The present report is a part of our continuing efforts to explore the utility of the rat epidermal keratinocyte organotypic culture (ROC) as an alternative model to human skin in transdermal drug delivery and skin irritation studies of new chemical entities and formulations. The aim of the present study was to compare the stratum corneum lipid content of ROC with the corresponding material from human skin. The lipid composition was determined by thin-layer chromatography (TLC) and mass-spectrometry, and the thermal phase transitions of stratum corneum were studied by differential scanning calorimetry (DSC). All major lipid classes of the stratum corneum were present in ROC in a similar ratio as found in human stratum corneum. Compared to human skin, the level of non-hydroxyacid-sphingosine ceramide (NS) was increased in ROC, while α-hydroxyacid-phytosphingosine ceramide (AP) and non-hydroxyacid-phytosphingosine ceramides (NP) were absent. Also some alterations in fatty acid profiles of ROC ceramides were noted, e.g., esterified ω-hydroxyacid-sphingosine contained increased levels of oleic acid instead of linoleic acid. The fraction of lipids covalently bound to corneocyte proteins was distinctly lower in ROC compared to human skin, in agreement with the results from DSC. ROC underwent a lipid lamellar order to disorder transition (T2) at a slightly lower temperature (68 °C) than human skin (74 °C). These differences in stratum corneum lipid composition and the thermal phase transitions may explain the minor differences previously observed in drug permeation between ROC and human skin. 相似文献

Sex-biased natal dispersal in long-lived species may resultin interactions between parents and mature young of the philopatricsex. To investigate the evolutionary basis of natal philopatryin a noncooperative species, the common goldeneye Bucephalaclangula, we studied possible costs and benefits of simultaneousbreeding of females and philopatric daughters. We did not find
any fitness consequences of a daughter's breeding on their mother'sbreeding in terms of nest-site selection, body weight, clutchsize, hatching date, or hatching success. Our results, therefore,did not support the assumption of the local resource competitionhypothesis, that the natally philopatric sex should be morecostly to a breeding parent. As possible benefits for daughters
returning to their natal area, we tested inheritance of nestsites from mothers and explored whether daughters utilize thepresence of their mother by parasitically sneaking into hermother's nest. Daughters' nest-site selection was not associatedwith the presence of their mothers. A comparison between daughtersand control females revealed that daughters chose their nestsite closer to their natal nest than expected by nest-siteavailability alone. Daughters could not expect to inherit anest site from their mother, and we did not find other indicationsof cooperation between relatives either. The mother's clutchsize did not increase in the year breeding with the daughter,

A method for the preparation of a fluorescent phosphatidylinositol analogue, 1-acl,-2-prinaroyl-sn-glycero-3-phospho-sn-1-myo-inositol has been developed. This method makes use of yeast phosphatidylinositol as the starting material and includes the following steps: (1) acetylation of the free hydroxyl groups of the inositol moeity; (2) removal of the fatty acid from the sn-2-position of the glycerol moiety by phospholipase A2; (3) reacylation with parimaroyl anhydride; (4) removal of the protecting acetyl groups by alkaline solvolysis. The identity of the product was established by thin-layer chromatography (TLC), UV absorption spectroscopy, enzymatic degradation and by a transfer assay using the phosphatidylinositol transfer protein from bovine brain.Some properties of the fluorescent phosphatidylinositol analogue are reported. 相似文献

Life-history theory assumes a trade-off between current reproductive effort and future reproductive success. There are a large number of studies demonstrating reproductive trade-offs in different animal taxa, particularly in birds. Most bird studies have focused on the costs of chick rearing in altricial species. These costs have been assumed to be low in precocial species, but this aspect has been little studied. We used long-term individual reproductive data from the common goldeneye Bucephala clangula , an iteroparous precocial duck with uniparental female care, to examine whether brood rearing carries costs that affect future reproductive performance. All females were experienced breeders, and possible differences in female quality were ruled out. We compared within-individual (between-year) changes in clutch size, hatching date and body mass between females that had reared a brood in the previous year and females that had not. It turned out that brood rearing involved a cost in terms of clutch size and hatching date the next year, but not in terms of body mass: females that had reared a brood in the previous year laid relatively smaller clutches and laid relatively later than females that had not reared a brood. Summary The uptake, incorporation and distribution of tritiated thymidine and uridine into visceral organs of immature female and male rats was studied. Radioautographs of various organs were prepared 5, 15 and 30 min after intraperitoneal administration. The distribution of the radioactivity was compared with sections obtained 30 min after intravenous injection of the same amount of nucleotides.The intraperitoneal injection was performed by directing the needle toward the right side of the abdominal cavity. Visceral organs from the injected side contained consistently more silver grains than those from the opposite side. Sections of the abdominal organs showed a decreasing concentration gradient of reduced silver grains extending from the serosal part of the organ to the deeper layers. In addition, initial labeling was observed in sectors of organs which protruded into the peritoneal cavity. In contrast, following intravenous administration of nucleotides a concentration gradient was not observed in any of the visceral organs.The results of the present study suggest that nucleotides administered intraperitoneally may penetrate the abdominal organs directly via the serosal surface without prior entry into the general circulation. 相似文献

