Rachels' subject is the performance of philosophers/ethicists as commentators on public events, such as the decision of a California couple, the Ayalas, to have another child with the hope that the infant's bone marrow cells could be used to save the life of a daughter dying of leukemia. Rachels charges that instead of carefully analyzing new developments and their implications, ethicists often function as defenders of existing social consensus.     

Effect of monensin on receptor recycling during continuous endocytosis of asialoorosomucoid

The binding of asialoglycoproteins to their liver cell receptor results in internalization of the ligand-receptor complex. These complexes rapidly appear in intracellular compartments termed endosomes whose acidification results in ligand-receptor dissociation. Ligand and receptor subsequently segregate: ligand is transported to lysosomes and is degraded while receptor recycles to the cell surface. The proton ionophore monensin prevents acidification of endosomes and reversibly inhibits this acid-dependent dissociation of ligand from receptor. The present study determined the effect of monensin treatment of short-term cultured rat hepatocytes on cell-surface-receptor content, determined both by their binding activity and immunologically, following continuous endocytosis of asialoorosomucoid. Inclusion of 5 microM monensin in the incubation medium reduced the number of immunologically detectable cell-surface receptors by 20% in the absence of ligand. During continuous endocytosis of asialoorosomucoid, inclusion of monensin resulted in a 30-40% reduction of cell-surface receptor detectable either by ligand binding or immunologically. These results suggest that the reduced liver-cell-surface content of receptor in monensin is due to intracellular trapping of ligand-receptor complexes. The reduction of surface receptor during monensin incubation in the absence of ligand suggests that "constitutive recycling" of plasma membrane components also requires intracellular acidification.     

Interference Competition among Coprophilous Fungi: Production of (+)-Isoepoxydon by Poronia punctata

(+)-Isoepoxydon has been established as the major causative agent of interference competition between Poronia punctata (NRRL 6457), a late fungal colonist of cattle dung, and two early-occurring dung colonists, Ascobolus furfuraceus (NRRL 6460) and Sordaria fimicola (NRRL 6459). This compound was isolated from ethyl acetate extracts of liquid cultures of P. punctata by silica gel chromatography and identified by mass spectrometry and proton and carbon nuclear magnetic resonance spectroscopy. The isolation process was guided by in vitro bioassays for antifungal activity against A. furfuraceus and S. fimicola. (+)-Isoepoxydon has been implicated as an intermediate in the biosynthesis of patulin, a mycotoxin produced by Penicillium spp., but no patulin could be detected in cultures of P. punctata.     

The effect of glucocorticoids on the in vivo conversion of androstenedione to oestrone

In vitro studies have previously shown that the activity of the aromatase enzyme system, which is responsible for the conversion of androstenedione to oestrone, can be stimulated by natural and synthetic glucocorticoids and also by ACTH. In view of the potential physiological importance of such a regulatory mechanism we have examined the effect of administration of dexamethasone, and of ACTH on the conversion of androstenedione to oestrone in vivo. The transfer constants for the conversion of androstenedione to oestrone [( rho]AEBU) measured in two women before administration of dexamethasone were 1.0% and 1.1% and after were 0.9% and 1.2%. Similarly no increase in conversion of androstenedione to oestrone [( rho]AE1BB) was detected after ACTH stimulation (pre = 0.74%, post = 0.77%). It is concluded from this study that glucocorticoids and ACTH do not have a role in regulating aromatase activity in vivo.     

Changing patterns of cerium reactivity in developing ovarian oocytes of the hamster

In developing ovarian oocytes of 2-day-old, 12-day-old and adult hamsters, oxidative enzyme activity as demonstrated by the cerium technique is strongest at 12 days and occurs principally in those oocytes which have begun rapid growth. The reaction is associated with the outer mitochondrial membrane, nearby cytoplasm, endoplasmic reticulum and nuclear envelope. Once antrum formation has begun and the mitochondria have reached the dormant (storage) stage, cerium reactivity ceases. In adults, reaction occurs at the same stages of oocyte development but is weaker and limited to the mitochondria and their immediate vicinity.     

The giant (gt) mutants of Drosophila melanogaster alter DNA metabolism

Summary Abnormalities in DNA metabolism have been found in third-instar females of Drosophila melanogaster that are heteroallelic or homoallelic for X-chromosomal giant (gt) mutations. Analysis of DNA metabolism in larval brain ganglia was carried out using alkaline sucrose gradient centrifugation, incorporation assays and a neutral filter elution assay. These analyses show that gt stocks synthesize DNA of a reduced molecular weight, have an unusually high frequency of spontaneous single and doublestrand breaks, and exhibit a reduction in the normal inhibition of DNA synthesis following treatment with UV and the carcinogen AAAF. These phenomena are not associated with a defect in the repair of X-ray induced DNA breaks nor are they accompanied by any alterations in chromosome stability. Analysis of homozygous 1(2)gl larvae also reveal that these phenomena are specific to the gt locus and are thus not attributable solely to an extended developmental program. These findings strengthen the suggestion that the genetic instability associated with gt is related to perturbations in chromosome metabolism (Green 1982).Abbreviations used UV ultraviolet radiation-principal wavelength 313 nm - AAAF N-acetoxy-2-acetylaminofluorene     

Rationalization of some genetic anticodonic assignments

The genetic code appears to be a logic matrix in which, generally speaking, there is a correlation between the hydrophobicities of amino acids and their anticodonic nucleotides. There are several exceptions to this generality, however, and using previous data on hydrophobicity and binding constants, coupled with new data on reaction rates, we rationalize several of the anticodonic assignments.
