Summary Ustilago maydis was surveyed for cyclic AMP-dependent protein kinase activity. Using a combination of ion-exchange and molecular filtration techniques, we demonstrate that there is only one form of cyclic AMP-dependent protein kinase in the cytosolic fraction of the fungus. The kinase activity is specifically activated by cyclic AMP and utilizes protamine and kemptide as substrates. Most, if not all, of the cyclic AMP binding detected in the soluble fraction is associated with the protein kinase activity. The right ventricles of pig heart were perfused with hypoxic blood and the left ventricles were perfused with normally ventilated arterial blood. Free carnitine and short-chain acylcarnitines in hypoxic ventricles were lower than in perfused controls, and much lower than in non-perfused heart. Acetylcarnitine levels decreased and the branched-chain acylcarnitines and propionylcarnitine were elevated in the hypoxic perfused ventricles. These data indicate that both hypoxia and anaesthesia caused loss of carnitine and short-chain acylcarnitines from the heart and hypoxia also changed the distribution of short-chain acylcarnitines in the heart.  相似文献

DEAE-cellulose chromatography of mycelial extracts of Mucor rouxii grown to mid-exponential phase resolves two types of low-Km cyclic AMP phosphodiesterase (EC 3.1.4.17; PDE): PDE I, highly activatable (4-6-fold) by phosphorylation or proteolysis, and PDE II, unresponsive to activation. The enzymic profile of PDE activity obtained from germlings shows only PDE I activity, whereas PDE activity from mycelia grown to stationary phase is eluted from the DEAE-cellulose column at the position of PDE II, and like PDE II is unresponsive to activation. Endogenous proteolysis or controlled trypsin treatment transforms PDE I into PDE II. The insensitive forms of PDE exhibit a slightly smaller sedimentation coefficient than the activatable forms, as judged by sucrose-gradient centrifugation. The basal activity of the highly activatable form of PDE is elevated almost to the value in the presence of trypsin on storage at 4 degrees C in the absence of proteinase inhibitors. Benzamidine, leupeptin, antipain or EGTA prevents the activation produced by storage. PDE I remains strongly activatable by phosphorylation and proteolysis after resolution by polyacrylamide-gel electrophoresis.  相似文献

Monosodium glutamate (MSG) was used to evaluate the importance of the arcuate nucleus of the hypothalamus in the expression of daily gonadotropin rhythms in female golden hamsters. These daily rhythms of luteinizing hormone (LH) and follicle-stimulating hormone (FSH), which also occur in prepubertal females, are characterized by afternoon surges. Neonatal administration of MSG induces degeneration of perikarya in the arcuate nucleus and renders females permanently anovulatory. MSG was injected at 8 days of age; at 21 days, the animals were weaned and sorted by sex into groups of 5-7. Blood samples were obtained at 1300 and 1700 h at 25, 30, 35, 40, 50, 62, and 192 days of age from MSG-sterilized animals. Saline-injected controls were bled at 25 days and after estrous cycles had been initiated (29-37 days of age). In both control and MSG-injected groups, there was an afternoon surge of LH and FSH at 25 days of age. These daily surges persisted in MSG-injected animals. The ovaries of these animals were characterized by an abundant interstitium and arrested follicular development. Progesterone levels of MSG-anovulatory animals also reflected the rhythmicity of LH and FSH, with a significant increase occurring between 1300 and 1700 h. Thus, MSG did not affect the daily circadian-based rhythmicity in gonadotropin secretion even though adult-age animals were infertile. These results suggest that perikarya of the arcuate nucleus affected by MSG are not required for generation of daily LH and FSH rhythms.  相似文献

Time courses for the effect of electrical stimulation, fasting and sampling mode (anesthesia vs decapitation) on the quantitative distribution of carnitine and its esters in fast white and fast red skeletal muscle fibres of rats were determined. Both fibre types responded similarly to electrical stimulation with respect to changes in acetyl- and propionylcarnitine, but the time course was very different. The proportion of esterified carnitine decreased with fasting and anesthesia in both fibres compared to the fed decapitated group. This shift in the acylation state of carnitine was mainly due to the decrease of acetylcarnitine levels. The data show that the use of anesthetics may induce significant quantitative changes in specific acylcarnitine levels, presumably reflecting changes in specific acylcoenzyme A levels.  相似文献

It is shown from the statistical-mechanical overview of Volterra's ecological model how to reckon the fluctuations of collective variables such as the total population of a genus: and that these fluctuations are much decreased (or that the collective populationsteadiness is enhanced) as the speciation is increased. (A niching of species in time, or phase-niching, is entailed here.) Secondly, it is shown how Preston's log-normal distribution describing the species-abundance relationship, as well as a generalization of such distributions, come forth simply and naturally from the statistical-Volterra-dynamics.  相似文献