The impact of nutrient enrichment on the phytoplankton community structure, and particularly cyanobacteria, was studied in a 3-week mesocosm experiment conducted in August 2001 in the Archipelago Sea, a part of the northern Baltic Sea. The factorial design experiment included daily additions of nitrogen (N) and phosphorus (P) at two mass ratios, 1N:1P and 7N:1P, respectively, additions of iron (Fe) and a synthetic chelator, ethylenediaminetetraacetic acid (EDTA). The floating enclosures (400 l) were sampled for analyses of phytoplankton biomass and community structure, phytoplankton primary production, chlorophyll a, nutrients, and hepatotoxins. Chlorophyll a concentration, phytoplankton biomass and primary production increased most in the 7N:1P treatment. The increase was mainly due to an abundant growth of chlorophytes (Dictyosphaerium subsolitarium, Kirchneriella spp., Monoraphidium contortum, and Oocystis spp.), pennate diatoms (especially Nitzschia spp.), dinophytes and the chroococcalean cyanobacterium Synechococcus sp. The nutrient enrichments had no effect on the total biomass of N2-fixing cyanobacteria. Nevertheless, the biomass of Anabaena spp. was highest in the enrichments with a low N/P ratio. Chlorophyll a concentration and total phytoplankton biomass were not affected by Fe or EDTA, but Fe alone had a positive effect on the chlorophyte Kirchneriella sp. The N2-fixing cyanobacteria Aphanizomenon sp. responded positively to Fe alone and to both Fe and EDTA added together. The hepatotoxin concentration increased during the experiment, but no clear responses to nutrient enrichments were found. Our study showed species-specific responses to nutrient enrichments among the N2-fixing cyanobacteria. Recent, primarily structural observations indicate that related viruses, harboring no sequence similarity, infect hosts of different domains of life. One such clade of viruses, defined by common capsid architecture and coat protein fold, is the so-called PRD1-adenovirus lineage. Here we report the structure of the marine lipid-containing bacteriophage PM2 determined by crystallographic analyses of the entire approximately 45 MDa virion and of the outer coat proteins P1 and P2, revealing PM2 to be a primeval member of the PRD1-adenovirus lineage with an icosahedral shell and canonical double beta barrel major coat protein. The view of the lipid bilayer, richly decorated with membrane proteins, constitutes a rare visualization of an in vivo membrane. The viral membrane proteins P3 and P6 are organized into a lattice, suggesting a possible assembly pathway to produce the mature virus. 相似文献

Replicated, factorial mesocosm experiments were conducted across Europe to study the effects of nutrient enrichment and fish
density on macrophytes and on periphyton chlorophyll a (chl-a) with regard to latitude. Periphyton chl-a densities and plant decline were significantly related to nutrient loading in all countries. Fish effects were significant
in a few sites only, mostly because of their contribution to the nutrient pool. A saturation-response type curve in periphyton
chl-a with nutrients was found, and northern lakes achieved higher densities than southern lakes. Nutrient concentration and phytoplankton
chl-a necessary for a 50% plant reduction followed a latitudinal gradient. Total phosphorus values for 50% plant disappearance
were similar from Sweden (0.27 mg L−1) to northern Spain (0.35 mg L−1), but with a sharp increase in southern Spain (0.9 mg L−1). Planktonic chl-a values for 50% plant reduction increased monotonically from Sweden (30 μg L−1) to València (150 μg L−1). Longer plant growing-season, higher light intensities and temperature, and strong water-level fluctuations characteristic
of southern latitudes can lead to greater persistence of macrophyte biomass at higher turbidities and nutrient concentration
than in northern lakes. Results support the evidence that latitudinal differences in the functioning of shallow lakes should

Tawny owl reproduction and offspring sex ratios have been considered to depend on the abundance of small voles. We studied
reproductive performance (laying date, clutch and brood size) during 1995–2003 and offspring sex ratios from 1999 to 2003
in relation to the abundance of small voles and food delivered to the nest in a tawny owl population in southern Finland.
Abundance of small voles (field and bank voles) was based on trappings in the field, and estimates of food delivery was based
on diet analysis of food remains in the nest boxes. In this population, reproductive output was not related to the abundance
of small voles. Analysis of food delivered to the nest showed that the prey weight per offspring varied more than twofold
between years and revealed that this difference was mainly related to the proportion of water voles in the diet. Only the
number of water voles correlated with laying dates. Offspring sex ratios were weakly male biased (55%) but did not differ
from parity. Sex ratios were not related to the abundance of small voles, and we found no evidence that parents delivered
more food to nests with proportionally more offspring of the larger (female) sex. Our results underline the notion that populations
may differ in their sex allocation pattern, and suggest such differences may be due to diet. 相似文献
