Summary Three polymorphic loci APOC2, CKMM and p134C were used to haplotype 15 Finnish dystrophia myotonica (DM) families representing about one third of all DM patients in this isolated population. Compound APOC2 and CKMM haplotypes reveal linkage disequilibrium: 90% of DM chromosomes co-occur with the haplotypes that occur in 31% of normal chromosomes only. The same disequilibrium is present when only polymorphisms occurring at the APOC2 locus are used. Surprisingly, no statistically significant linkage disequilibrium was discovered at the CKMM locus alone. Of the meiotic events, 84% were informative when both APO2 and CKMM loci were used. When studied selectively, 60% of meiotic events were informative at the APOC2 locus, whereas CKMM alone resulted in 65% meiotic informativeness. The distal marker p134C was found to have an unfortunately low information content in our population.  相似文献

Summary The case of a healthy 29-year-old woman is reported who had a history of three early spontaneous abortions. Chromosomal analysis of the mother of the patient showed a balanced karotype of 46,XX,t(6;11)(q24; q21), whereas the chromosomes of her father were normal. The karotype of this patient is thus a combination of a familial translocation 6;11 and a de novo translocation 4;11, which is very rare.  相似文献

Bark beetle diversity at different spatial scales   总被引:3,自引:1,他引:2  
To determine how the scale of observation affects ecological patterns we studied bark beetle (Coleoptera, Scolytidae) diversity in southern Finland. A block covering 160 × 160 m of a forest was delimited in four stands of different site types. Each block was divided into 256 squares (10 × 10 m) in which the occurrence of bark beetle species was recorded. In addition, environmental variables describing site type, trees, and breeding material appropriate for bark beetles were measured. The species presence/absence data were combined at different scales of resolution (10 × 10 m. 20 × 20 m, 40 × 40 m, 80 × 80 m, 160 × 160 m). At the finest scale a recently thinned pine stand showed relatively high diversity compared to other study stands due to a few evenly distributed and abundant species. However, the species diversity increased faster toward larger scales in mature spruce stands with several sporadically distributed species. According to logistic regression analyses, breeding material and site characteristics explained the occurrence of most beetle species. However, these variables did not explain the occurrence of the six most frequent species, probably because the factors regulating their distribution and occurrence operate at larger scales.  相似文献

Multi-sea-winter Atlantic salmon (75–115 cm fork length L F, 2–4-winter fish) were radio-tagged in the Tanafjord (700 N), Norway, in 1992–1993, and 130 fish entered the large subarctic River Tana (Teno). They entered the fresh water at any time of the tidal cycle but more so during the high and ebbing tides. No diel rhythm was detected in river entry under polar day conditions. There were no differences in the change of flow between days when salmon moved and when they did not, but during active migration increasing discharge was associated with increased swimming activity of salmon, especially later in the summer. Increasing air temperature was also associated with enhanced migration activity. Low river flow was associated with increasing delay in salmon passing the first riffle area of the river, 35 km from the sea.  相似文献

Hepatic tyrosine aminotransferase (l-tyrosine:2-oxoglutarate aminotransferase, EC 2.6.1.5) is known to be induced by α-methyl-p-tyrosine, a well-known catecholamine depletor, in both intact and adrenalectomized rats. The authors have studied this subject further and their results show that α-methyl-p-tyrosine does not influence the activity of tyrosine aminotransferase in the isolated, perfused liver and that hypophysectomy totally abolishes the induction of the enzyme by this agent. The involvement of hypophyseal hormones is discussed.  相似文献

