Adding jasmonic acid (JA) to autotrophic, photomixotrophic, or heterotrophic suspension cultures of soybean specifically increased the level of the Mr 30,000 subunit of soybean vegetative storage protein (VSP-30) and a polypeptide at Mr 18,000 that interacted with antibody raised against VSP. Using photomixotrophic cells, the increase was observed at concentrations as low as 10 nM JA and the increase was evident within 2 h following treatment. Below 10 M, JA did not inhibit growth of the cells but did cause browning at higher concentrations. Other plant growth regulators, including abscisic acid (ABA), gibberellic acid, and benzyl adenine, did not alter the level of VSP-30 either in the presence or absence of JA. Methyl jasmonate (JA-Me), 3-oxo-2-butyl-cyclopentane-1-acetate, and 3-oxo-2-pentyl-cyclopentane-1-acetate also increased VSP-30 but at higher concentrations than JA. Altering the level of reduced nitrogen or sucrose in the medium did not alter VSP-30 levels in the cells, but at higher sucrose concentrations, sensitivity to JA was reduced. Effects of sublethal doses of metal (Cu, Cd, Zn) mixtures on the activities of key respiratory enzymes (succinate dehydrogenase, SDH and glyceraldehyde dehydrogenase, GDH) and their recovery following withdrawal of treatments were studied in the freshwater fish O. mossambicus. On the basis of 96 hr LC50 Cu was highly toxic followed by Zn and Cd, and the trimetal combination (Cu+Zn+Cd) was extremely toxic than any other combination; combination of Zn+Cd was least toxic. A significant gradual decrease in SDH with a concomitant increase in GDH activity observed in liver, brain, muscle and gill of animals exposed to metal suggest a metabolic shift from aerobiosis to anaerobiosis due to metal action. Exposed individuals when transferred to metal impoverished water showed an improvement in SDH activity and decline in GDH activity suggesting slow reversal to aerobic metabolism. Prions are proteins that play a central role in transmissible spongiform encephalopathies in a variety of mammals. Among the most notable prion disorders in ungulates are scrapie in sheep, bovine spongiform encephalopathy in cattle, and chronic wasting disease in deer. Single nucleotide polymorphisms in the sheep prion gene (PRNP) have been correlated with susceptibility to natural scrapie in some populations. Similar correlations have not been reported in cattle or deer; however, characterization of PRNP nucleotide diversity in those species is incomplete. This report describes nucleotide sequence variation and frequency estimates for the PRNP locus within diverse groups of U.S. sheep, U.S. beef cattle, and free-ranging deer (Odocoileus
virginianus and O. hemionus from Wyoming). DNA segments corresponding to the complete prion coding sequence and a 596-bp portion of the PRNP promoter region were amplified and sequenced from DNA panels with 90 sheep, 96 cattle, and 94 deer. Each panel was designed to contain the most diverse germplasm available from their respective populations to facilitate polymorphism detection. Sequence comparisons identified a total of 86 polymorphisms. Previously unreported polymorphisms were identified in sheep (9), cattle (13), and deer (32). The number of individuals sampled within each population was sufficient to detect more than 95% of all alleles present at a frequency greater than 0.02. The estimation of PRNP allele and genotype frequencies within these diverse groups of sheep, cattle, and deer provides a framework for designing accurate genotype assays for use in genetic epidemiology, allele management, and disease control. 相似文献

The sulfur present in both agricultural and uncultivated soils is largely in the form of sulfonates and sulfate esters and not as free, bioavailable inorganic sulfate. Desulfurization of the former compounds in vitro has previously been studied in Pseudomonas putida, a common rhizosphere inhabitant. Survival of P. putida strains was now investigated in three sulfur-deficient Danish soils which were found to contain 60 to 70% of their sulfur in sulfonate or sulfate ester form, as determined by X-ray near-edge spectroscopy. The soil fitness of P. putida S-313 was compared with that of isogenic strains with mutations in the sftR and asfA genes (required for in vitro desulfurization of sulfate esters and arylsulfonates, respectively) and in the ssu locus (required in vitro for the desulfurization of both sulfonates and sulfate esters). asfA or sftR mutants showed significantly reduced survival compared to the parent strain in bulk soil that had been enriched with carbon and nitrogen to mimic rhizosphere conditions, but this reduced survival was not observed in the absence of these additives. In a tomato rhizosphere grown in compost, survival of sftR and ssu mutants was reduced relative to the parent strain. The results demonstrate that the ability to desulfurize sulfonates and sulfate esters is critical for survival of bacteria in the rhizosphere but less so in bulk soils outside the influence of plant roots, where carbon is the limiting nutrient for growth. 相似文献

Introduction
Rheumatoid arthritis (RA) is a chronic disease associated with inflammation and destruction of bone and cartilage. Although inhibition of TNFα is widely used to treat RA, a significant number of patients do not respond to TNFα blockade, and therefore there is a compelling need to continue to identify alternative therapeutic strategies for treating chronic inflammatory diseases such as RA. The anti-epidermal growth factor (anti-EGF) receptor antibody trastuzumab has revolutionised the treatment of patients with EGF receptor-positive breast cancer. Expression of EGF ligands and receptors (known as HER) has also been documented in RA. The highly unique compound RB200 is a bispecific ligand trap that is composed of full-length extracellular domains of HER1 and HER3 EGF receptors. Because of its pan-HER specificity, RB200 inhibits responses mediated by HER1, HER2 and HER3 in vitro and in vivo.
