Several independent criteria indicate 2 mol of terbium(III) bind to yeast enolase in the absence of substrate-fluorescence titrations of enzyme and metal, effects on thermal stability and published ultrafiltration and inhibition experiments. These measurements also suggest the terbium binding sites are the same as those normally occupied by “conformational” magnesium. Terbium binds much more strongly than magnesium, however, and measurements of the kinetics of the absorbance change in the terbium-enzyme on adding excess EDTA suggest the terbium-enzyme dissociation constant is about 1500 that of the magnesium-enzyme. Measurements of enzyme activity as a function of substrate concentration show that terbium permits no enzymatic activity. However, magnesium competes more effectively with the lanthanide if the substrate analogue 3-aminoenolpyruvate 2-phosphate (AEP) is present.The fluorescence of the lanthanide is not readily observed on exciting the terbium-enzyme at 280 nm, indicating the absence of tyrosines or tryptophans in the coordination sphere of the metal. Excitation of terbium using 488 nm radiation from an argon ion laser shows the fluorescence of the metal is enhanced by binding to the enzyme. EDTA and carbonate have similar effects. This suggests carboxyl groups are involved in binding metal at the conformational sites of yeast enolase. Measurements of lifetimes of enzyme-bound terbium in the presence and absence of D2O indicated three moles of water remained on each of the bound metals, independently of the buffer used. If enzyme-bound terbium is assumed to be nine-coordinate, the metal must bind to six groups from the enzyme. The presence of substrate does not markedly affect the emission spectrum of the bound terbium or the number of water molecules remaining on the metal, but calorimetric measurements show that substrate binds to the terbium enzyme.  相似文献

This study analyzed distributions of Euclidean displacements in gaze (i.e. “gaze steps”) to evaluate the degree of componential cognitive constraints on audio-visual speech perception tasks. Children performing these tasks exhibited distributions of gaze steps that were closest to power-law or lognormal distributions, suggesting a multiplicatively interactive, flexible, self-organizing cognitive system rather than a component-dominant stipulated cognitive structure. Younger children and children diagnosed with an autism spectrum disorder (ASD) exhibited distributions that were closer to power-law than lognormal, indicating a reduced degree of self-organized structure. The relative goodness of lognormal fit was also a significant predictor of ASD, suggesting that this type of analysis may point towards a promising diagnostic tool. These results lend further support to an interaction-dominant framework that casts cognitive processing and development in terms of self-organization instead of fixed components and show that these analytical methods are sensitive to important developmental and neuropsychological differences.  相似文献

Paraquat (10–30 μm) exerted a dose-dependent and light-dependent toxicity on Chlorella sorokiniana. Paraquat was also seen to increase the superoxide dismutase content of these cells and to cause the appearance of a new electrophoretically distinct isozyme. Cells grown in the absence of paraquat contained one manganese-superoxide dismutase and two iron-superoxide dismutases, while the paraquat-grown cells contained an additional manganese-superoxide dismutase. Cells which were grown in the presence of 25 μm Paraquat, and which therefore possessed elevated levels of superoxide dismutase, were resistant to 30 μm Paraquat, whereas control cells were bleached and killed by this level of Paraquat. Electron micrography and chemical analysis revealed that Paraquat decreased the starch content of the cells and caused a failure of dividing cells to separate. It appears that Paraquat increases the photoproduction of O2? in C. sorokiniana and that an increase in the cell content of superoxide dismutase is an adaptive response which provides protection against this herbicide.  相似文献

Muscarinic and β-adrenergic ligands associate with polymorphonuclear leukocytes to show high-affinity, saturable accumulation. This association can be distinguished from specific receptor binding by its temperature dependence, sensitivity to pH, requirement of an energy source, inhibition by ionophores, and inhibition by a variety of permeable basic amines.Our results suggest that these amines accumulate in acidic lysosomes which are plentiful in these cells.
