We compared the occurrence of peroxidase isozymes in protein extract from roots, hypocotyls and cotyledons of 10 dayCucurbita pepo plants and of adult leaves of older plants by means of starch gel and polyacryl amide gel electrophoresis. We reached maximum discrimination by means of starch gel electrophoresis: 11 zones were ascertained on the cathode side and about 2 on the anode side at pH 3.1. Two zones occurred regularly:A and (the latter having a more complicated structure). ZoneD is characteristic for roots, but is it suppressed and seldom found with leaves. On the other hand zonesC 1 andC 2 are clearly discernible with leaves but are substantially less evident with roots. The character of anodic zoneZ is discussed later in this paper.  相似文献

An investigation was made of the anatomical structure of the shoot apex ofSenecio vulgaris L. a photoperiodically neutral plant, and compared with the formation of successive leaf primordia along the axis up to the initiation of the terminal inflorescence. In the shoot apex of a germinating plant a central zone can first be distinguished from the peripheral zone which is composed of small and intensely stained cells. Later, a rib meristem appears. At the time of the initiation of the middle (the largest) leaves, the shoot apex has a distinct small central zone and a well developed peripheral zone and rib meristem. Between these zones there is a group of cells dividing in all directions, the subcentral zone. At the time of initiation of the last leaves, the central zone extends to the flanks and gradually ceases to be distinguishable. At the same time, the subcentral zone increases in size. This is caused first by cell division and later, with the initiation of the last, most reduced leaves, by enlargement of the cells. Vacuolization in the inner part of the apex and the arrangement of the superficial cells in rows parallel to the surface of the apex, is a preparatory step to the initiation of the inflorescence.  相似文献

Calamagrostis villosa stands occurring in areas deforested by air-pollution impact in the Moravian-Silesian Beskydy Mountains were characterized by a high dry mass of total underground biomass (3 300 g. m?2—the slope site, 2 850 g. m?2—the flat site). The percentage of living roots and rhizomes in total underground biomass was very high (about 70%). The total aboveground biomass was respectively, 321 g.m?2 (the slope site) and 726 g. m?2 (the flat site). In unstabilized habitats on steep slope, the higher plant biomass produced was allocated to a more developed root system.  相似文献

The Epstein-Barr virus (EBV) immediate-early gene product, BRLF1, transactivates the human immunodeficiency virus type 1 (HIV-1) long terminal repeat. BRLF1-induced transactivation of HIV-1 promoter constructs is accompanied by an increase in plasmid mRNA and is reporter gene independent. Previously, BRLF1 transactivation of EBV promoters has been mapped to regions which function as enhancer elements. Deletional analysis demonstrates that BRLF1 transactivation of the HIV-1 promoter does not require the HIV-1 enhancer. Thus, the EBV BRLF1 gene product may transactivate by at least two different mechanisms, one mechanism involving certain enhancer elements and another mechanism which is enhancer independent.  相似文献

The Epstein-Barr virus (EBV) BZLF1 gene product is thought to mediate the disruption of latent EBV infection. We have examined the regulatory effects of BZLF1 by studying its transactivating effects on seven different EBV promoters. We find that whereas the BZLF1 gene product increases the activity of the two early promoters, BMLF1 and BMRF1, it decreases the activity of three latent promoters (the BamHI-C and BamHI-W Epstein-Barr nuclear antigen promoters and the latent membrane protein promoter). The BZLF1-induced changes in promoter-directed chloramphenicol acetyltransferase activity occur in EBV-negative as well as EBV-positive cell lines and are accompanied by a similar change in chloramphenicol acetyltransferase mRNA. Deletion analysis of the BamHI Z fragment indicates that in a portion of the amino-terminal half of the BZLF1 gene product (amino acids 24 to 86) is not essential for positive transactivating effects but is required for down-regulating effects. Thus, different domains of the same EBV immediate-early gene product can either increase the function of EBV promoters active in productive infection or decrease the function of key promoters active in latent infection.  相似文献

