The mobility of heavy metals in contaminated soils is dependent on the kinetics release from soils. Metal extraction over time is commonly used to distinguish two or more fractions of metal based on differences of extraction or release rates. Kinetic studies using 0.01 M CaCl2, 0.01 M malic acid, and 0.01 M EDTA extractions were performed to characterize nickel (Ni) and lead (Pb) kinetic release in 10 contaminated calcareous soils. Proportions of Ni and Pb extracted with EDTA were higher than when using malic acid and CaCl2, respectively. The release of Ni and Pb was characterized by an initial fast rate followed by a slower rate and could best be described by a two first-order reactions model with rate constants k1 and k2 and two metal pools: readily labile (Q 1) and less labile (Q 2). In an EDTA extractant, different Q1 /Q2 ratios for Ni and Pb were observed, indicating binding energies to soil constituents is not comparable. The k1 of the model for Ni (average of 10 soils: 0.2204 h?1 and 0.2359 h?1 for 0.01 M CaCl2 and 0.01 EDTA, respectively) was higher than Pb (0.1044 h?1 and 0.1631 h?1 for 0.01 M CaCl2 and 0.01 M EDTA, respectively), indicating a higher potential of Ni for leaching and groundwater contamination in contaminated calcareous soils. Relationships between the fraction of Ni and Pb determined with the two first-order reactions model and the soil composition and Pb fractions were established. The results indicated that the efficiency of the extractions Ni and Pb from soils depends both on the Ni and Pb content and soil composition. Overall, the results indicated that the use of a 0.01 M malic acid washing solution would be preferred in the field condition.  相似文献

A trypsin‐like proteinase was purified and characterized in the midgut of Ectomyelois ceratoniae. A purification process that used Sepharyl G‐100 and DEAE‐cellulose fast flow chromatographies revealed a proteinase with specific activity of 66.7 μmol/min/mg protein, recovery of 27.04 and purification fold of 23.35. Molecular weight of the purified protein was found to be 35.8 kDa. Optimal pH and temperature were obtained 9 and 20°C for the purified trypsin proteinase, respectively. The purified enzyme was significantly inhibited by PMSF, TLCK, and SBTI as specific inhibitors of trypsins in which TLCK showed the highest inhibitory effect. Trypsin proteinase inhibitors were extracted from four varieties of pomegranate including Brait, Torsh‐Sabz, May‐Khosh, and Shirin by ion exchange chromatography. It was found that fractions 17–20 of Brait; fractions 18 and 21–26 of Torsh‐Sabz; fractions 1–7, 11–17, and 19–21 of May‐Khosh and fraction 8 for Shirin showed presence of trypsin inhibitor in these host. Comparison of their inhibitory effects on the purified trypsin proteinase of E. ceratoniae demonstrated that fractions from May‐khosh variety had the highest effect on the enzyme among other extracted fractions. Characterization of serine proteinases of insects mainly trypsins is one of the promising methods to decrease population and damages via extracting their inhibitors and providing resistant varieties.  相似文献

Background

Population differentiation is the result of demographic and evolutionary forces. Whole genome datasets from the 1000 Genomes Project (October 2012) provide an unbiased view of genetic variation across populations from Europe, Asia, Africa and the Americas. Common population-specific SNPs (MAF > 0.05) reflect a deep history and may have important consequences for health and wellbeing. Their interpretation is contextualised by currently available genome data.

Results

The identification of common population-specific (CPS) variants (SNPs and SSV) is influenced by admixture and the sample size under investigation. Nine of the populations in the 1000 Genomes Project (2 African, 2 Asian (including a merged Chinese group) and 5 European) revealed that the African populations (LWK and YRI), followed by the Japanese (JPT) have the highest number of CPS SNPs, in concordance with their histories and given the populations studied. Using two methods, sliding 50-SNP and 5-kb windows, the CPS SNPs showed distinct clustering across large genome segments and little overlap of clusters between populations. iHS enrichment score and the population branch statistic (PBS) analyses suggest that selective sweeps are unlikely to account for the clustering and population specificity. Of interest is the association of clusters close to recombination hotspots. Functional analysis of genes associated with the CPS SNPs revealed over-representation of genes in pathways associated with neuronal development, including axonal guidance signalling and CREB signalling in neurones.

