Summary Metaphase chromosomes isolated from human fibroblasts by lysis of mitotic cells in the presence of the intercalating DNA-specific fluorochrome propidium iodide appear relatively long, even after exposure to vinblastine sulfate overnight. Therefore, they can be easily banded and thereby unequivocally identified. Chromosomes isolated in this way may be employed in flow analysis and sorting without loosing the inducibility of their band patterns.  相似文献

A model is proposed for the temporal characteristics of X-and Y-type responses of ganglion cells in the primate retina. The main suggestions of the model are: (I) The X-type temporal response is determined primarily by the delay between center and surround contributions. (II) The Y-type response is generated in the inner plexiform layer by a derivativelike operation on the bipolar cell's input, followed by a rectification in the convergence of these inputs onto the Y-ganglion-cell. (III) The derivative-like operation is obtained by recurrent inhibition in the dyad synaptic structure.The X-and Y-type responses predicted by the model, for a variety of stimuli, were examined and compared with available electrophysiological recordings. Finally, certain predictions derived from the model are discussed.  相似文献

Summary Nerve growth factor (NGF) was localized in the submandibular, sublingual, and parotid salivary glands of male and female diabetic mice and their normal littermates by immunoperoxidase staining usingp-phenylenediamine-pyrocatechol as a chromogen for the cytochemical demonstration of peroxidase activity. In the normal male submandibular gland, immunoreactive NGF was localized in the apical regions of granular, intercalated and collecting duct cells, while in the normal female submandibular gland, NGF was present throughout the cytoplasm of granular duct cells. The localization of NGF in the diabetic male and female submandibular glands was similar and resembled that of the normal female. NGF immunoreactivity was also observed in the striated duct cells in the sublingual and parotid glands of all four types of mice.The sympathetic innervation of the submandibular glands of normal and diabetic mice was demonstrated using glyoxylic acid-induced histofluorescence. The pattern of sympathetic innervation and the intensity of catecholamine fluorescence was consistently different in the four types of mice. In the normal male submandibular gland the fluorescence was very intense, particularly in nerves adjacent to the granular ducts. In the normal female submandibular gland, the fluorescence was weak, while in the diabetic male and female the fluorescence was moderate.The correlation between the intensity of the immunocytochemical staining for NGF and the catecholamine fluorescence adjacent to the granular ducts suggests a trophic influence of the NGF-containing granular ducts on their sympathetic innervation.  相似文献

Effects of rabbit anti-embryonal carcinoma IgG on embryonal carcinoma cells and their differentiated derivatives were studied at different levels of cell-cell interaction. Fab fragments of anti-EC IgG were found to inhibit aggregation of the majority of EC cell lines. Two, however, were insensitive. Anti-EC Fab fragments act also on the transfer of metabolites between EC cells: the rescue of HPRT? EC cells by HPRT+ EC cells in selective medium is abolished. These findings are correlated with the disappearance of tight and gap junctions from the surface of EC cells (Dunia et al., 1979). The presence of the surface structure involved in the action of anti-EC Fab fragments was tested by absorption experiments followed by decompaction test on PCC4 Aza R1 cells. All EC cell lines and two embryonic cell lines—a trophectodermal and an endodermal line—were found to bear material absorbing the decompacting activity. The results are discussed in terms of state of differentiation of the cell lines and of complexity of aggregation of embyronic cells.  相似文献

