Isolated abdomens of crayfish were maintained in vitro and lysine (-3H) was intracellularly injected into one of the giant motoneuron of the ventral cord ganglia by iontophoresis. Membrane potentials ranging between -60 and -70 mV were recorded all along the experiment. Light microscope radioautographs showed an intense reaction over the injected nerve cell body and the initial segment of its axon; most of the surrounding tissues were free of radioactivity when the diffusion of the injected lysine (-3H) was prevented by adding cold lysine to the bathing medium. Some exchange of label was however noted with electrically coupled axons and glial sheaths. No radioactive protein could be traced in the numerous nerve endings of the neuromuscular junction. Nonetheless when a ligature was placed on the nerve root, the amount of accumulated radioactivity was increased from 3 to 6 h. in the axon of the injected motoneuron only. Electron microscope radioautographs indicate that the fast transported proteins were mainly associated with the endoplasmic reticulum and the axonal membrane. It is concluded that the visualization of the nerve endings was limited by the dispersion of the label into the numerous thin terminal branchs of the axon; however the combination of iontophoretic injection and radioautography permits to trace endogenous protein along the axon and to study molecular exchanges with other cells.  相似文献

A detailed comparative assessment was made of the use of solid-phase-coupled antibodies in radioimmunoassay, by using an assay for human placental lactogen as a model system. The major advantages of the solid-phase technique are: (1) in common with the use of a second antibody, it is universally applicable; (2) separation can be carried out rapidly; (3) in contrast with some other techniques, the separation of antibody-bound and free antigen is virtually complete. The disadvantages when compared with other procedures are: (1) a considerable proportion of the antibody may be lost during the initial coupling reaction; (2) the tubes must be continuously mixed during incubation, and much effort is expended in removing and replacing the caps; (3) there is a decrease in the apparent affinity constant of the antibody after coupling, which is reflected in a lower sensitivity of the assay system. It is concluded that solid-phase antibodies are of greatest value in those systems in which the supply of antiserum is abundant, and in which the achievement of high sensitivity is not a requirement.  相似文献

The role of phospholipases from inflammatory macrophages in demyelination   总被引:3,自引:0,他引:3  
Activated macrophages harvested from rat peritoneum were shown to contain phospholipase A1, A2 and lysophospholipase activities which were defined on a series of radiolabelled phospholipid substrates. During in vitro culture of these elicited macrophage populations, phospholipase enzymes were secreted into the culture medium. Radiolabelled myelin, prepared from young rats after intracerebral injection of14C acetate, was used as a substrate to analyze the susceptibility of central nervous system (CNS) myelin to attack by cell-associated and secreted macrophage enzymes. Homogenates of peritoneal macrophages degraded the myelin lipids at acid pH; phosphatidyl choline (PC) and ethanolamine phosphatide (EP) were both degraded with liberation of free fatty acid and small amounts of lysolipids. The ethanolamine lipids were most vulnerable; up to 20% of this fraction was degraded in six hours. Selected batches of macrophage culture supernatant similarly degraded the myelin EP at acid pH. These results suggest that phospholipase enzymes, released from activated macrophages in close proximity to the myelin sheath, may participate in primary demyelination in inflammatory CNS lesions.  相似文献

Maltoporin, a protein spanning Escherichia coli outer membranes, modifies electrical conductance of membranes due to its channel-forming properties. This observation was made by conductance measurements across planar bilayers which were derived from unextracted, isolated outer membrane vesicles using a porin-deficient E. coli strain. Alternatively, proteoliposomes reconstituted with detergent-solubilized homogeneous maltoporin and phospholipids were used. With either membrane preparation, channel conductance was observed, although no discrete conductance levels were detected. The presence of lipopolysaccharide, a bacterial glycolipid, was not required, nor did it affect channel activity. In the presence of the water-soluble periplasmic maltose-binding protein, conductance fluctuations occurred in discrete steps, demonstrating opening and closing events of channels. Multiple step sizes (1/3, 2/3 and 1 ns in 1 M KCl) in single channel traces suggest cooperative opening and closing of up to three channels. The action of maltose-binding protein is highly asymmetrical, and its affinity to maltoporin is very high (KD = 1.5 X 10(-7) M). Association of maltose-binding protein to maltoporin shifts, for a given polarity, the equilibrium between open and closed states in favour of closed states. This result matches earlier in vivo studies, and supports the physiological significance of the observations made.  相似文献