Conclusions

Common population-specific SNPs are non-randomly distributed throughout the genome and are significantly associated with recombination hotspots. Since the variant alleles of most CPS SNPs are the derived allele, they likely arose in the specific population after a split from a common ancestor. Their proximity to genes involved in specific pathways, including neuronal development, suggests evolutionary plasticity of selected genomic regions. Contrary to expectation, selective sweeps did not play a large role in the persistence of population-specific variation. This suggests a stochastic process towards population-specific variation which reflects demographic histories and may have some interesting implications for health and susceptibility to disease.

The abundance of coccinellids on pistachio trees was monitored at both a cultivated pistachio plantation and a wild pistachio growing area in Rafsanjan, Iran. Eleven ladybird species were identified from planted pistachio trees with Adalia bipunctata (Linnaeus) and Oenopia conglobata contaminata (Menetries) the most abundant ladybirds. At the wild pistachio plantation site, 17 coccinellid species were collected, with Brumus undulatus Weise the most abundant ladybird, followed by A. bipunctata and O. conglobata contaminata. Three species of Coccinellidae were evaluated for their potential for biological control of the common pistachio psylla (CPP), Agonoscena pistaciae Burckhardt and Lauterer. Coccinella undecimpunctata aegyptica (Reiche), Exochomus nigripennis (Erichson) and Hippodamia variegata (Goeze) developed successfully on A. pistaciae nymphs and reproduced successfully. Development and reproduction of these ladybirds were also examined on an alternative prey, the cotton aphid Aphis gossypii Glover, the most abundant aphid species on herbal weeds in pistachio orchards. The highest intrinsic rate of natural increase (rm value) was observed in C. undecimpunctata aegyptica, 0.22 and 0.23 when it was fed on A. pistaciae and A. gossypii Glover, respectively. In comparison, the lowest rm values (0.12 and 0.13) were found in E. nigripennis on the CPP and cotton aphid diets, respectively. Exochomus nigripennis consumed the largest number of psyllid nymphs during larval period, while the adults consumed the lowest number of psyllid nymphs daily. Although eight psyllophagous coccinellid species were collected in colonies of the CPP on pistachio trees, they did not regulate psyllid numbers below damaging levels. At present, conservation should assume a central role in biological control strategies for this pest.  相似文献

We investigated the effect on lead (Pb) fractionation of the addition of plant residues and incubation time in three metal-amended calcareous soils of Iran. Four amended treatments with plant residues (helianthus, potato, rape, and wheat) were established at the rate of 2% and incubated for four weeks at 25°C and constant moisture. Lead was added to amended-soils at the rate of 200 mg kg?1 as chloride. A control without amendments but with Pb (control) was also set up. The samples were incubated for 3, 72, 168, 336, 672, 1008, and 1344 h at 25°C and constant moisture. After incubation, the soils were sequentially fractionated into water-soluble plus exchangeable (EXCH), organically bound (OM), inorganically bound (CARB), and residual (RES) forms. There were changes in the proportional distribution of Pb in all three studied soils during 1344 h of incubation with spiked Pb. In general, the proportions of Pb associated with the most weakly bound fraction (EXCH) tended to decrease, with corresponding increases in the other three more strongly binding fractions during the incubation. Application of plant residues increased Pb significantly in EXCH fractions in studied soils. Among the plant residues, application of potato residue gave the highest increase in EXCH fractions in two soils, which may be related to its lower C/N ratio. The higher proportions of EXCH fraction of spiked and amended soils in these calcareous soils indicate the higher potential of downward leaching and runoff transport, especially at the early stage of pollution.  相似文献
