Summary A comparison of the protein patterns of the 70S and 80S ribosomes from various plants, E. coli and yeast by disc-gel electrophoresis has shown the following relations: 1. There is a greater similarity between chloroplast ribosomes from various plants than between chloroplast and cytoplasmic ribosomes obtained from the same plant. 2. The protein patterns of the cytoplasmic ribosomes from bean, spinach and tobacco are more similar to each other than when compared to that of wheat germ. 3. At least one band is common to cytoplasmic ribosomes from all plants tested. 4. Only very few bands with identical mobilities are observed between chloroplast and E. coli ribosomes and between cytoplasmic plant and yeast ribosomes.  相似文献

The addition of 0.5 mM salicylic acid (SA) to the hydroponic growth solution of young maize (Zea mays L.) plants under normal growth conditions provided protection against subsequent low-temperature stress. This observation was confirmed by chlorophyll fluorescence parameters and electrolyte leakage measurements. In addition, 1 d of 0.5 mM SA pre-treatment decreased net photosynthesis, stomatal conductivity and transpiration at the growth temperature (22/20 °C). Since there was only a slight decrease in the ratio of variable to maximal fluorescence (Fv/Fm) the decrease in photosynthetic activity is not due to a depression in photosystem II. The analysis of antioxidant enzymes showed that whereas SA treatment did not cause any change in ascorbate peroxidase (EC 1.11.1.11) and superoxide dismutase (EC 1.15.1.1) activities, there was a decrease in catalase (EC 1.11.1.6) activity, and an increase in guaiacol peroxidase (EC 1.11.1.7) and glutathione reductase (EC 1.6.4.2) activities after the 1-d SA treatment at 22/20 °C. In native polyacrylamide gels there was, among the peroxidase isoenzymes, a band which could be seen only in SA-treated plants. It is suggested that the pre-treatment of maize plants with SA at normal growth temperature may induce antioxidant enzymes which lead to increased chilling tolerance. The present study investigated the possible mediatory role of salicylic acid (SA) in protecting plants from cadmium (Cd) toxicity. The exposure of pea plants to increasing Cd concentrations (0.5, 1.0, 2.0 and 5.0 μM) during early stages of their establishment, caused a gradual decrease in shoot and root fresh weight accumulation, the rate of CO2 fixation and the activity of ribulose-1,5-bisphosphate carboxylase (RuBPC, E.C. 4.1.1.39), the effect being most expressed at higher Cd concentrations. In vivo the excess of Cd-induced alterations in the redox cycling of oxygen-evolving centers and the assimilatory capacity of the pea leaves as revealed by changes in thermoluminescence emission after flash illumination. The levels of some important parameters associated with oxidative stress, namely lipid peroxidation, electrolyte leakage and proline production were increased. Seed pretreatment with SA alleviated the negative effect of Cd on growth, photosynthesis, carboxylation reactions, thermoluminescence characteristics and chlorophyll content, and led to decrease in oxidative injuries caused by Cd. The data suggest that the beneficial effect of SA during an earlier growth period could be related to avoidance of cumulative damage upon exposure to cadmium thus reducing the negative consequences of oxidative stress caused by heavy metal toxicity. In addition, the observed high endogenous levels of SA after treatment with Cd suggests that SA may act directly as an antioxidant to scavenge the reactive oxygen species and/or indirectly modulate redox balance through activation of antioxidant responses.Taken together these evidences could explain at some extend the protective role of SA on photochemical activity of chloroplast membranes and photosynthetic carboxylation reactions in Cd-stressed pea plants.  相似文献

Alterations in temperature adaptation processes and changes in the content of stress-related compounds, polyamines and salicylic acid were evaluated in Atnoa1 (NO-associated 1) Arabidopsis mutant. The Fv/Fm chlorophyll-a fluorescence induction parameter and the actual quantum yield were significantly lower in the Atnoa1 mutant than in the wild-type. In the wild-type Col-0, the fastest increase in the non-photochemical quenching (NPQ) occurred in plants pre-treated at low temperature (4 °C), while the slowest was in those adapted to 30 °C. The NPQ showed not only a substantially increased level in the light-adapted state, but also more rapid light induction after the dark-adapted state in the Atnoa1 mutant than in the wild-type. The results of freezing tests indicated that both the wild-type and the mutant had better freezing tolerance after cold hardening, since no significant differences were found between the genotypes. The level of putrescine increased substantially, while that of spermine decreased by the end of the cold-hardening (4 °C, 4 d) period. The quantity of spermidine in Atnoa1 was significantly higher than in Col-0, at both control and cold-hardening temperatures. A similar trend was observed for spermine, but only under control conditions. The mutant plants showed substantially higher salicylic acid (SA) contents for both the free and bound forms. This difference was significant not only in the control, but also in the cold-hardened plants. These results suggest that there is a compensation mechanism in Atnoa1 mutant Arabidopsis plants to reduce the negative effects of the mutation. These adaptation processes include the stimulation of photoprotection and alterations in the SA and polyamine compositions.  相似文献

In the postantibiotic era, available treatment options for severe bacterial infections caused by methicillin-resistant Staphylococcus aureus have become limited. Therefore, new and innovative approaches are needed to combat such life-threatening infections. Virulence factor expression in S. aureus is regulated in a cell density-dependent manner using “quorum sensing,” which involves generation and secretion of autoinducing peptides (AIPs) into the surrounding environment to activate a bacterial sensor kinase at a particular threshold concentration. Mouse monoclonal antibody AP4-24H11 was shown previously to blunt quorum sensing-mediated changes in gene expression in vitro and protect mice from a lethal dose of S. aureus by sequestering the AIP signal. We have elucidated the crystal structure of the AP4-24H11 Fab in complex with AIP-4 at 2.5 Å resolution to determine its mechanism of ligand recognition. A key GluH95 provides much of the binding specificity through formation of hydrogen bonds with each of the four amide nitrogens in the AIP-4 macrocyclic ring. Importantly, these structural data give clues as to the interactions between the cognate staphylococcal AIP receptors AgrC and the AIPs, as AP4-24H11·AIP-4 binding recapitulates features that have been proposed for AgrC-AIP recognition. Additionally, these structural insights may enable the engineering of AIP cross-reactive antibodies or quorum quenching vaccines for use in active or passive immunotherapy for prevention or treatment of S. aureus infections.  相似文献
