ABC transporters from the multidrug resistance-associated protein (MRP) subfamily are glutathione S-conjugate pumps exhibiting a broad substrate specificity illustrated by numerous xenobiotics, such as anticancer drugs, herbicides, pesticides and heavy metals. The engineering of MRP transporters into plants might be interesting either to reduce the quantity of xenobiotics taken up by the plant in the context of “safe-food” strategies or, conversely, in the development of phytoremediation strategies in which xenobiotics are sequestered in the vacuolar compartment. In this report, we obtained Arabidopsis transgenic plants overexpressing human MRP1. In these plants, expression of MRP1 did not increase plant resistance to antimony salts (Sb(III)), a classical glutathione-conjugate substrate of MRP1. However, the transporter was fully translated in roots and shoots, and targeted to the plasma membrane. In order to investigate the functionality of MRP1 in Arabidopsis, mesophyll cell protoplasts (MCPs) were isolated from transgenic plants and transport activities were measured by using calcein or Sb(III) as substrates. Expression of MRP1 at the plasma membrane was correlated with an increase in the MCPs resistance to Sb(III) and a limitation of the metalloid content in the protoplasts due to an improvement in Sb(III) efflux. Moreover, Sb(III) transport was sensitive to classical inhibitors of the human MRP1, such as MK571 or glibenclamide. These results demonstrate that a human ABC transporter can be functionally introduced in Arabidopsis, which might be useful, with the help of stronger promoters, to reduce the accumulation of xenobiotics in plants, such as heavy metals from multi-contaminated soils.  相似文献

Despite numerous studies on the response of Mediterranean ecosystems to fire, few have measured the respective resilience of vegetation and fauna compartments. For 28 years, we conducted an annual monitoring of avifauna composition and vegetation structure (cover profile) following a severe wildfire in a holm oak (Quercus ilex) stand in southern France. Our aim was to estimate the time necessary for this bird–vegetation system to return to a state analogous to its pre-fire state. In the burned plots, low herbaceous and shrub layers were gradually replaced by higher, woody layers of vegetation. Neither bird species richness nor inter-annual bird species turnover showed significant differences from one year to the next over the study period. In contrast, bird species composition did change steadily, leading to an almost complete replacement of early-successional species by late-successional ones. Using the first axes of multivariate analyses as ‘proxy variables’ of vegetation or avifauna recoveries, we estimated by extrapolation the recovery times of these two ecosystem components at ca. 50 and 35 years, respectively. Towards the end of the study period, the rate of change in avifauna composition decreased comparatively to that of vegetation structure. Our results show that holm oak woodlands are highly resilient and seem to tolerate a ~50-year fire interval, even if it remains to be assessed how resilient they would be in the case of increased fire frequency. More generally, our multivariate approach, which allows comparative estimations of resilience in different components of an ecosystem using qualitative as well as quantitative criteria, could be applied to various case studies in disturbance and restoration ecology.  相似文献

The objective of this research is to evaluate the feasibility and locations of using cellulosic biomass both from crop residues and from dedicated energy crops to supply 200-million-liter-biodiesel plants in France. The estimation of the potential amount of agricultural residue available in 2015 in each region of France is calculated. The residues considered in this study come from cereal straw and corn stover. Results show that eight out of the twenty one French regions have enough agricultural residues available to supply at least one 200 million liter biofuel plant. Region Centre has the largest potential, with enough residues to supply three to five plants. Finally, cost of supplying one biodiesel plant of 200 million liters in the region Centre is estimated. Results show that collection of biomass will be effective in an area with a radius of 58 Km to 168 Km depending of the raw material considered and its abundance. The cost of supplying a plant with miscanthus is much higher than with residues only. Thus, crop residues appear to offer a lower cost to produce biodiesel in the near term compared to a dedicated crop. Results show that production of biofuel from cellulosic biomass should not be limited by the supply of raw material, but costs of conversion to liquid fuels clearly will play a key role in the development of cellulosic biofuels. Energy prices and policies will have a significant impact on second generation biofuel development.  相似文献

