The change in optimal quantum efficiency (F v/F m) of the Arctic species Laminaria saccharina and Palmaria palmata was investigated in a long-term experiment in situ under different radiation levels during the summer of 1997 in the Kongsfjord (Ny-Ålesund, Spitsbergen, Norway, 78°55.5′N, 11°56.0′E). Whole plants were incubated in an open box system made of UV-transparent Perspex and exposed to solar radiation (λ>295?nm), solar radiation excluding UVB (λ?>?320?nm) and solar radiation excluding UVA?+ UVB (λ?>?400?nm). Increasing radiation levels were simulated by transplantation of the pre-adapted algae from their growth depth at 2?m to a water depth of 1?m. Sensitivity to artificially increased UV radiation was determined by exposure of algae from the three treatments to 6?h of strong UV radiation. P. palmata was relatively insensitive to increasing UV radiation and recovered very fast and almost completely in 2?h. Even plants pre-cultured in ambient radiation levels excluding UVA?+?UVB or UVB only showed no photoinhibition after exposure to extra UV radiation in the laboratory. L. saccharina was, in comparison to P. palmata, more sensitive and showed photoinhibition under solar radiation and solar minus UVB radiation after transplantation from 2 to 1?m water depth. However, after 3?weeks at 1?m depth, F v/F m of L. saccharina was equal in all treatments and restored to the original values at the start of the experiment. Sensitivity to extra UV radiation in the laboratory increased in time, although recovery was also fast and occurred within 20?h.  相似文献

The Westerschelde estuary is a very polluted and turbid estuary, but the last decade the waterquality improved. Dredging activity also increased in 1997 to allow bigger ships to enter the port of Antwerpen. This could potentially decrease the light conditions for the phytoplankton. Because of all these recent changes in the estuary we studied primary productivity in 2001 and compared it to values in 1991. The results show that due to a decrease in discharge in particulate and dissolved organic carbon the oxygen concentrations in general have increased in the upstream region, although in spring and summer low oxygen concentrations (10–30% saturation) can still be found. Phosphate and ammonia concentrations have decreased and the zone of nitrification which was very large in 1991 has become very small and is now located in the uppermost upstream region of the estuary. Si-concentrations have remained the same. All nutrient concentrations are still high enough not to limit phytoplankton growth. Turbidity remained unaltered as a result of the dredging works, and as a result phytoplankton biomass in most of the estuary did not show a decrease, although there were signs that in the upstream region phytoplankton biomass decreased, possible caused by increased grazing pressure. The relationship between phytoplankton biomass and primary productivity did not change, and from the data it can be concluded that the dredging activity will not influence the gross and net primary productivity of the phytoplankton.  相似文献

Hemidesmosomes (HDs) are multiprotein adhesion complexes that promote attachment of epithelial cells to the basement membrane. The binding of alpha6beta4 to plectin plays a central role in their assembly. We have defined three regions on beta4 that together harbor all the serine and threonine phosphorylation sites and show that three serines (S1356, S1360, and S1364), previously implicated in HD regulation, prevent the interaction of beta4 with the plectin actin-binding domain when phosphorylated. We have also established that epidermal growth factor receptor activation, which is known to function upstream of HD disassembly, results in the phosphorylation of only one or more of these three residues and the partial disassembly of HDs in keratinocytes. Additionally, we show that S1360 and S1364 of beta4 are the only residues phosphorylated by PKC and PKA in cells, respectively. Taken together, our studies indicate that multiple kinases act in concert to breakdown the structural integrity of HDs in keratinocytes, which is primarily achieved through the phosphorylation of S1356, S1360, and S1364 on the beta4 subunit.  相似文献

