Central Asian mountain voles Alticola is one of the least known groups of voles both in evolution and life history. This genus includes three subgenera Alticola s.str., Aschizomys and Platycranius, and belongs to the tribe Clethrionomyini comprising also red‐backed voles Clethrionomys and oriental voles Eothenomys. In order to elucidate the phylogenetic relationships within Alticola and to examine its position within the tribe, mitochondrial cytochrome b (cyt b) gene variation was estimated, and the results were compared with morphological and palaeontological data. Maximum likelihood (ML), neighbor‐joining (NJ), maximum parsimony (MP) and Bayesian phylogenetic analyses show that the genus Alticola does not appear to be a monophyletic group since the representatives of Aschizomys branch within Clethrionomys, whereas two other subgenera (Alticola and Platycranius) form a separate monophyletic clade. Flat‐headed vole Alticola (Platycranius) strelzowi is nested within the nominative subgenus showing close association with A. (Alticola) semicanus. Surprisingly, the two species of Aschizomys do not form a monophyletic group. The results of the relaxed‐clock analysis suggest that the Alticola clade splits from the Clethrionomys stem in early Middle Pliocene while basal cladogenetic events within Alticola s.str. dates back to the late Middle to early Late Pliocene. A scenario of evolution in Clethrionomyini is put forward implying rapid parallel morphological changes in different lineages leading to the formation of Alticola‐like biomorphs adapted to mountain and arid petrophilous habitats. Carcinoma of the hypopharynx is an uncommon disease, with an annual incidence of approximately 1 in 100,000. Post-cricoid carcinoma is more common in women and is not usually associated with tobacco and alcohol abuse. Reconstruction of large pharyngeal defects following surgery for squamous cell carcinoma is complex and often requires microvascular free tissue transfer to achieve the best oncological and functional outcomes. The most common complications of such procedures include fistulas and strictures of the neopharynx. Here, we describe a case of a female patient admitted to the Head and Neck Department at our hospital to undergo delayed reconstruction following pharyngolaryngectomy and removal of the cervical oesophagus. Several complications occurred during post-operative care, including stricture and skin dehiscence. At present, the patient is able to swallow saliva and is currently being prepared to return to a normal diet.  相似文献

The influence of alkaline and the neutral grade of magnesium aluminometasilicate as a porous solid carrier for the liquid self-emulsifying formulation with ibuprofen is investigated. Ibuprofen is dissolved in Labrasol, then this solution is adsorbed on the silicates. The drug to the silicate ratio is 1:2, 1:4, and 1:6, respectively. The properties of formulations obtained are analyzed, using morphological, porosity, crystallinity, and dissolution studies. Three solid self-emulsifying (S-SE) formulations containing Neusilin SG2 and six consisting of Neusilin US2 are in the form of powder without agglomerates. The nitrogen adsorption method shows that the solid carriers are mesoporous but they differ in a specific surface area, pore area, and the volume of pores. The adsorption of liquid SE formulation on solid silicate particles results in a decrease in their porosity. If the neutral grade of magnesium aluminometasilicate is used, the smallest pores, below 10 nm, are completely filled with liquid formulation, but there is still a certain number of pores of 40–100 nm. Dissolution studies of liquid SEDDS carried out in pH = 1.2 show that Labrasol improves the dissolution of ibuprofen as compared to the pure drug. Ibuprofen dissolution from liquid SE formulations examined in pH of 7.2 is immediate. The adsorption of the liquid onto the particles of the silicate causes a decrease in the amount of the drug released. Finally, more ibuprofen is dissolved from S-SE that consist of the neutral grade of magnesium aluminometasilicate than from the formulations containing the alkaline silicate.KEY WORDS: dissolution, ibuprofen, labrasol, magnesium aluminometasilicate, self-emulsifying powder  相似文献

Recessive dystrophic epidermolysis bullosa (RDEB) is caused by mutations in COL7A1 resulting in reduced or absent type VII collagen, aberrant anchoring fibril formation and subsequent dermal-epidermal fragility. Here, we identify a significant decrease in PLOD3 expression and its encoded protein, the collagen modifying enzyme lysyl hydroxylase 3 (LH3), in RDEB. We show abundant LH3 localising to the basement membrane in normal skin which is severely depleted in RDEB patient skin. We demonstrate expression is in-part regulated by endogenous type VII collagen and that, in agreement with previous studies, even small reductions in LH3 expression lead to significantly less secreted LH3 protein. Exogenous type VII collagen did not alter LH3 expression in cultured RDEB keratinocytes and we show that RDEB patients receiving bone marrow transplantation who demonstrate significant increase in type VII collagen do not show increased levels of LH3 at the basement membrane. Our data report a direct link between LH3 and endogenous type VII collagen expression concluding that reduction of LH3 at the basement membrane in patients with RDEB will likely have significant implications for disease progression and therapeutic intervention.  相似文献

