The prospects for persistence of bees living in fragmented landscapes is a topic of considerable interest due to bees’ importance
as pollinators of agricultural crops and wild plants, coupled with the ubiquity of native habitat loss and evidence that bees
may be declining worldwide. Population persistence in fragmented areas depends on dispersal potential and maintenance of gene
flow among fragments of habitat. Here we used population genetic techniques to characterize, for two equally abundant orchid
bee species that differ in their physiology and ecology, levels of genetic differentiation among fragments of tropical forest
in southeastern Costa Rica in a ~200 km2 landscape. We measured population differentiation with ϕPT (an analogue to the traditional summary statistic Fst), as well as two measures that may more accurately reflect the level
of differentiation when highly variable loci are used: G’st and Dest. We also calculated pairwise genetic distances among individuals and conducted Mantel tests to test the correlation of genetic
and geographic distance, for each species. We found strong differences in genetic structure between the species. Contrary
to our expectations, each measure of genetic structure revealed that the larger-bodied species, Eulaema bombiformis, had higher levels of differentiation than the smaller species, Euglossa championi. Furthermore, for Eulaema bombiformis there was a significant positive correlation of genetic and geographic distance while for Euglossa championi there was no significant positive correlation. Our results demonstrate that bee species can have strikingly different levels
of gene flow in fragmented habitats, and that body size may not always act as a useful proxy for dispersal, even in closely

The occurrence of boar taint and the European Commission recommendation to discontinue the surgical castration of pigs by the year 2018 creates an urgent need for new analytical methods that are simple, affordable, and suitable for field testing. We describe the generation and engineering of a skatole-specific antibody derived from a synthetic antibody library and the development of ELISA for its detection. The immunoassay is capable of detecting skatole with IC50 of 222 μg L−1, which is within the analytical threshold level suggested for skatole, and with low cross-reactivity interference from other indolic compounds. 相似文献

Fungal laccases are attracting enzymes for sustainable valorization of biorefinery lignins. To improve the lignin oxidation capacity of two previously characterized laccase isoenzymes from the white-rot fungus Obba rivulosa, we mutated their substrate-binding site at T1. As a result, the pH optimum of the recombinantly produced laccase variant rOrLcc2-D206N shifted by three units towards neutral pH. O. rivulosa laccase variants with redox mediators oxidized both the dimeric lignin model compound and biorefinery poplar lignin. Significant structural changes, such as selective benzylic α-oxidation, were detected by nuclear magnetic resonance analysis, although no polymerization of lignin was observed by gel permeation chromatography. This suggests that especially rOrLcc2-D206N is a promising candidate for lignin-related applications. 相似文献

Allozyme analyses have suggested that Neotropical orchid bee (Euglossini) pollinators are vulnerable because of putative high frequencies of diploid males, a result of loss of sex allele diversity in small hymenopteran populations with single locus complementary sex determination. Our analysis of 1010 males from 27 species of euglossine bees sampled across the Neotropics at 2–11 polymorphic microsatellite loci revealed only five diploid males at an overall frequency of 0.005 (95% CIs 0.002–0.010); errors through genetic nondetection of diploid males were likely small. In contrast to allozyme‐based studies, we detected very weak or insignificant population genetic structure, even for a pair of populations >500 km apart, possibly accounting for low diploid male frequencies. Technical flaws in previous allozyme‐based analyses have probably led to considerable overestimation of diploid male production in orchid bees. Other factors may have a more immediate impact on population persistence than the genetic load imposed by diploid males on these important Neotropical pollinators. 相似文献

Human-caused genetic changes in two Atlanticsalmon (Salmo salar L.) stocks, from therivers Iijoki and Oulujoki in Finland, wereassessed by comparing the genetic parameters ofthese stocks before and after the hatcherybreeding of several successive generations,corresponding to 40 and 33 years since the wildstate. The changes were also compared withthose observed in a large wild salmon stock inthe River Teno during 56 years. In all, thevariation at seven microsatellite DNA loci wasexamined in 11 Atlantic salmon samplesoriginating from these three rivers. Theeffective population size, Ne, duringbreeding of the Iijoki broodstock and for theTeno salmon was also estimated by the temporalmethod based on allele frequency changes. Forthe Iijoki broodstock, the changes could betracked generation by generation from thefounding of the stock. Statisticallysignificant changes in allele frequencies werecommon in the hatchery stocks (F = 0.029, forIijoki), but not in the wild Teno stock, whichwas temporally very stable (F = 0.007). Allelicrichness decreased statistically significantly(24.8%) in the Oulujoki broodstock, from 62.1to 46.7 alleles at nine loci. On average, therewere 9.7 fewer alleles (15.7%) in thecontemporary broodstocks than in thecorresponding historical stocks. The meanheterozygosity was 6.6% lower in thecontemporary Oulujoki broodstock, but remainedunchanged in the Iijoki broodstock. Theestimated Ne for the Iijoki broodstock wasunder 80 for 4.5 generations from 1962 to 1995and for the wild Teno salmon over 900 for 56years from 1939 to 1995. 相似文献

Previously, various persistent pesticides were used extensively in the production of seedlings at Finnish forest nurseries. The extent and magnitude of the risks arising from the consequent environmental contamination are largely unknown. Therefore, we selected two representative nurseries for which we conducted tiered health risk assessments (HRA) using risk-based benchmarks and two calculation tools (SSL and Risc-Human software). Ecological risk assessments (ERA) involved comparisons of environmental concentrations with ecotoxicological benchmarks. Site investigations revealed that the concentration of several pesticides exceeded the Finnish soil quality guidelines in some places. The compost pile for organic residues and the pond receiving runoffs contained traces of pesticides and the maximum concentration of atrazine and terbuthylazine in groundwater exceeded the corresponding guideline for household water. Hexachlorobenzene proved to pose the highest health risks, the maximum hazard quotient being around 10 (carcinogenity-based) in the residential land use scenario. Owing to the conservative assumptions, health risks are expected to remain insignificant, however. Risks to the local terrestrial ecosystem would also remain low, while only further studies will reveal the actual risks to the adjoining aquatic ecosystem. Both calculation tools showed shortcomings that generate uncertainty in the HRA, whereas the ERA was hampered particularly by the lack of benchmarks. 相似文献