Lamellipodial protrusion and directional migration of carcinoma cells towards chemoattractants, such as epidermal growth factor (EGF), depend upon the spatial and temporal regulation of actin cytoskeleton by actin-binding proteins (ABPs). It is generally hypothesized that the activity of many ABPs are temporally and spatially regulated by PIP2; however, this is mainly based on in vitro–binding and structural studies, and generally in vivo evidence is lacking. Here, we provide the first in vivo data that directly visualize the spatial and temporal regulation of cofilin by PIP2 in living cells. We show that EGF induces a rapid loss of PIP2 through PLC activity, resulting in a release and activation of a membrane-bound pool of cofilin. Upon release, we find that cofilin binds to and severs F-actin, which is coincident with actin polymerization and lamellipod formation. Moreover, our data provide evidence for how PLC is involved in the formation of protrusions in breast carcinoma cells during chemotaxis and metastasis towards EGF.  相似文献

The photosynthesis of plankton sampled from the eutrophic Lake Loosdrecht was studied in Laboratory Scale Enclosures (LSEs) with regard to the rate of mixing. First, two LSEs were operated at different mixing rates. No significant differences in photosynthetic performance were found, with the exception of a depressed photosynthesis in the afternoon in the LSE which had a low mixing rate. Secondly, when mixing was stopped, the phytoplankton which stayed in the dark due to the steep light gradient in the LSE responded by changing its maximal photosynthetic capacity. The results show that the filamentous cyanobacteria in the lake can respond rapidly to changes in the depth of the mixed layer by altering their photosynthetic performance.This is publication 567 of the Centre for Estuarine and Coastal Ecology.  相似文献

We report a technique to evaluate the same tumor microenvironment over multiple intravital imaging sessions in living mice. We optically marked individual tumor cells expressing photoswitchable proteins in an orthotopic mammary carcinoma and followed them for extended periods through a mammary imaging window. We found that two distinct microenvironments in the same orthotopic mammary tumor affected differently the invasion and intravasation of tumor cells.  相似文献

Stable isotopes are increasingly being used to infer past and present trophic interactions in light of environmental changes. The Lake Victoria haplochromine cichlids have experienced severe environmental changes in the past decades that, amongst others, resulted in a dietary shift towards larger prey. We investigated how the changed environment and diet of the haplochromines influenced stable isotope values of formalin-then-ethanol-preserved cichlid specimens, and then investigated how these values differed among species before (1977–1982) and after substantial environmental changes (2005–2007). We found a small preservation effect on both δ13C and δ15N values, and significant differences in isotope values among haplochromine species collected before the environmental changes. In contrast, there was a remarkable similarity in δ13C and δ15N values among species collected from the contemporary ecosystem and two out three species showed significantly different stable isotope values compared to species of the historic ecosystem. In addition, we found a putative isotopic gradient effect along our 5-km-long research transect indicating that the studied demersal species are more stenotopic than previously thought. The environmental changes have resulted in dietary change and overlap of the haplochromines which provides insight into the trophic plasticity of these species, which are often considered trophic specialists.  相似文献

Integrins control motile strategy through a Rho-cofilin pathway   总被引:1,自引:0,他引:1  
During wound healing, angiogenesis, and tumor invasion, cells often change their expression profiles of fibronectin-binding integrins. Here, we show that beta1 integrins promote random migration, whereas beta3 integrins promote persistent migration in the same epithelial cell background. Adhesion to fibronectin by alpha(v)beta3 supports extensive actin cytoskeletal reorganization through the actin-severing protein cofilin, resulting in a single broad lamellipod with static cell-matrix adhesions at the leading edge. Adhesion by alpha5beta1 instead leads to the phosphorylation/inactivation of cofilin, and these cells fail to polarize their cytoskeleton but extend thin protrusions containing highly dynamic cell-matrix adhesions in multiple directions. The activity of the small GTPase RhoA is particularly high in cells adhering by alpha5beta1, and inhibition of Rho signaling causes a switch from a beta1- to a beta3-associated mode of migration, whereas increased Rho activity has the opposite effect. Thus, alterations in integrin expression profiles allow cells to modulate several critical aspects of the motile machinery through Rho GTPases.  相似文献
