Neurites grow faster towards the cathode than the anode in a steady field.   总被引:9,自引:0,他引:9  
We explanted fragments of embryonic chick dorsal root ganglia on to polylysine coated glass and cultured them in a medium containing one unit of nerve growth factor plus enough methylcellulose to give viscosities from 0.01-3,000 poise. We allowed them to grow out in the absence of a field, and then selected explants with halos of neurites which were relatively dense, relatively symmetrical, and practically free of glial cells. These selected explants were then exposed to electrical fields of up to 140 mV/mm for some hours. In media with viscosities of one poise or less, the field some times dragged the central cell mass of an explant towards the anode. However, in cases where the central cell mass did not move, fields of 70-140 mV/mm induced that sector of each neurite halo which faced the cathode to grow out several times faster than the one facing the anode.  相似文献

Summary Experiments were done to relate the presence of tannin vacuoles to plant movements. When surveyed, 4 out of 10 species exhibited rapid thigmonastic or nyctinastic movements, and only those 4 species had tannin vacuoles in their motor cells. Interestingly, plants ofAlbizzia julibrissin whose seed was obtained in New Haven, Connecticut had rapid nyctinasty and tannin vacuoles in the tertiary pulvini, whereasAlbizzia collected in Winston-Salem, North Carolina had neither. Nyctinasty ofMimosa pudica andAlbizzia NH, both of which had tannin vacuoles, was at the rate of 4.2 °/min, whereasAlbizzia WS andSamanea saman, neither of which had tannin vacuoles, exhibited leaflet closure at the rate of 2.0–2. 1 °/min. Pretreatment with the Ca2+ channel agonist Bay K-8644 increased, and with the antagonist verapamil decreased, the rate of leaflet closure only inMimosa andAlbizzia NH. Pretreatment with the Ca2+ inhibitor Erythrosin B substantially blocked reopening only inMimosa andAlbizzia NH. Hydrocortisone (10?6 – 10?7M) completely inhibited the production of hemoglobin by DMSO- and DMF-treated Friend erythroleukemia cells (FLC) in vitro without affecting either cell replication or general protein synthesis. Only 11, 17-dihydroxycorticosteroids were effective in inhibiting this expression of differentiation. Addition of hydrocortisone as late as 48 hours after the addition of DMSO (at a time at which cells were committed to differentiation) still resulted in significant inhibition of hemoglobin synthesis. Although the mechanism of this action is unknown, since it was not reversed by the addition of arachidonic acid nor a number of prostaglandins, it appears to be unrelated to the ability of corticosteroids to inhibit endogenous prostaglandin synthesis.  相似文献

Crude homogenates of adult Dirofilaria immitis females were able to incorporate choline into phosphatidylcholine (PC) and also were able to methylate phosphatidyl (N,N-dimethyl)-ethanolamine, using S-adenosylmethionine as the methyl donor, to form PC. The finding of choline phosphotransferase (EC 2·7·8·2) and phosphatidyl (N,N-dimethyl) ethanolamine methyltransferase activity in the paniculate (mainly microsomal) fraction of the homogenates provided further evidence that adult D. immitis females can synthesize PC by way of choline and cytidine 5'-diphosphocholine (Kennedy pathway) and also by way of S-adenosylmethionine-mediated sequential methylation of phosphatidylethanolamine (Bremer-Greenberg pathway).  相似文献

We have phenotypically and molecularly analyzed the cutlet locus in Drosophila. Homozygous cutlet flies exhibit abnormal development of a subset of adult tissues, including the eye, wing, and ovary. We show that abnormal development of these tissues is due to a defect in normal cell growth. Surprisingly, cell growth is affected in all developing precursor tissues in cutlet mutant animals, including those that give rise to phenotypically wild-type adult structures. The cutlet gene encodes a Drosophila homologue of yeast CHL12 and has similarity to mammalian replication factor C. In addition, cutlet genetically interacts with multiple subunits of Drosophila replication factor C. Our results suggest that the cutlet gene product acts as an accessory factor for DNA replication and has different requirements for the formation of various adult structures during Drosophila development.  相似文献

Thrombin stimulation of prostacyclin (PGI2) synthesis by cultured human umbilical vein endothelial cells (HUVEC) requires the active site of thrombin and involves rapid and transient rises in cytoplasmic free calcium [Ca2+]i. In this study, we investigated whether or not the anion-binding exosite for fibrinogen recognition of thrombin (which confers certain substrate specificities) is also necessary for the induction of rises in [Ca2+]i and PGI2 production. Thrombin variants which lack either the catalytic site (DIP-alpha-thrombin) or anion-binding exosite (gamma-thrombin) either alone or in combination failed to induce rises in [Ca2+]i or PGI2 production in HUVEC. To further study the role of the anion-binding exosite of thrombin in the activation of HUVEC, COOH-terminal fragments of hirudin were used. This portion of hirudin interacts with the anion-binding exosite of thrombin and inhibits thrombin-induced fibrinogen coagulation while leaving the catalytic activity of thrombin intact. A 21-amino acid COOH-terminal peptide of hirudin (N alpha-acetyldesulfato-hirudin45-65 or Hir45-65) inhibited thrombin-induced (0.5 U/ml) rises in [Ca2+]i and PGI2 production with IC50 of 0.13 and 0.71 microM, respectively. Similar results were obtained using shorter hirudin-derived peptides. Thus, the fibrinogen anion-binding exosite of thrombin is required for alpha-thrombin-induced rises in [Ca2+]i and PGI2 production in HUVEC.  相似文献
