Attempts were made to cryopreserve in liquid nitrogen six different photomixotrophic suspension cultured lines of five different species:Amaranthus powellii Wats.,Datura innoxia Mill.,Glycine max (L.) Merr.,Gossypium hirsutum L. andNicotiana tabacum xNicotiana glutinosa L. fusion hybrid. Only theD. innoxia line, DAT, and theG. max line, SB1, could be successfully recovered as viable, growing, dark green cultures. The successful method utilized a preculture treatment of from 2 to 8 days in a medium containing 3% starch and 3% sorbitol for DAT and 3% sucrose and 3% sorbitol for SB1 cells. The cells survived if frozen with 10% dimethylsulfoxide (DMSO) and 9.1% sorbitol or with 10% DMSO and 8% sucrose. Following a programmed slow-cooling, the cells were thawed in a 40° C bath and could be recovered directly when added to fresh liquid medium. Cryostorage of these lines will save labor and prevent further genetic changes from occurring in these unique suspension cultures.  相似文献

We tested the Plant Vigor Hypothesis by determining the distribution of galls formed on leaves of witch hazel, Hamamelis virginiana , by the aphid Hormaphis hamamelidis , and by determining various factors that affect the fecundity of the gall-forming fundatrices. We also studied the role of the fundatrix in host plant manipulation. While the mean number of galls per leaf was low, galls had an aggregated distribution among leaves. Among trees, the average number of galls per leaf was not related to the mean leaf size, contrary to the preference prediction of the Plant Vigor Hypothesis. While fundatrices preferred the distal leaves of buds, which grew more than the proximal leaves, being on distal leaves conferred no increase in fecundity for fundatrices, contrary to predictions of the Plant Vigor Hypothesis. Gall size was the factor that explained the largest proportion of variation in fundatrix fecundity; fundatrix size explained somewhat less of the variation. Also, gall position on the leaf, number of aphid galls on the leaf, and on which leaf of the bud the gall was located all played small, statistically significant roles in explaining fundatrix fecundity, but their effects were variable between experiments. Removal of fundatrices shortly after galls had enclosed them limited the growth of galls, indicating the role of the fundatrices in gall growth. We compare and contrast this system versus other gall-forming insects, as well as discuss the adaptive significance of the aphid manipulation of the host plant. Much of the data contradict predictions of the Plant Vigor Hypothesis, and we discuss how gall size, as a measure of plant growth caused by insect manipulation, explains the observed patterns.  相似文献

The Alzheimer's disease-linked amyloid-beta precursor protein (APP) belongs to a superfamily of proteins, which also comprises the amyloid-beta precursor-like proteins, APLP1 and APLP2. Whereas APP has been identified in both lower and higher vertebrates, thus far, APLP1 and 2 have been characterized only in human and rodents. Here we identify the first nonmammalian APLP2 protein in the South African claw-toed frog Xenopus laevis. The identity between the Xenopus and mammalian APLP2 proteins is approximately 75%, with the highest degree of conservation in a number of amino-terminal regions, the transmembrane domain and the cytoplasmic tail. Furthermore, amino acid residues known to be phosphorylated and glycosylated in mammalian APLP2 are conserved in Xenopus. The availability of the Xenopus APLP2 protein sequence allowed a phylogenetic analysis of APP superfamily members that suggested the occurrence of APP and preAPLP lineages with their separation predating the mammalian-amphibian split. As in mammals, Xenopus APLP2 mRNA was ubiquitously expressed and alternatively spliced forms were detected. However, the expression ratios between the mRNA forms in the various tissues examined were different between Xenopus and mammals, most prominently for the alternatively spliced forms containing the Kunitz protease inhibitor-coding region that were less abundantly expressed than the corresponding mammalian forms. Thus, the identification of APLP2 in Xenopus has revealed evolutionarily conserved regions that may help to delineate functionally important domains, and its overall high degree of conservation suggests an important role for this APP superfamily member.  相似文献

Oxidative stress contributes to the vascular and neurological complications of diabetes mellitus. The aim was to evaluate the effects of treatment with the radical scavenger and transition metal chelator, alpha-lipoic acid, on endothelium-dependent relaxation of the mesenteric vasculature and on superior cervical ganglion blood flow in 8 week streptozotocin-induced diabetic rats. alpha-Lipoic acid effects on small nerve fiber-mediated nociception were also monitored. For the in vitro phenylephrine-precontracted mesenteric vascular bed, diabetes caused a 31% deficit in maximum endothelium-dependent relaxation to acetylcholine, and a 4-fold reduction in sensitivity. alpha-Lipoic acid gave 85% protection against these defects. Acetylcholine responses are mediated by nitric oxide and endothelium-derived hyperpolarizing factor: isolation of the latter by nitric oxide synthase blockade revealed a 74% diabetic deficit that was halved by alpha-lipoic acid. Superior cervical ganglion blood flow, 52% reduced by diabetes, was dose-dependently restored by alpha-lipoic acid (ED(50), 44 mg/kg/d). Diabetic rats exhibited mechanical and thermal hyperalgesia, which were abolished by alpha-lipoic acid treatment. Thus, diabetes impairs nitric oxide and endothelium-derived hyperpolarizing factor-mediated vasodilation. This contributes to reduced neural perfusion, and may be responsible for altered nociceptive function. The effect of alpha-lipoic acid strongly implicates oxidative stress in these events and suggests a potential therapeutic approach.  相似文献

