Nuclear disasters at Chernobyl and Fukushima provide examples of effects of acute ionizing radiation on mutations that can affect the fitness and distribution of species. Here, we investigated the prevalence of Microbotryum lychnidis‐dioicae, a pollinator‐transmitted fungal pathogen of plants causing anther‐smut disease in Chernobyl, its viability, fertility and karyotype variation, and the accumulation of nonsynonymous mutations in its genome. We collected diseased flowers of Silene latifolia from locations ranging by more than two orders of magnitude in background radiation, from 0.05 to 21.03 μGy/h. Disease prevalence decreased significantly with increasing radiation level, possibly due to lower pollinator abundance and altered pollinator behaviour. Viability and fertility, measured as the budding rate of haploid sporidia following meiosis from the diploid teliospores, did not vary with increasing radiation levels and neither did karyotype overall structure and level of chromosomal size heterozygosity. We sequenced the genomes of twelve samples from Chernobyl and of four samples collected from uncontaminated areas and analysed alignments of 6068 predicted genes, corresponding to 1.04 × 107 base pairs. We found no dose‐dependent differences in substitution rates (neither dN, dS, nor dN/dS). Thus, we found no significant evidence of increased deleterious mutation rates at higher levels of background radiation in this plant pathogen. We even found lower levels of nonsynonymous substitution rates in contaminated areas compared to control regions, suggesting that purifying selection was stronger in contaminated than uncontaminated areas. We briefly discuss the possibilities for a mechanistic basis of radio resistance in this nonmelanized fungus.  相似文献

Models of herbivore diversification rely heavily on adaptations that reflect the nutritional quality of foods consumed. In particular, browsers and grazers are expected to show dichotomous adaptations to deal with high quality (concentrate) browse-based and poor quality grass-based diets, respectively. In this study, we test the widespread assumption that browse represents a higher quality food source than grass. We analyzed plants from a South African savanna, collected over one dry and one wet season across several habitat types, for percent nitrogen (%N), neutral detergent fiber (NDF), acid detergent fiber (ADF), and acid detergent lignin (ADL) to compare variations in nutritional value of different food types. Results show consistently higher %N and lower NDF and ADF of tree foliage and forbs compared to monocots, but the former have consistently higher ADL, implying a higher fiber digestibility in grass compared with browse. Some fruit species have a high NDF and ADL content, implying poorer nutritional value than is commonly assumed. Our findings are in agreement with several other studies depicting relatively poor digestibility of browse (tree foliage and fruit) compared to grass. Reference to browse as high quality foods is therefore misleading, and models of herbivory that rest on this assumption require revision. The more efficient fiber digestibility recorded in grazers compared to browsers cannot be treated as an adaptation to poor quality diets, but rather to maximize benefits of higher fiber digestibility of grass. Spatio-seasonal variations in plant nutritional seem to reflect seasonal and spatial diet changes expected for grazers and intermediate (mixed) feeders. We propose that future studies require further detail on variations in diet, diet quality, and digestive efficiency to properly understand mechanisms of adaptation.  相似文献

Two large-scale phenotyping efforts, the European Mouse Disease Clinic (EUMODIC) and the Wellcome Trust Sanger Institute Mouse Genetics Project (SANGER-MGP), started during the late 2000s with the aim to deliver a comprehensive assessment of phenotypes or to screen for robust indicators of diseases in mouse mutants. They both took advantage of available mouse mutant lines but predominantly of the embryonic stem (ES) cells resources derived from the European Conditional Mouse Mutagenesis programme (EUCOMM) and the Knockout Mouse Project (KOMP) to produce and study 799 mouse models that were systematically analysed with a comprehensive set of physiological and behavioural paradigms. They captured more than 400 variables and an additional panel of metadata describing the conditions of the tests. All the data are now available through EuroPhenome database (www.europhenome.org) and the WTSI mouse portal (http://www.sanger.ac.uk/mouseportal/), and the corresponding mouse lines are available through the European Mouse Mutant Archive (EMMA), the International Knockout Mouse Consortium (IKMC), or the Knockout Mouse Project (KOMP) Repository. Overall conclusions from both studies converged, with at least one phenotype scored in at least 80?% of the mutant lines. In addition, 57?% of the lines were viable, 13?% subviable, 30?% embryonic lethal, and 7?% displayed fertility impairments. These efforts provide an important underpinning for a future global programme that will undertake the complete functional annotation of the mammalian genome in the mouse model.  相似文献

The investigation of canine personality has failed to find strong agreement between studies as to its structure. The area has been hampered by a reliance on human personality models and by a tendency to limit the types of dogs used as subjects. These problems were recently addressed during the development of the Monash Canine Personality Questionnaire (MCPQ). In this follow-up study, over 450 participants provided demographic information about themselves and their dog and completed the MCPQ for their dog. Structural Equation Modelling results necessitated reassessing the original data and reanalysis suggested a more succinct questionnaire, the MCPQ-R, to measure five dimensions of canine personality very similar to those revealed in earlier work (extraversion, motivation, training focus, amicability and neuroticism). Owner reports of personality were generally consistent across demographic variables for all five canine personality subscales. There was no association between any subscale score and owner gender or education level, or dog sex or sexual status (desexed or not desexed). Significant, but generally weak associations were found for owner age and Extraversion (r = 0.17, P < 0.001), owner knowledge of their dog and Training Focus (r = 0.22, P < 0.001), time spent inside and Extraversion (r = ?0.13, P = 0.007), dog age and Extraversion (r = 0.14, P = 0.004), Training Focus and Extraversion (r = 0.13, P = 0.007), dog height and Neuroticism (r = ?0.20, P < 0.001) and dog height and Amicability (r = 0.2, P < 0.001), dog weight and Neuroticism (r = ?0.17, P < 0.01) and dog weight and Amicability (r = 0.19, P < 0.01). There were also few differences in personality ratings across recognised purebred dog breed groups, although Working Dogs and Terriers scored significantly more highly on the Extraverted subscale than other groups (P < 0.001) and Working Dogs and Gundogs scored more highly on the Training Focus subscale (P < 0.001). These results are consistent with the view that the MCPQ-R assesses a construct, canine personality, which is relatively stable and comprised of five dimensions.  相似文献

