This paper seeks to show how the traditional societies in western Serengeti have coexisted and continue to coexist with wildlife. It also recognizes the relevancy of this coexistence in furthering contemporary conservation efforts although there are practical constraints to putting this into practice. The following questions are examined: (1) How did/do traditional societies in Serengeti interact with their nature? (2) Which traditional management institutions governed/govern interaction between people and wildlife species, resources and ecosystems and, how do they operate? (3) Which factors were (or are) responsible for erosion of traditional management institutions? (4) What can the traditional practices offer to contemporary conservation efforts and what are the limitations? The paper identifies four ways in which traditional institutions and practices can contribute to current conservation efforts: regulating the overexploitation of resources; complementing the current incentives aiming at diffusing prevailing conflicts between conservation authorities and communities; minimising the costs of law enforcement and; complementing the modern scientific knowledge in monitoring and responding to ecosystem processes and functions. The practical constraints likely to limit adoption of these practices are presented as: methodological complications of acquiring indigenous knowledge; prevailing historical conflicts; human population growth; poverty and lack of appreciation among the conservation planners and managers. Since, proteins carry out many functional roles in a cell with different concentrations, proteomics is likely a more appropriate approach to explain biological processes and cellular events than mRNA studies. Although, gene ontology provides a systematic and organized vocabulary of biological terms for proteins, we need more details to decide about the correct duty and annotation of proteins in a specific condition. One can assume that a change of protein concentration is related to a biological process of that protein with negligible error. Therefore, we can obtain more information about the function of proteins by studying these profiles. In this study, we used time-course proteomic data of a twenty day differentiation study of embryonic stem cells (ESCs) differentiating to embryoid bodies (EBs). Hierarchical clustering was used to cluster time-series concentration profile of proteins. Our results demonstrate that there are eleven active processes with distinct concentration profiles in this initial differentiation. According to the concentration profiles of proteins, we suggest new change points (or equivalently, new stages) in the course of embryonic differentiation. 相似文献

Natural products are considered recently as one of the source for production of efficient therapeutical agents for breast cancer treatment. In this study, a sesquiterpene lactone, 13-O-acetylsolstitialin A (13ASA), isolated from Centaurea cyanus, showed cytotoxic activities against MCF-7 and MDA-MB-231 breast cancer cell lines using standard 3-(4, 5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay. To find the mechanism of action of cytotoxicity, annexin V/propidium iodide (PI) staining was performed for evaluation of apoptosis. This process was further confirmed by immunoblotting of anti- and proapoptotic, Bcl-2 and Bax, proteins. Cell cycle arrest was evaluated by measurement of fluorescence intensity of PI dye and further confirmed by immunoblotting of Cdk-4 and cyclin D1. Mitochondrial transmembrane potential (ΔΨm) and generation of reactive oxygen species (ROS) were measured using the JC-1 and DCFDA fluorescence probes, respectively. These experiments showed that 13ASA is a potent cytotoxic agent, which activates apoptosis-mediated cell death. In response to this compound, Bax/Bcl-2 ratio was noticeably increased in MCF-7 and MDA-MB-231 cells. Moreover, 13ASA induced cell cycle arrest at subG1 and G1 phases by decreasing protein levels of cyclin D1 and Cdk-4. It was done possibly through the decrease of ΔΨm and increase of ROS levels which induce apoptosis. In conclusion, this study mentioned that 13ASA inhibit the growth of MCF-7 and MDA-MB-231 breast cancer cell lines through the induction of cell cycle arrest, which triggers apoptotic pathways. 13ASA can be considered as a susceptible compound for further investigation in breast cancer study. 相似文献

Various clinical protocols have been developed to aid in the clinical diagnosis of classical bovine spongiform encephalopathy (BSE), which is confirmed by postmortem examinations based on vacuolation and accumulation of disease-associated prion protein (PrPd) in the brain. The present study investigated the occurrence and progression of sixty selected clinical signs and behaviour combinations in 513 experimentally exposed cattle subsequently categorised postmortem as confirmed or unconfirmed BSE cases. Given sufficient space, it is possible for gliding animals to reach an equilibrium state with no net forces acting on the body. In contrast, every gliding trajectory must begin with a non-steady component, and the relative importance of this phase is not well understood. Of any terrestrial animal glider, snakes exhibit the greatest active movements, which may affect their trajectory dynamics. Our primary aim was to determine the characteristics of snake gliding during the transition to equilibrium, quantifying changes in velocity, acceleration, and body orientation in the late phase of a glide sequence. We launched 'flying' snakes (Chrysopelea paradisi) from a 15 m tower and recorded the mid-to-end portion of trajectories with four videocameras to reconstruct the snake's body position with mm to cm accuracy. Additionally, we developed a simple analytical model of gliding assuming only steady-state forces of lift, drag and weight acting on the body and used it to explore effects of wing loading, lift-to-drag ratio, and initial velocity on trajectory dynamics. Despite the vertical space provided to transition to steady-state gliding, snakes did not exhibit equilibrium gliding and in fact displayed a net positive acceleration in the vertical axis, an effect also predicted by the analytical model. 相似文献

The use of animal models for initially screening anti-aging drugs is a promising approach for drug discovery. However, there a number of potential artifacts, confounds and errors that can arise in such research programs. The following rules are intended to minimize such problems: (1) since aging occupies an increasing proportion of human adulthood, data that conflate aging and late life should not be extrapolated to human aging; (2) the response to candidate medications should show a normal dose-response pattern, although not necessarily a linear response; (3) medicated animal models should not be hypometabolic; (4) medicated animal models should not show pronounced reductions in fertility; (5) medicated animal models should not exhibit general nervous system depression; (6) the effect of the medication should not be highly sensitive to the culture environment; (7) the effect of the medication should not be highly dependent on the genetic ancestry of the stock employed, leaving aside inbreeding, which should be avoided because humans are not generally inbred. While these rules do not guarantee successful extrapolation of successful drug results from the animal model to humans in a clinical setting, the failure to adhere to these rules should raise doubts about such extrapolation. 相似文献

Natural killer (NK) cells are cytotoxic lymphocytes that lack CD3 and express variable levels of CD16, CD56 and CD57. In recent years NK cells have been categorised into two major groups based on the level of CD56 expression. This phenotypic classification correlates with functional activity as CD56bright NK cells are the major cytokine producing subset whereas CD56dim NK cells exhibit greater cytotoxic activity.
