The competitive neuromuscular blocking agents, gallamine and pancuronium, enhanced the nictitating membrane contraction, in the cat, resulting from muscarine ganglionic transmission. Inhibition of ganglionic muscarinic hyperpolarization, in response to short tetanic bouts of preganglionic cervical sympathetic stimulation, was an associated event and is considered by us to be causally related. The neuroleptic drug, haloperidol, enhanced ganglionic hyperpolarization under similar stimulatory conditions, and reduced the nictitating membrane contraction elicited via ganglionic muscarine pathways, effects opposite to those produced by the skeletal muscle relaxants. Apomorphine reduced both ganglionic hyperpolarization and the ganglionic muscarinic-induced nictitating membrane contractions. The action of gallamine and pancuronium conforms to a speculative cholinergic antagonism at the specific muscarinic receptors, termed Mi, on the ganglionic dopaminergic interneuron. Haloperiodol and apomorphine are anticipated to be exerting distinct antagonistic and agonistic actions, respectively, on prejunctional dopamine receptors of the ganglionic interneuron. Ganglionic slow depolarization mediated through the muscarinic receptors, termed Me, was unaltered by any of the agents studied.  相似文献

The interaction of lipoprotein lipase (LpL) and a nonhydrolyzable phosphatidylcholine, 1,2-ditetradecyl-rac-glycero-3-phosphocholine (C14-ether-PC), has been studied by several physical methods. Analysis of the circular dichroic spectrum of LpL gave the following fractional conformation: 35% alpha-helix, 30% beta-pleated sheet, and 45% remaining structure. No significant change in the circular dichroic spectrum of LpL was observed on addition of C14-ether-PC vesicles. The quenching of LpL fluorescence by acrylamide and iodide ion was decreased only slightly by addition of C14-ether-PC vesicles. Addition of LpL to sonicated C14-ether-PC vesicles containing entrapped carboxyfluorescein caused the release of less than 15% of the vesicle contents in 20 min, indicating that the enzyme did not disrupt the structure of the lipid. In contrast, greater than 80% of the vesicle contents were released with the addition of apolipoprotein A-I to an identical vesicle preparation. The temperature dependence of the fluorescence polarization of 1,6-diphenyl-1,3,5-hexatriene incorporated into C14-ether-PC vesicles was not significantly altered by the addition of LpL. When LpL is added to vesicles, the bilayer structure of the vesicles is not disrupted as observed by freeze-fracture electron microscopy. However, at low ionic strength (0.1-0.25 M NaCl) significant aggregation of intact vesicles is observed by light scattering and electron microscopy. Vesicle aggregation is prevented and reversed by 1 M NaCl and by heparin. These data demonstrate that LpL binds to the surface of a lipid interface, without dramatic changes in lipid bilayer or protein structure.  相似文献

Long-distance signalling from roots to shoots assessed: the flooding story.   总被引:16,自引:0,他引:16  
Several kinds of signal may be generated when roots are exposed to an environmental stress. Some, but not all, are conveyed to shoots in the transpiration stream. Principles are summarized that may help establish experimentally the presence or intensity of root signals transported by transpiration. In many dryland species, flooding of the soil induces developmental responses in the shoot such as epinastic leaf curvature, stomatal closure and slowing of leaf expansion. These reactions compensate for diminished input of resources from the roots. They lend themselves to the study of root-to-shoot signalling by commencing after a time lag of only a few hours, by persisting for several days and by being highly reproducible. Evidence implicating chemical and hydraulic signals in promoting stomatal closure and epinastic curvature in flooded plants is reviewed. Further progress will depend upon examining a wider range of putative signals, accounting for any interactions between them and improving methods for the evaluation of signal durability in transit, and effectiveness at target sites.  相似文献

The efficacy of prophylactic penicillin and of 14 valent pneumococcal vaccine in preventing pneumococcal infection in homozygous sickle cell (SS) disease was investigated in 242 children aged 6 months to 3 years at entry. In the first five years of the trial there were 11 pneumococcal infections in the pneumococcal vaccine treated group, 10 by serotypes present in the vaccine. Type 23 accounted for five of these, and there was evidence of higher infection rates in those given the vaccine before age 1. No pneumococcal isolations occurred in the penicillin group while receiving penicillin, although four isolations occurred within one year of stopping penicillin. Probably the most effective prophylaxis against pneumococcal infection requires penicillin beyond the age of 3. The age at which pneumococcal vaccine should be given must await further data on antibody response and clinical efficacy in these patients.  相似文献
