Two methods, digestion and elution, were used to recover parasitic nematodes from 470 flatfish belonging to species in the family Pleuronectidae. Samples of similar fish were collected from market lots; half of each sample was subjected to digestion, and half was subjected to elution (sedimentation). The edible (flesh) and the inedible (viscera) portions of each fish were analyzed separately. The total number of nematodes recovered by digestion was 1,110, which was not significantly greater than the 922 nematodes recovered by elution. However, digestion recovered 1,062 nematodes of the anisakine genera Anisakis and Phocanema, which are potentially pathogenic for human consumers of raw of semiraw fish. This number is significantly greater than the 608 pathogenic nematodes recovered by elution. Digestion also recovered 242 more nematodes from the edible flesh than did elution. Conversely, more nonpathogenic nematodes were recovered by elution. Approximately half the fish (240) had been collected in Boston markets, and the other half (230) had been collected in San Francisco markets. Fish from San Francisco each contained an average of eight nematodes, and those from Boston contained an average of less than one nematode per fish.  相似文献

The synthesis of a membrane-bound MalE β-galactosidase hybrid protein, when induced by growth of Escherichia coli on maltose, leads to inhibition of cell division and eventually a reduced rate of mass increase. In addition, the relative rate of synthesis of outer membrane proteins, but not that of inner membrane proteins, was reduced by about 50%. Kinetic experiments demonstrated that this reduction coincided with the period of maximum synthesis of the hybrid protein (and another maltose-inducible protein, LamB). The accumulation of this abnormal protein in the envelope therefore appeared specifically to inhibit the synthesis, the assembly of outer membrane proteins, or both, indicating that the hybrid protein blocks some export site or causes the sequestration of some limiting factor(s) involved in the export process. Since the MalE protein is normally located in the periplasm, the results also suggest that the synthesis of periplasmic and outer membrane proteins may involve some steps in common. The reduced rate of synthesis of outer membrane proteins was also accompanied by the accumulation in the envelope of at least one outer membrane protein and at least two inner membrane proteins as higher-molecular-weight forms, indicating that processing (removal of the N-terminal signal sequence) was also disrupted by the presence of the hybrid protein. These results may indicate that the assembly of these membrane proteins is blocked at a relatively late step rather than at the level of primary recognition of some site by the signal sequence. In addition, the results suggest that some step common to the biogenesis of quite different kinds of envelope protein is blocked by the presence of the hybrid protein.  相似文献

Summary Insecticide treatment of a small, Appalachian forest stream caused massive downstream insect drift and reduced aquatic insect densities to <10% of an adjacent untreated reference stream. Reduction in breakdown rates of leaf detritus was accompanied by differences in quantity and composition of benthic organic matter between the two streams. Following treatment, transport of particulate organic matter was significantly lower in the treated stream than in the reference stream whereas no significant differences existed prior to treatment. Our results indicate that macroinvertebrate consumers, primarily insects, are important in regulating rates of detritus processing and availability to downstream communities.  相似文献

Effects of 2,4-dinitrophenol (DNP) and several other substituted phenols on permeability of barley roots (Hordeum vulgare var. Trebi) to ions were assayed as a function of pH and phenol concentration. Solutions containing 0.1 micromolar undissociated DNP increase the permeability of barley root cells to small ions such as K+, Na+, Ca2+, and Cl with no inhibition of respiration. Undissociated forms of the other phenols increase permeability also, but they are less effective than DNP. Only the undissociated DNP is effective. Anionic DNP does not increase permeability or inhibit ion uptake, although it is the major species accumulated by the roots, both at pH 5 and pH 7. At pH 7, in contrast to pH 5, 10 micromolar DNP has no effect on ion permeability of barley roots yet it uncouples oxidative phosphorylation of barley root mitochondria. This indicates that the all too common use of DNP as a test for active transport or involvement of ATP synthesis can be misleading.  相似文献

By analysing rumen content samples from 105 dead specimens, the annual diet of the Roe deer ( Capreolus capreolus ) in the New Forest, Hampshire, was determined over the period from November 1970 to March 1973. Bramble and rose formed the bulk of the diet throughout the year, comprising the largest food fraction in all months except January and April. In January to March, foliage from felled conifers, Calluna , and ivy were major foods, plus lesser amounts of grasses, herbs and fungi. From May until early winter, herbs and grasses were important. New, tender, spring and summer growth from broadleaved deciduous trees, bilberry and holly was eaten in appreciable quantities. Acorns were a characteristic autumnal food. Abundant potential foods which Roe deer seldom selected included rushes, sedges, mosses, bark and dead hardwood leaves. Widespread species that Roe rarely consumed were Corsican pine, Erica tetralix, E. cinerea , gorses, bog myrtle and the grasses Agrostis setacea, Brachypodium sylvaticum, Deschampsia caespitosa and Molinia caerulea .  相似文献