Die nächtliche Zugunruhe setzt auch unter konstanten Lichtbedingungen (Dauerdunkel 0,06 Lux, Dauerhell 250 Lux) etwa mit der Sonnenuntergangszeit ein und erreicht im Frühjahr etwa um 21h ihr Maximum (etwas später bei Dauerhell).2. Während der Nacht werden vom zugaktiven Rotkehlchen in stärkerem Maße als bei Tag die dunkleren Abteilungen einer Lichtorgel aufgesucht.3. Die höchste Zugaktivität wird bei Beleuchtungsstärken von 1–10 Lux erreicht, aber auch bei völliger Dunkelheit wird noch etwa 7 % der Maximalaktivität gemessen. Sommer-Nachtaktivität erreicht ebenfalls bei ca. 5 Lux ein Maximum, erlischt aber fast völlig bei Beleuchtungsstärken unter 1 Lux.4. Bei dunklem Untergrund ist die gemessene Aktivität bei gleicher Beleuchtungsstärke deutlich geringer als bei hellem Untergrund.5. Niedrigere Tagesbeleuchtungsstärken im Versuchsraum (250 gegenüber 700 Lux) bewirken ein Ansteigen der Zugaktivität in der folgenden Nacht, bei gleichzeitig zunehmender Futteraufnahme während des Tages.6. Einzelne Lichtpunkte wirken aktivitätssteigernd unabhängig von der von ihnen bewirkten Beleuchtungsstärke. Eine größere Zahl von Lichtpunkten, gleichmäßig über die Decke verteilt, hemmt dagegen die Zugaktivität, auch wenn die dabei erreichte Beleuchtungsstärke höher ist als bei einer Diffusionsbeleuchtung, unter der dieselben Tiere eine höhere Aktivität zeigen.
Nocturnal migratory restlessness in the robin under different light conditions and patterns of light spots
Summary 1. Nocturnal migratory restlessness even under constant LL or DD lighting conditions (250 Lux, 0,006 Lux) starts at the time of sunset, and in spring reaches a maximum at 9 pm under constant dark, and a little later in constant light (Fig. 1).2. At night, during nocturnal restlessness, the birds tend to be more active in the darker parts of a set of differently lighted chambers than during daytime (Fig. 3 b).3. Highest migratory restlessness in spring and autumn is recorded under an illumination of 1 to 10 Lux, though even in total darkness a high level of activity persists. Nocturnal activity in summer also reaches a maximum at an illumination level of 5 Lux, but drops to zero at intensities below 1 Lux (Fig. 4 a).4. In cages painted black the nocturnal activity is lower than in white cages with the same overhead illumination intensity (Fig. 5).5. Low illumination intensity during daytime (250 against 700 Lux) causes high activity during the following night and raises diurnal food consumption considerably.6. A few single light spots (stars) on the ceiling strongly activate migratory restlessness (almost independent of their brightness), while higher numbers of evenly spaced light spots lower the birds activity even though total illumination intensity is higher than under the diffuse control lighting under which the birds show a higher amount of activity (Fig. 7, 8).

Endo-beta-N-acetylglucosaminidase H released four major oligosaccharides from high-mannose glycopeptides prepared from embryonal carcinoma cells. The oligosacchaides were indistinguishable from (Man)9GlcNAc, (Man)8GlcNAc, (Man)7GlcNAc, and (Man)6GlcNAc isolated from fibroblasts. This result suggests that the biosynthetic pathway of asparagine-linked oligosaccharides in early embryonic cells is controlled as in adult cells, at least to the initial stage of processing of the nascent oligosaccharide transferred from lipid-linked intermediate.  相似文献

A method of fractionation of hnRNP constituents adaptable to large-scale preparation is presented. It is based on differential resistance to salt dissociation of the two classes of units of hnRNP, the 30--50S monoparticles and the heterogeneous complexes. The monoparticle proteins were released from hnRNP by 0.4 M NaCl. They were separated from the salt-resistant RNP corresponding to the heterogeneous complexes in three steps: chromatography on DEAE-cellulose, high-speed centrifugation, and Bio-Gel chromatography. The latter chromatography permitted a first fractionation of monoparticle proteins according to molecular weight. Such fractions may serve for purification of individual proteins of molecular weight below 80 000. After the two first steps, two fractions of salt-resistant RNP were obtained. In addition to heterogeneous RNA up to 30 S, small nuclear RNAs were detected which represented 6% of total RNA. The protein pattern was complex, and no clear-cut segregation of groups of proteins could be observed between the two fractions. They were both highly enriched in phosphoproteins as compared to nomoparticle proteins. In another fraction corresponding to the void volume of Bio-Gel chromatography, one-third of the RNA was small nuclear RNA. It is suggested that this fraction contains snRNP in addition to free proteins of molecular weight above 80 000 and to salt-resistant RNP similar to those described above but of small size.  相似文献

Distinctive properties of fucosyl glycopeptides on human teratoma cells   总被引:1,自引:0,他引:1  
Fucose-labeled glycopeptides from four human teratoma cell lines of independent origin show similar elution profiles on Sephadex G-50 column chromatography. The fucosyl glycopeptides elute in two major regions: one near the void volume, the other in fractions corresponding to a molecular weight of 2500-3000. These elution profiles are very different from those obtained with the other human cell lines examined which included 3 lymphomas, 2 colon carcinomas, and HeLa. The elution profiles of the human teratomas, however, show remarkable similarities to those obtained with murine embryonal carcinoma cell culture and early mouse embryos. These results suggest that the excluded G-50 fraction may well contain glycopeptides playing a role in mammalian embryogenesis.  相似文献

A strain of Fusarium solani (Mart.) Sacc. (IMI-216517), isolated from a patient of mycotic keratitis, produced experimental keratomycosis in albino rabbit cornea and survived in internal tissues of albino mice for varying periods. Alantolactone, isolated from the plant — Inula racemosa Hook. f. exhibited marked in vitro fungistatic activity against this strain of F. solani at 100–200 g/ml concentrations. The strain was less sensitive to amphotericin B and showed more acid than alkaline proteinase and phosphatase activities.Communication No. 2526.  相似文献