Seven young (Y, 22-28 yr) and seven middle-aged (MA, 49-60 yr) normotensive men of similar body size, fatness, and maximal oxygen uptake (VO2max) were exposed to a heat challenge in an environmental chamber (48 degrees C, 15% relative humidity). Tests were performed in two hydration states: hydrated (H, 25 ml water/kg body wt 1 h before the test, 2.5 h before exercise) and hypohydrated (Hypo, after 18-20 h of water deprivation). Each test began with a 90-min rest period during which the transiently increased plasma volume and decreased osmolality after drinking in the H condition returned to base line. This period was followed by 30 min of cycle exercise at a mean intensity of 43% VO2max and a 60-min resting recovery period with water ad libitum. Although prior drinking caused no sustained changes in plasma osmolality, Hypo increased plasma osmolality by 7-10 mosmol/kg in both groups. There were no significant age differences in water intake, urine output or osmolality, overall change in body weight, or sweating rate. In the H state, the percent change in plasma volume was less (P less than 0.01) during exercise for the Y group (-5.9 +/- 0.7%) than for the MA group (-9.4 +/- 0.6%). Esophageal temperature (Tes) was higher in the Hypo condition for both groups with no age-related differences. The p53 tumor suppressor protein, which is commonly mutated in human cancers, has been shown to interact directly with virally encoded from papillomavirus, adenovirus, and simian virus 40. The disruption of p53 function may be required for efficient replication of certain viruses and may also play a role in the development of virally induced malignancies. Infection with Epstein-Barr virus (EBV) has been associated with the development of B-cell lymphomas and nasopharyngeal carcinoma. Here we show that the EBV immediate-early protein, BZLF1 (Z), which is responsible for initiating the switch from latent to lytic infection, can interact directly in vitro and in vivo with the tumor suppressor protein, p53. This interaction requires the coiled-coil dimerization domain of the Z protein and the carboxy-terminal portion of p53. Overexpression of wild-type p53 inhibits the ability of Z to disrupt viral latency. Likewise, Z inhibits p53-dependent transactivation in lymphoid cells. The direct interaction between Z and p53 may play a role in regulating the switch from latent to lytic viral infection.  相似文献

von Hippel–Lindau (VHL) disease is a hereditary tumor syndrome predisposing to multifocal bilateral renal cell carcinomas (RCCs), pheochromocytomas, and pancreatic tumors, as well as angiomas and hemangioblastomas of the CNS. A candidate gene for VHL was recently identified, which led to the isolation of a partial cDNA clone with extended open reading frame, without significant homology to known genes or obvious functional motifs, except for an acidic pentamer repeat domain. To further characterize the functional domains of the VHL gene and assess its involvement in hereditary and nonhereditary tumors, we performed mutation analyses and studied its expression in normal and tumor tissue. We identified germ-line mutations in 39% of VHL disease families. Moreover, 33% of sporadic RCCs and all (6/6) sporadic RCC cell lines analyzed showed mutations within the VHL gene. Both germ-line and somatic mutations included deletions, insertions, splice-site mutations, and missense and nonsense mutations, all of which clustered at the 3' end of the corresponding partial VHL cDNA open reading frame, including an alternatively spliced exon 123 nt in length, suggesting functionally important domains encoded by the VHL gene in this region. Over 180 sporadic tumors of other types have shown no detectable base changes within the presumed coding sequence of the VHL gene to date. We conclude that the gene causing VHL has an important and specific role in the etiology of sporadic RCCs, acts as a recessive tumor-suppressor gene, and appears to encode important functional domains within the 3' end of the known open reading frame.  相似文献

Bacillus megaterium strain B6 producing extracellular β-amylase was isolated and grown in a medium supplemented with waste potato starch. It showed highest enzyme synthesis in the early stationary phase. The partially purified β-amylase had a temperature optimum at 60°C and a pH optimum at 6.9 and was not affected by Schardinger dextrins. Summary Mortierella alpina S-17 cultivated in shaker flasks in the semisynthetic liquid medium with 3% glucose at 28 °C for 14 days accumulated 2.1 g/l of intracellular lipid rich in arachidonic acid (ARA). The content of ARA was 50 % of the total fatty acids. The simple method for high pure methyl arachidonate preparation was developed.  相似文献