Nitric oxide (NO) is involved together with reactive oxygen species (ROS) in the activation of various stress responses in plants. We have used ozone (O3) as a tool to elicit ROS-activated stress responses, and to activate cell death in plant leaves. Here, we have investigated the roles and interactions of ROS and NO in the induction and regulation of O3-induced cell death. Treatment with O3 induced a rapid accumulation of NO, which started from guard cells, spread to adjacent epidermal cells and eventually moved to mesophyll cells. During the later time points, NO production coincided with the formation of hypersensitive response (HR)-like lesions. The NO donor sodium nitroprusside (SNP) and O3 individually induced a large set of defence-related genes; however, in a combined treatment SNP attenuated the O3 induction of salicylic acid (SA) biosynthesis and other defence-related genes. Consistent with this, SNP treatment also decreased O3-induced SA accumulation. The O3-sensitive mutant rcd1 was found to be an NO overproducer; in contrast, Atnoa1/rif1 ( Arabidopsis nitric oxide associated 1/resistant to inhibition by FSM1 ), a mutant with decreased production of NO, was also O3 sensitive. This, together with experiments combining O3 and the NO donor SNP suggested that NO can modify signalling, hormone biosynthesis and gene expression in plants during O3 exposure, and that a functional NO production is needed for a proper O3 response. In summary, NO is an important signalling molecule in the response to O3.  相似文献

Batch experiments were conducted to investigate the thermophilic biohydrogen production using an enrichment culture from a Turkish hot spring. Following the enrichment, the culture was heat treated at 100 °C for 10 min to select for spore-forming bacteria. H2 production was accompanied by production of acetate, butyrate, lactate and ethanol. H2 production was associated by acetate–butyrate type fermentation while accumulation of lactate and ethanol negatively affected the H2 yield. H2 production was highest in the temperature range from 49.6 to 54.8 °C and optimum values for initial pH and concentrations of iron, yeast extract and glucose were 6.5, 40 mg/l, 4–13.5 g/l, respectively. PCR–DGGE profiling showed that the heat treated culture consisted of species closely affiliated to genus Thermoanaerobacterium.  相似文献

Diabetic nephropathy (DN) is the primary cause of morbidity and mortality in patients with type 1 diabetes mellitus (T1DM) and affects about 30% of these patients. We have previously localized a DN locus on chromosome 3q with suggestive linkage in Finnish individuals. Linkage to this region has also been reported earlier by several other groups. To fine map this locus, we conducted a multistage case-control association study in T1DM patients, comprising 1822 cases with nephropathy and 1874 T1DM patients free of nephropathy, from Finland, Iceland, and the British Isles. At the screening stage, we genotyped 3072 tag SNPs, spanning a 28 Mb region, in 234 patients and 215 controls from Finland. SNPs that met the significance threshold of p < 0.01 at this stage were followed up by a series of sample sets. A genetic variant, rs1866813, in the noncoding region at 3q22 was associated with increased risk of DN (overall p = 7.07 × 10−6, combined odds ratio [OR] of the allele = 1.33). The estimated genotypic ORs of this variant in all Finnish samples suggested a codominant effect, resulting in significant association, with a p value of 4.7 × 10−5 (OR = 1.38; 95% confidence interval = 1.18–1.62). Additionally, an 11 kb segment flanked by rs62408925 and rs1866813, two strongly correlated variants (r2 = 0.95), contains three elements highly conserved across multiple species. Independent replication will clarify the role of the associated variants at 3q22 in influencing the risk of DN.  相似文献

A thermophilic, rod-shaped, motile, Gram-positive, spore-forming bacterium strain 70BT was isolated from a geothermally active underground mine in Japan. The temperature and pH range for growth was 50–81°C (optimum 71°C) and 6.2–9.8 (optimum pH 7–7.5), respectively. Growth occurred in the presence 0–2% NaCl (optimum 1% NaCl). Strain 70BT could utilize glucose, fructose, mannose, mannitol, pyruvate, cellobiose and tryptone as substrates. Thiosulfate was used as electron acceptor. Major whole-cell fatty acids were iso-C15:0, C16:0 DMA (dimethyl acetal), C16:0 and anteiso-C15:0. The G+C mol% of the DNA was 44.2%. Phylogenetic analysis based on the 16S rRNA gene sequence revealed that the closest relatives of strain 70BT were Thermosediminibacter oceani DSM 16646T (94% similarity) and Thermosediminibacter litoriperuensis DSM 16647 (93% similarity). The phenotypic, chemotaxonomic and phylogenetic properties suggest that strain 70BT represents a novel species in a new genus, for which the name Thermovorax subterraneus gen. nov., sp. nov. is proposed. The type strain of Thermovorax subterraneus is 70BT (=DSM 21563 = JCM 15541).  相似文献
