We determined the DNA sequences surrounding the junctions of IS91 in two insertion derivatives: pSU234 (pACYC184::IS91) and pSU240 (pBR322::IS91). The termini of IS91 consist of two imperfect inverted repeats eight base pairs long. Their sequence is 5'-TCGAGTAGG. . . CCTATCGA-3'. Insertion of IS91 did not generate direct repetitions in the target DNAs.  相似文献

Tubulin-folding cofactor E (TBCE) is an alpha-tubulin-binding protein involved in the formation of the tubulin dimer and in microtubule dynamics, through the regulation of tubulin heterodimer dissociation. TBCE has also been implicated in two important related human disorders, the Kenny-Caffey and Sanjad-Sakati syndromes. The expression of TBCE as a recombinant protein in bacteria results in the formation of insoluble inclusion bodies in the absence of denaturing agents. Although the active protein can be obtained from mammalian tissues, biochemical studies of TBCE present a special challenge. To express and purify native TBCE, a recombinant baculovirus expression system was used. Native wild-type TBCE purified from Sf9 extracts was sequentially purified chromatographically through cation exchange, hydrophobic interaction, and high-resolution gel-filtration columns. Mass spectrometric analysis identified 30% of the sequence of human TBCE. A stoichiometric excess of purified TBCE dissociated tubulin heterodimers. This reaction produced a highly unstable TBCE-alpha-tubulin complex, which formed aggregates. To distinguish between the aggregation of tubulin dimers induced by TBCE and tubulin dissociation, TBCE and tubulin were incubated with tubulin-folding cofactor A (TBCA). This cofactor captures the beta-tubulin released from the heterodimer with a stoichiometry of 1:1, as previously demonstrated. The beta-tubulin polypeptide was recovered as TBCA-beta-tubulin complexes, as demonstrated by non-denaturing gel electrophoresis and specific antibodies directed against beta-tubulin and TBCA.  相似文献

A quantitative GC method for conjugated linoleic acid (CLA) isomers of physiological significance (cis-9, trans-11 CLA and trans-10, cis-12 CLA) as non-esterified fatty acids (NEFA) or triacylglycerols (TAG) was developed. Furthermore, the effect of the internal standard addition point (sample or fat extract) was studied. Response linearity, recovery and precision assays, detection and quantification limits were determined. Linearity was demonstrated over a range from 0.1 to 10 microg/mL. When CLA isomers were present as NEFA, the recovery significantly decreased (P< or =0.05) from 76% to 27.1% (cis-9, trans-11 CLA) and 28.5% (trans-10, cis-12 CLA) when the standards were added to the fat extract or to the initial tissue, respectively. As an application, liver samples from hamsters fed a diet supplemented with both CLA isomers were analyzed. The CLA isomers in liver samples were detected with reasonable reproducibility.  相似文献

Methods of evaluating population trends have recently received particular attention because of perceived declines in several species during the twentieth century. We have studied demographic traits of the peregrine falcon (Falco peregrinus) population in Bizkaia (Northern Spain) for 11 years. This species suffered a severe decline in the 1950s and 1960s and started to recover in the 1980s and 1990s, although the recovery trends differed between areas. In our study area the peregrine falcon density is one of the highest found in Spain. The frequency of juvenile breeders was 2.0%, the mean age at first breeding being 3.7 calendar years for males and 4.0 cy for females. The territorial fidelity was at least 3.4 years for males and 3.7 years for females. Females dispersed on average 80.5 km and males 51.8 km during their pre-breeding movements, whilst distance between birthplace and breeding territories was on average 108.5 km for females and 64.5 km for males. We studied the relationships between adult turnover, recruitment age, territorial fidelity and dispersal in a healthy population in order to establish population dynamics. A combination of these parameters, not just age at first breeding, could be used as a potential early warning signal indicating future changes, prompting their consequences to be evaluated. This approach could lead to the reclassification of the large-scale health of a population. Its utilization would allow resources to be directed into helping “ailing” subpopulations, detecting causes of decline, and developing adequate recovery strategies.  相似文献

Chromosome territories constitute the most conspicuous feature of nuclear architecture, and they exhibit non-random distribution patterns in the interphase nucleus. We observed that in cell nuclei from humans with Down Syndrome two chromosomes 21 frequently localize proximal to one another and distant from the third chromosome. To systematically investigate whether the proximally positioned chromosomes were always the same in all cells, we developed an approach consisting of sequential FISH and CISH combined with laser-microdissection of chromosomes from the interphase nucleus and followed by subsequent chromosome identification by microsatellite allele genotyping. This approach identified proximally positioned chromosomes from cultured cells, and the analysis showed that the identity of the chromosomes proximally positioned varies. However, the data suggest that there may be a tendency of the same chromosomes to be positioned close to each other in the interphase nucleus of trisomic cells. The protocol described here represents a powerful new method for genome analysis.  相似文献

Giant tortoises were once a megafaunal element widespread in tropical and subtropical ecosystems. The role of giant tortoises as herbivores and seed dispersers, however, is poorly known. We evaluated tortoise impacts on Opuntia cactus (Cactaceae) in the Galápagos Islands, one of the last areas where giant tortoises remain extant, where the cactus is a keystone resource for many animals. We contrasted cactus populations immediately inside and outside natural habitats where tortoises had been held captive for several decades. Through browsing primarily and trampling secondarily tortoises strongly reduced densities of small (0.5–1.5 m high) cacti, especially near adult cacti, and thereby reduced densities of cacti in larger size classes. Tortoises also caused a shift from vegetative to sexual modes of reproduction in cacti. We conclude that giant tortoises promote a sparse and scattered distribution in Opuntia cactus and its associated biota in the Galápagos Islands.  相似文献
