In this study we examined the biosynthesis of abscisic acid (ABA) by developing corn (Zea mays L.) embryos. Three comparisons were made: ABA biosynthesis in embryos isolated from kernels grown in vitro with those grown in the field; the developmental profile of ABA content with that of biosynthesis; and ABA biosynthesis in corn embryos lacking carotenoid precursors with ABA biosynthesis in normal embryos. Embryos were harvested at various times during seed development and divided into two groups. Endogenous levels of ABA were measured in one group of embryos and ABA biosynthetic capacity was measured in the other group. The ABA biosynthetic capacity was measured with and without tetcyclacis (an inhibitor of ABA degradation) in embryos from both field-grown and in-vitro-grown corn kernels. Reduced-carotenoid (either fluridone-treated or genetically viviparous) embryos were also included in the study. Corn kernels developing under field and in-vitro conditions differed from each other in their responses to tetcyclacis and in their profiles of ABA biosynthesis during development. Therefore, in-vitro kernel culture may not be an appropriate substitute for field conditions for studies of embryo development. The developmental profiles of endogenous ABA content differed from those of ABA biosynthesis in isolated embryos of both in-vitro-and field-grown kernels. This indicated that ABA levels in the developing embryos were determined by import from the maternal tissues available to the embryos rather than by in-situ biosynthesis. In embryos with reduced levels of carotenoids, either fluridone-treated or genetically viviparous embryos, ABA biosynthesis was low or nonexistent. The effect of an acute increase in temperature, exhaustive exercise and hypoxia on the haematology of the benthic Antarctic teleost, Trematomus bernacchii was investigated. High temperature and hypoxia caused the biggest changes to the blood, with increases in haematocrit, haemoglobin concentrations and plasma chloride levels. The spleen decreased in mass. Exercise produced the smallest changes. Changes were substantially less than reported for the more active cryopelagic species Pagothenia borchgrevinki. The magnitude of the haematocrit increase is discussed in relation to life-style of fish living in the Antarctic.  相似文献

RasG is the most abundant Ras protein in growing Dictyostelium cells and the closest relative of mammalian Ras proteins. We have generated null mutants in which expression of RasG is completely abolished. Unexpectedly, RasG cells are able to grow at nearly wild-type rates. However, they exhibit defective cell movement and a wide range of defects in the control of the actin cytoskeleton, including a loss of cell polarity, absence of normal lamellipodia, formation of unusual small, punctate polymerized actin structures, and a large number of abnormally long filopodia. Despite their lack of polarity and abnormal cytoskeleton, mutant cells perform normal chemotaxis. However, rasG cells are unable to perform normal cytokinesis, becoming multinucleate when grown in suspension culture. Taken together, these data suggest a principal role for RasG in coordination of cell movement and control of the cytoskeleton.  相似文献

An anaerobic liquid enrichment method followed by plating on a selective medium revealed that the soft rot coliform bacterium Erwinia carotovora subsp. carotovora was generally present in water from drains, ditches, streams, rivers and lakes (including reservoirs) in southern Scotland and in Colorado, United States, in mountainous, upland and arable areas through the year. Many sites were remote from susceptible or diseased crops. Erwinia carotovora subsp. atroseptica was isolated much less frequently and no Erwinia bacteria were isolated from underground waters. Erwinia bacteria were also found in rain-water in Scotland, in winter snow from mountain passes in Colorado, and in sea water from the west and east coasts of Scotland and from the coasts of Oregon, California, Texas, Louisiana and Florida. The significance of the occurrence of these bacteria in water is discussed in relation to the control of blackleg and soft rot diseases of potato by production of Erwinia -free stocks.  相似文献