Winter frosts caused by radiative cooling were hypothesized to limit successful reintroduction of Hawaiian plants other than Acacia koa to alien‐dominated grasslands above 1700 m elevation. We determined, in the laboratory, the temperature at which irreversible tissue damage occurred to Metrosideros polymorpha leaves. We also conducted a field study of this species to determine if (1) leaf damage was correlated with sub‐zero leaf temperatures, (2) radiative cooling could be moderated by canopies of A. koa, and (3) low soil temperatures contributed to seedling damage. The last was evaluated by thermally buffering seedlings with water‐filled bladders placed at their base to keep roots warm, or by installing a radiation shield to reduce early morning transpiration when water uptake from cold soils would be least. Leaf temperatures were monitored between midnight and 7:00 a.m. using fine‐wire thermocouples, and leaf damage was recorded monthly. In the laboratory, supercooling protected leaves from mild sub‐zero temperatures; irreversible tissue damage occurred at about ?8°C. In the field, leaf damage was strongly correlated with degree‐hours below freezing. Unprotected seedlings suffered the greatest leaf damage. Those sheltered under A. koa trees rarely experienced temperatures below ?3°C, and damage was minimal. Shaded and thermally buffered seedlings suffered less damage than unprotected plants, probably due to elevated leaf temperatures rather than improved water relations. Using A. koa or artificial devices to reduce radiative cooling during winter nights should enhance establishment of M. polymorpha in high‐elevation rangeland.  相似文献

We describe a population of colonial cyanobacteria (waterwarts) that develops as the dominant primary producer in a bottom‐fed, O2‐poor, warm spring in the Cuatro Ciénegas karstic region of the Mexican Chihuahuan Desert. The centimeter‐sized waterwarts were suspended within a central, conically shaped, 6‐m deep well by upwelling waters. Waterwarts were built by an Aphanothece‐like unicellular cyanobacterium and supported a community of epiphytic filamentous cyanobacteria and diatoms but were free of heterotrophic bacteria inside. Sequence analysis of 16S rRNA genes revealed that this cyanobacterium is only distantly related to several strains of other unicellular cyanobacteria (Merismopedia, Cyanothece, Microcystis). Waterwarts contained orderly arrangements of mineral crystallites, made up of microcrystalline low‐magnesium calcite with high levels of strontium and sulfur. Waterwarts were 95.9% (v/v) glycan, 2.8% cells, and 1.3% mineral grains and had a buoyant density of 1.034 kg·L?1. An analysis of the hydrological properties of the spring well and the waterwarts demonstrated that both large colony size and the presence of controlled amounts of mineral ballast are required to prevent the population from being washed out of the well. The unique hydrological characteristics of the spring have likely selected for both traits. The mechanisms by which controlled nucleation of extracellular calcite is achieved remain to be explored.  相似文献

The present study compared bereavement responses of 325 monozygotic (MZ) and 176 dizygotic (DZ) adolescent and adult twins following the loss of their co-twins. A subset of twins completed the Grief Experience Inventory using a retrospective time frame, while a second subset completed it using a current time frame. It was hypothesized that MZ twins (in both retrospective and current groups) would report higher levels of grief-related behavior than DZ twins, consistent with Hamilton's (1964) concept of inclusive fitness. Discriminant function and profile analyses yielded supportive findings, but only for the retrospective MZ and DZ twin comparisons. Females in both groups expressed higher levels of bereavement-related behavior than males. Findings are discussed with reference to theoretical aspects of grief and mourning.  相似文献

Skewed sex ratios are common among several species of Poeciliopsis, a viviparous fish from northwestern Mexico. Since previous, unrelated studies from this laboratory (Angus and Schultz, 1983) suggested that deviation from a 1:1 sex ratio might be influenced by temperature, two inbred strains of P. lucida were tested for temperature-dependent sex determination by comparing sex ratios of offspring from pregnant females held at different water temperatures. Different sex ratios were produced by the two strains at the same temperature: one strain produced almost all-male offspring at 30°C and female-biased sex ratios at 24°C, while the other strain produced a 1:1 sex ratio at both temperatures. At intermediate temperatures, the labile strain produced sequentially fewer males with decreasing temperatures. The other strain produced a consistent sex ratio regardless of temperature. Poeciliopsis lucida apparently has a genetic polymorphism for temperature-influenced sex determination. An hypothesis is offered for the evolutionary origin of environmental sex determination.  相似文献

A small population of cells representing 1% or less of those in the root-tip meristem was identified as the precursor of vascular parenchyma and certain root-cap cells in carbohydrate starved cultured pea roots. Autoradiography and cytophotometric measurements of nuclei labeled with [3H]-thymidine showed that in the absence of carbohydrate the precursor cells replicate their DNA discontinuously accumulating temporarily in late S phase prior to differentiating from the G2 phase. Besides discontinuity of DNA synthesis, the nuclei of precursor cells undergo a change in morphology. The nuclei are shaped round when replicating DNA but later on, while differentiating, they become oblong. This transformation occurs within 72 hr after the starved roots are fed sucrose. Autoradiograms of serial cross-sections of pulse-labeled roots indicate that the cells in late S phase differentiate forming a ring around the stelar cylinder and a ring around the periphery of the root. These observations suggest that during the last half of the final S phase the precursor cells modify their chromosomal DNA and that this modification is associated with the initial steps of differentiation.  相似文献