Clostridium difficile is the etiological agent of diarrhoea and colitis, especially in elderly patients. The incidence of these diseases has increased during the last 10 years. Emergence of so-called hypervirulent strains is considered as one of the main factors responsible for the more severe disease and changed profile of sensitivity to antimicrobial agents. The aim of this work was to determine the sensitivity profile of toxigenic strains of C. difficile in the Czech Republic in 2011–2012 to selected antibiotics. The antibiotics clindamycin, metronidazole, vancomycin and amoxicillin with clavulanic acid were used for this purpose. Isolates cultured on Brazier's C. difficile selective agar were analysed for the presence of toxin genes using Xpert detection system. Xpert analysis revealed that 33 strains carried the genes for toxins tcdB, cdt and tcdCΔ117, thus showing characteristics typical for the hypervirulent ribotype 027/PFGE type NAP1/REA type B1. The remaining 29 strains carried only the gene for toxin B (tcdB) and not cdt and tcdCΔ117. Our results indicate the higher susceptibility of C. difficile hypertoxigenic strains to three out of four tested antibiotics (except vancomycin) than it is for the other toxigenic strains. We found that only 10.34 % of other toxigenic strains were resistant to clindamycin, and no resistance was found in all other cases. All the isolates were sensitive to amoxicillin/clavulanic acid in vitro. However, its use is not recommended for therapy of infections caused by C. difficile.  相似文献

Rhinanthoid Orobanchaceae form a monophyletic lineage that include the hemiparasitic genera Euphrasia, Melampyrum, Tozzia, Bartsia, Nothobartsia, Odontites (s.l.), Rhinanthus, Rhynchocorys, Parentucellia, Hedbergia and holoparasitic Lathraea. In this study, we aimed to reconstruct the phylogeny, evolution of life-history traits (life cycle and seed size) and explain the extant biogeographical patterns in this group. For phylogenetic reconstruction, we used molecular data obtained by sequencing the nuclear ITS region and the chloroplast trnT-trnL intergenic spacer and matK?+?trnK regions. The genus Melampyrum was found to occupy the sister position to the rest of the group. The other genera were assembled in the sister Rhinanthus-Rhynchocorys-Lathraea and Bartsia-Euphrasia-Odontites subclades. The reconstruction of life-cycle evolution yielded ambiguous results suggesting nonetheless a substantially higher likelihood of perenniality compared to annuality in most ancestor lineages. Seed size varied across two orders of magnitude (average weight per seed: 0.02–7.22 mg) and tended to decrease in the Bartsia-Euphrasia-Odontites subclade compared to the rest of the group. Seed-size evolution was correlated with life-history evolution in the group if the generally small-seeded Bartsia-Euphrasia-Odontites subclade is excluded. We formulated hypotheses relating the extant biogeographical affinities of individual genera to the geological history of the Euro-Caucasian diversity center of the group. Notable dispersal events in Euphrasia and Bartsia were hypothesized to be allowed or at least facilitated by a specific combination of the life-history traits.  相似文献

A method that was based on non‐invasive sampling of genetic material was used to determine the rates of extra‐pair paternity (EPP) and conspecific brood parasitism (CBP) in mallards. Maternal and offspring DNA were extracted from feathers in nest material and hatched eggshell membranes. Using 8 microsatelite loci, extra‐pair offspring were detected in 48% of nests and accounted for 9.3% of all offspring. In addition, 10.1% of the offspring were confirmed to result from CBP, and 24% of all nests contained at least 1 offspring from CBP. Rates of conspecific nest parasitism were higher than those of related species, which might have been due to higher breeding densities at our study site. The incidence of EPP was distributed randomly (i.e. did not deviate from bionomial distribution) throughout the population, indicating that variations in pre‐copulatory (e.g. female choice, mate guarding) or post‐copulatory processes (e.g. sperm competition, cryptic female choice) do not affect the distribution of EPP among breeding pairs markedly. Yet, our data provide evidence of variation in the risk of being parasitized among breeding females. The occurrence of CBP and EPP was unaffected by the timing of the breeding attempt or breeding synchrony.  相似文献
