assess the influence of various H(2)SeO(3) concentrations added to soil (0.05, 0.15, and 0.45mMkg(-1)) on selenium and adenosine triphosphate (ATP) content, and on the activity of the ascorbate-glutathione cycle enzymes in green parts of wheat and oilseed rape. Selenium uptake by the test plants was found to vary, with content increasing from one developmental stage to the next over four stages of the developmental cycle. At the lowest H(2)SeO(3) dose (0.05mMkg(-1)), the wheat plants took up much more selenium than did the oilseed rape plants, while the amount of selenium taken up at higher doses (0.15 and 0.45mMkg(-1)) was markedly higher in rape. The increasing Se content in the wheat to about 10mgkg(-1) (in the dark) and to about 16mgkg(-1) (in the light) was accompanied by a concurrent increase in the ATP content, which remained unchanged in the light-exposed plants, while clearly decreasing in those kept in the dark. On the other hand, the ATP content of the light-exposed oilseed rape was maintained at a stable level to about 10mg Sekg(-1), following which ATP content was observed to decrease. In contrast, the tendency for the ATP content to decrease appeared immediately in the dark. The increasing plant selenium concentration was accompanied by decreased APX activity in wheat, increased activity in oilseed rape, no major change in the dehydroascorbate reductase (DHAR) activity in oilseed rape and a slight increase in wheat to about 8mg Sekg(-1), followed by a reduction. The glutathione reductase (GR) activity in wheat differed from the activity of DHAR; an increase in the selenium content to about 8mgkg(-1) was accompanied by a distinct reduction, while a significant increase was observed at higher selenium contents; in oilseed rape, the activity was observed to increase slightly within a narrow range of selenium contents (up to 5mgkg(-1)), and to decrease thereafter.  相似文献