Defensins have been identified as key elements of innate immunity against microbial infections. In the present study, human beta-defensin-2 (hBD-2) mRNA and peptide expression were evaluated by RT-PCR and Western blotting in normal human keratinocytes, in function of their stage of differentiation. In proliferating, non-differentiating keratinocytes generated in serum-free, low-calcium medium, a very low hBD-2 mRNA expression was found. A significantly higher expression was detected in high-calcium cultivated keratinocytes grown either as monolayers or as multilayers under submerged conditions. In an air-liquid interface culture of keratinocytes, allowing epidermis to be reconstructed, hBD-2 mRNA expression level was significantly higher than in the other conditions and displayed inter-individual variability as observed in native epidermis. The peptide was detected only in reconstructed epidermis. These results indicate that hBD-2 gene expression in normal human keratinocytes is dependent upon their stage of differentiation. The level of expression of hBD-1 mRNA was lower and that of hBD-3 was higher than that of hBD-2 in reconstructed epidermis. Exposure of reconstructed epidermis to bacterial lipopolysaccharide (LPS) resulted in an average 4-fold increase in hBD-2 mRNA 18 h after challenge, but not of hBD-1 and hBD-3 gene expression. These results show the selective regulation of hBD-2-encoding gene in an organotypic epidermal model, in response to LPS. They also provide evidence that in vitro reconstructed epidermis represents a useful model for studying regulation of expression of beta-defensins after skin challenge with pathogenic microorganisms in conditions as close as possible to the in vivo situation.  相似文献

Experiments performed in laboratory animals suggest that ionizing radiation can induce DNA damage in the germ cells of exposed individuals and lead to various deleterious effects in their progeny, including miscarriage, low birth weight, congenital abnormalities and perhaps cancer. However, no clear evidence for such effects has been found in epidemiological studies of people exposed to radiation. The predicted risks of hereditary effects of any kinds resulting from parental exposure to relatively low doses of ionizing radiation remain very low, compared to the spontaneous risks in the absence of irradiation. Irradiation of the mouse embryo can lead to various effects (lethality, growth retardation, congenital abnormalities), depending on the period of gestation at which irradiation occurs. In humans, prenatal irradiation has only been exceptionally associated with congenital abnormalities, but irradiation between weeks 8-25 has been shown to be able to induce severe mental retardation. Although being not proven, the risk of developing a childhood cancer following prenatal irradiation may also not be excluded. Like for genetic effects, the risk of adverse effects following exposure of the embryo to relatively low doses remains quite low compared to the natural risks.  相似文献

Dendritic cells (DC) are potent antigen - presenting cells that can orientate the immune response towards a Th1 or a Th2 type. DC produce chemokines that are involved in the recruitment of either Th1 cells, such as IP10 (CXCL10), Th2 cells such as TARC (CCL17) and MDC (CCL22), or non-polarized T cells such as RANTES (CCL5) and MIP-lalpha (CCL3). We investigated whether monocyte-derived DC (MD-DC) generated from healthy donors or from patients sensitive to Dermatophagoides pteronyssinus (Dpt) and exposed to the cysteine-protease Der p 1(allergen of Dpt), could upregulate the expression of chemokines involved in type 1 or type 2 T cell recruitment. MD-DC were pulsed with either Der p 1 or with LPS as the control and the chemokines produced were evaluated using ELISA and chemotaxis assays. Der p 1-pulsed DC from allergic patients showed increased TARC (CCL17) and MDC (CCL22) production without modifying IP-10 (CXCL10) release. Der p 1-pulsed DC from healthy donors showed only increased IP-10 (CXCL10) secretion. RANTES (CCL5) and MIP-lalpha (CCL3) production were similarly increased when DC were from healthy or allergic donors. The selective Th2 clone recruitment activity of supernatants from Der p 1-pulsed DC of allergic patients was inhibited by anti-TARC (CCL17) and anti-MDC (CCL22) neutralizing Abs. By using anti-IP10 (CXCL10) blocking Abs, supernatants of Der p 1-pulsed DC from healthy donors were shown to be involved in the recruitment of Th1 cells. These results suggest that in allergic patients exposed to house dust mites, DC may favour the exacerbation of the Th2 response via the increase in type 2 chemokine production.  相似文献