The design and development of a series of highly selective pyrrolidine carboxamide 11β-HSD1 inhibitors are described. These compounds including PF-877423 demonstrated potent in vitro activity against both human and mouse 11β-HSD1 enzymes. In an in vivo assay, PF-877423 inhibited the conversion of cortisone to cortisol. Structure guided optimization effort yielded potent and stable 11β-HSD1 selective inhibitor 42.  相似文献

Some species described as gynodioecious are truly gynomonoecious-gynodioecious. Three distinct phenotypes may be found in their natural populations-female and hermaphrodite pure-sexed plants bearing either only pistillate or perfect flowers, respectively, and mixed plants bearing both types of flowers. In one such species, Dianthus sylvestris, we investigated mating system parameters using allozyme data. Outcrossing rates and correlations of outcrossed paternity were calculated for the three types of plants and separately for pistillate and perfect flowers on mixed plants. The mean outcrossing rate for the population was t(m) ± SD = 0.885 ± 0.032. Females were more outcrossed than hermaphrodites (0.987 ± 0.112 and 0.790 ± 0.076, respectively), whereas mixed plants were not significantly more or less outcrossed than hermaphrodites (0.840 ± 0.060). Within mixed plants, perfect flowers showed an intermediate outcrossing rate (0.898 ± 0.057), whereas pistillate flowers were as selfed as perfect flowers on hermaphrodite plants (0.782 ± 0.111). Family estimates of outcrossing rates were highly variable. Globally, no biparental inbreeding was detected in this species, and there was a mean of 61.5 ± 19.9% of full-sibs within families. Floral dimorphism between small pistillate and large perfect flowers together with pollinator preference for larger flowers could explain the observed patterns for both mating parameters. The advantages of gynomonoecy-gynodioecy are discussed. We conclude that mixed plants do not reduce selfing for all flowers on a plant, but perfect flowers on these plants seem to have an outcrossing advantage.  相似文献

Malaria parasites suffer severe losses in the mosquito as they cross the midgut, haemolymph and salivary gland tissues, in part caused by immune responses of the insect. The parasite compensates for these losses by multiplying during the oocyst stage to form the infectious sporozoites. Upon human infection, malaria parasites are again attenuated by sustained immune attack. Here, we report a single copy gene that is highly conserved amongst Plasmodium species that encodes a secreted protein named PxSR. The predicted protein is composed of a unique combination of metazoan protein domains that have been previously associated with immune recognition/activation and lipid/protein adhesion interactions at the cell surface, namely: (i) scavenger receptor cysteine rich (SRCR); (ii) pentraxin (PTX); (iii) polycystine-1, lipoxygenase, alpha toxin (LH2/PLAT); (iv) Limulus clotting factor C, Coch-5b2 and Lgl1 (LCCL). In our assessment the PxSR molecule is completely novel in biology and is only found in Apicomplexa parasites. We show that PxSR is expressed in sporozoites of both human and rodent malaria species. Disruption of the PbSR gene in the rodent malaria parasite P. berghei results in parasites that form normal numbers of oocysts, but fail to produce any sporozoites. We suggest that, in addition to a role in sporogonic development, PxSR may have a multiplicity of functions.  相似文献

A na?ve (unimmunized) human semisynthetic phage display library was employed to isolate recombinant antibody fragments against the cyanobacterial hepatotoxin microcystin-LR. Selected antibody scFv genes were cloned into a soluble expression vector and expressed in Escherichia coli for characterization against purified microcystin-LR by competition enzyme-linked immunosorbent assay (ELISA). The most sensitive single-chain antibody (scAb) isolated was capable of detecting microcystin-LR at levels below the World Health Organization limit in drinking water (1 microg liter(-1)) and cross-reacted with three other purified microcystin variants (microcystin-RR, -LW, and -LF) and the related cyanotoxin nodularin. Extracts of the cyanobacterium Microcystis aeruginosa were assayed by ELISA, and quantifications of microcystins in toxic samples showed good correlation with analysis by high-performance liquid chromatography. Immobilized scAb was also used to prepare immunoaffinity columns, which were assessed for the ability to concentrate microcystin-LR from water for subsequent analysis by high-performance liquid chromatography. Anti-microcystin-LR scAb was immobilized on columns via a hexahistidine tag, ensuring maximum exposure of antigen binding sites, and the performance of the columns was evaluated by directly applying 150 ml of distilled water spiked with 4 micro g of purified microcystin-LR. The procedure was simple, and a recovery rate of 94% was achieved following elution in 1 ml of 100% methanol. Large-scale, low-cost production of anti-microcystin-LR scAb in E. coli is an exciting prospect for the development of biosensors and on-line monitoring systems for microcystins and will also facilitate a range of immunoaffinity applications for the cleanup and concentration of these toxins from environmental samples.  相似文献
