Predicting whether microbial invaders will colonize an environment is critical for managing natural and engineered ecosystems, and controlling infectious disease. Invaders often face competition by resident microbes. But how invasions play out in communities dominated by facilitative interactions is less clear. We previously showed that growth medium toxicity can promote facilitation between four bacterial species, as species that cannot grow alone rely on others to survive. Following the same logic, here we allowed other bacterial species to invade the four-species community and found that invaders could more easily colonize a toxic medium when the community was present. In a more benign environment instead, invasive species that could survive alone colonized more successfully when the residents were absent. Next, we asked whether early colonists could exclude future ones through a priority effect, by inoculating the invaders into the resident community only after its members had co-evolved for 44 weeks. Compared to the ancestral community, the co-evolved resident community was more competitive toward invaders and less affected by them. Our experiments show how communities may assemble by facilitating one another in harsh, sterile environments, but that arriving after community members have co-evolved can limit invasion success.Subject terms: Microbial ecology, Microbial ecology 相似文献

Background and Aims
Proton pump inhibitors (PPIs) have been associated with adverse clinical outcomes amongst clopidogrel users after an acute coronary syndrome. Recent pre-clinical results suggest that this risk might extend to subjects without any prior history of cardiovascular disease. We explore this potential risk in the general population via data-mining approaches.Methods
Using a novel approach for mining clinical data for pharmacovigilance, we queried over 16 million clinical documents on 2.9 million individuals to examine whether PPI usage was associated with cardiovascular risk in the general population.Results
In multiple data sources, we found gastroesophageal reflux disease (GERD) patients exposed to PPIs to have a 1.16 fold increased association (95% CI 1.09–1.24) with myocardial infarction (MI). Survival analysis in a prospective cohort found a two-fold (HR = 2.00; 95% CI 1.07–3.78; P = 0.031) increase in association with cardiovascular mortality. We found that this association exists regardless of clopidogrel use. We also found that H2 blockers, an alternate treatment for GERD, were not associated with increased cardiovascular risk; had they been in place, such pharmacovigilance algorithms could have flagged this risk as early as the year 2000.Conclusions
Consistent with our pre-clinical findings that PPIs may adversely impact vascular function, our data-mining study supports the association of PPI exposure with risk for MI in the general population. Hepatitis C virus (HCV) infection causes chronic liver diseases and is a global public health problem. Detailed analyses of HCV have been hampered by the lack of viral culture systems. Subgenomic replicons of the JFH1 genotype 2a strain cloned from an individual with fulminant hepatitis replicate efficiently in cell culture. Here we show that the JFH1 genome replicates efficiently and supports secretion of viral particles after transfection into a human hepatoma cell line (Huh7). Particles have a density of about 1.15-1.17 g/ml and a spherical morphology with an average diameter of about 55 nm. Secreted virus is infectious for Huh7 cells and infectivity can be neutralized by CD81-specific antibodies and by immunoglobulins from chronically infected individuals. The cell culture-generated HCV is infectious for chimpanzee. This system provides a powerful tool for studying the viral life cycle and developing antiviral strategies. 相似文献

Hepatitis C virus (HCV) is a leading cause of chronic viral hepatitis worldwide. The study of antibody-mediated virus neutralization has been hampered by the lack of an efficient and high-throughput cell culture system for the study of virus neutralization. The HCV structural proteins have been shown to assemble into noninfectious HCV-like particles (HCV-LPs). Similar to serum-derived virions, HCV-LPs bind and enter human hepatocytes and hepatoma cell lines. In this study, we developed an HCV-LP-based model system for a systematic functional analysis of antiviral antibodies from patients with acute or chronic hepatitis C. We demonstrate that cellular HCV-LP binding was specifically inhibited by antiviral antibodies from patients with acute or chronic hepatitis C in a dose-dependent manner. Using a library of homologous overlapping envelope peptides covering the entire HCV envelope, we identified an epitope in the N-terminal E2 region (SQKIQLVNTNGSWHI; amino acid positions 408 to 422) as one target of human antiviral antibodies inhibiting cellular particle binding. Using a large panel of serum samples from patients with acute and chronic hepatitis C, we demonstrated that the presence of antibodies with inhibition of binding activity was not associated with viral clearance. In conclusion, antibody-mediated inhibition of cellular HCV-LP binding represents a convenient system for the functional characterization of human anti-HCV antibodies, allowing the mapping of envelope neutralization epitopes targeted by naturally occurring antiviral antibodies. 相似文献

The new genus Cieneguiticeras, assigned to the family Oppeliidae, is described on the basis of Andean lower-middle Tithonian ammonites from Arroyo Cieneguita, west-central part of the Neuquén-Mendoza Basin, Argentina. The macroconchs are closely homoeomorphic with Neochetoceras Spath and the microconchs have a ‘glochiceratid’-type morphology. The stratigraphic range of Cieneguiticeras nov. gen. includes the lower and middle Tithonian by means of a succession of two or three species which are interpreted as members of a phyletic lineage. Ammonites from the Tithonian of Cuba, Mexico and France are more or less confidently included in this new genus. 相似文献

Food web structure is well known to vary widely among ecosystems. Recent research indicates that there can be a high degree
of spatial heterogeneity within ecosystems as well. Xochimilco is a small heterogeneous freshwater system that has been transformed
into a network of canals, small lakes, and wetlands. Located within Mexico City, this ecosystem has been intensively managed
and highly impacted for more than 50 years. This system receives urban and agricultural runoff, with resulting impacts on
water quality. The aquatic community is dominated by exotics such as carp (Cyprinus carpio) and tilapia (Oreocrhomis niloticus), though the system still supports endemic species such as the aquatic salamander, axolotl (Ambystoma mexicanum), and crayfish (Cambarellus montezumae), which are both endangered. In this study, we used carbon and nitrogen stable isotopes for the whole food web and gut content
analysis from the exotic fishes to describe food web structure in different canals within Xochimilco. There were significant
isotopic differences among canals. These differences may result from isotopic baseline differences as well as differences
in actual food web structure: both are related to local spatial variation in water quality driven by nutrient inputs and exotic
fishes. Within-ecosystem variability is likely to be seen in other perturbed shallow systems as well, and should be explicitly