A study of the anterior sensory receptors of male and female Asplanchna brightwelli by scanning electron microscopy reveals some important differences in the region surrounding the mouth. In the male, the ventrolateral sensory bristles, the pseudotrochus, the inner and the outer buccal tufts and the mastax receptors are absent. The oral receptors are reduced. Transmission electron microscopy of these receptors shows that they consist of ciliated sensory cells surrounded by epithelial supporting cells. The distal ends of the cilia of the mastax receptors are modified; the cilia of the other receptors differ only in their length and rootlet structure from the locomotor cilia of the cingulum. A consideration of the feeding behaviour of Asplanchna leads us to suppose that these sensory cilia function in mechanoreception and in chemoreception.  相似文献

Summary Six Algerian patients with 0 thalassemia are presented, in addition to the two patients already reported (Godet et al., 1977). Family studies indicate that all the patients had homozygous thalassemia characterized by absence of globin chain synthesis in peripheral blood. The clinical severity varies from one family to the other and within the same family, from typical Cooley's anemia to thalassemia intermedia and appears to be related to the child death rate observed in each family. The / biosynthetic ratio was 0.36–0.40 in seven patients and 0.2 in the most seriously affected patient. The mRNA content in peripheral reticulocytes was less than 1.5% of mRNA in seven patients and 13.3% in one patient. These results indicate that Algerians homozygous for thalassemia are heterozygous at the clinical, biochemical and molecular levels.  相似文献

Changes in the ultrastructure and lipid composition of etioplasts have been evaluated in three regions from the base to the tip of 8-day-old darkgrown wheat leaves and in the upper-2/3 region of etiolated leaves of different ages. In developing darkgrown tissues, the main morphological changes that etioplasts undergo consist of an increase in the amount of thylakoïds which, in the most mature etioplasts, align in parallel arrays. Concomitantly, galactolipids and sulfolipid form an increasing proportion of the total lipids. Trans-3-hexadecenoic acid was not detectable in phosphatidylglycerol (PG) of etioplasts showing appressed thylakoïds isolated from 5-day-old leaves, but was present in significant amounts in etioplasts in the basal part of 8-day-ols leaves in which membrane appression was barely visible. The proportions of this acid increase as etioplasts develop, reaching 25% of the PG fatty acids (1.2% of the total fatty acids) in the most differentiated etioplasts. DNA levels were measured in the spinal cords of Lewis rats during the development of and recovery from experimental allergic encephalomyelitis (EAE). Spinal cord DNA was first increased 11 days after immunizing the rats with guinea pig myelin and rose to levels four times that of the Freund's adjuvant controls at day 14, then subsided after day 22. Spinal cord DNA was still 150% of control levels 60 days after immunization. These DNA changes were compared with fluctuations in spinal cord acid proteinase in the same animals. Acid proteinase activity in EAE spinal cord increased later than the rise in DNA and attained a level of 170% of control at days 15-17, then subsided. Spinal cord DNA was higher in rats immunized with whole myelin than in those administered equivalent amounts of purified myelin basic protein. Furthermore DNA was higher in spinal cords of rats immunized with a larger dose of myelin (1.0 mg) than with a lower amount (0.5 mg). Various protease inhibitors including pepstatin, nitrophenyl p-guanidino benzoate, polylysine, and dipropionyl rhein, previously shown to protect Lewis rats against EAE, suppressed the increase of DNA in the spinal cord. Measurement of DNA increases in the spinal cord of EAE animals provides a convenient reproducible measurement of the severity of inflammation in the CNS and provides an objective criterion for assessment of the efficacy of various agents screened as possible therapeutic treatment for multiple sclerosis.  相似文献

Summary The spinning apparatus of the uloborid spider Polenecia producta was studied to complete previous studies on the same family of spiders. The structure of spinnerets and spigots, under scanning electron microscopy, and the main anatomical and histochemical characteristics of the spinning glands of adult females and males are described. In addition some observations on the spinning apparatus at three successive stages of development are made. There are nine kinds of silk glands in Polenecia, i.e. one more (aciniform — B glands) than found in other uloborids. The spinning apparatus of Polenecia is, therefore, the most complex so far known. It is also more complex than that presently known of Araneoidea. The characteristics of the spinning glands of Polenecia are compared with those of other uloborids. Present knowledge of the spinning apparatus of uloborids leads to a renewed discussion of the origin of the orb web in this family and in araneids. It is concluded that these two types of orb webs emerged from independent evolutionary processes.  相似文献
