SYNOPSIS. Chloroplast development is inhibited in Euglena gracilis strain Z, when greened in a medium containing glucose. This inhibition is reflected not only in the pattern of chlorophyll accumulation but also in the chloroplast ultrastructure and activities of the 2 light reactions of photosynthesis. Chloroplasts of cells greening in the presence of glucose are delayed in déveloping certain structures. Photosystem I activity develops at about the same rate as that of the controls during the first 48 h of greening, after which it develops at a slower rate. The rate of development of photosystem II activity in cells greening in a glucose medium lags considerably behind that of the controls until the later hours of greening. There are similarities between glucose inhibition and chloramphenicol inhibition of chloroplast development. Glucose may inhibit a step in chloroplast development ultimately controlled by the chloroplast genome.  相似文献

The dispersal capabilities of intertidal organisms may represent a key factor to their survival in the face of global warming, as species that cannot adapt to the various effects of climate change will have to migrate to track suitable habitat. Although species with pelagic larval phases might be expected to have a greater capacity for dispersal than those with benthic larvae, interspecies comparisons have shown that this is not always the case. Consequently, population genetic approaches are being increasingly used to gain insights into dispersal through studying patterns of gene flow. In the present study, we used nuclear single‐nucleotide polymorphisms (SNPs) and mitochondrial DNA (mtDNA) sequencing to elucidate fine‐scale patterns of genetic variation between populations of the Black Katy Chiton, Katharina tunicata, separated by 15–150 km in south‐west Vancouver Island. Both the nuclear and mitochondrial data sets revealed no genetic differentiation between the populations studied, and an isolation‐with‐migration analysis indicated extensive local‐scale gene flow, suggesting an absence of barriers to dispersal. Population demographic analysis also revealed long‐term population stability through previous periods of climate change associated with the Pleistocene glaciations. Together, the findings of the present study suggest that this high potential for dispersal may allow K. tunicata to respond to current global warming by tracking suitable habitat, consistent with its long‐term demographic stability through previous changes in the Earth's climate. © 2012 The Linnean Society of London, Biological Journal of the Linnean Society, 2012, 106 , 589–597.  相似文献

The monophyletic family Zhangsolvidae comprises stout‐bodied brachyceran flies with a long proboscis and occurring only in the Cretaceous, originally known in shale from the Early Cretaceous Laiyang Formation (Fm.) in China (Zhangsolva Nagatomi & Yang), subsequently from limestones of the Early Cretaceous Crato Fm. of Brazil. Cratomyoides Wilkommen is synonymized with Cratomyia Mazzarolo & Amorim, both from the Crato Fm.; Cratomyiidae is synonymized with Zhangsolvidae. Two genera and three species of Zhangsolvidae are described: Buccinatormyia magnifica Arillo, Peñalver & Pérez‐de la Fuente, gen. et sp.n. and B. soplaensis Arillo, Peñalver & Pérez‐de la Fuente, sp.n. , in Albian amber from Las Peñosas Fm. in Spain; and Linguatormyia teletacta Grimaldi, gen. et sp.n. , in Upper Albian–Lower Cenomanian amber from Hukawng Valley in Myanmar. Buccinatormyia soplaensis and Linguatormyia teletacta are unique among all Brachycera, extant or extinct, by their remarkably long, flagellate antennae, about 1.6× the body length in the latter species. A phylogenetic analysis of 52 morphological characters for 35 taxa is presented, 11 taxa being Cretaceous species, which supports placement of the family within Stratiomyomorpha, although not to any particular family within the infraorder. Hexokinase (HXK) is present in all virtually living organisms and is central to carbohydrate metabolism catalysing the ATP‐dependent phosphorylation of hexoses. In plants, HXKs are supposed to act as sugar sensors and/or to interact with other enzymes directly supplying metabolic pathways such as glycolysis, the nucleotide phosphate monosaccharide (NDP‐glucose) pathway and the pentose phosphate pathway. We identified nine members of the tobacco HXK gene family and observed that among RNAi lines of these nine NtHXKs, only RNAi lines of NtHXK1 showed an altered phenotype, namely stunted growth and leaf chlorosis. NtHXK1 was also the isoform with highest relative expression levels among all NtHXKs. GFP‐tagging and immunolocalization indicated that NtHXK1 is associated with mitochondrial membranes. Overexpression of NtHXK1 resulted in elevated glucose phosphorylation activity in leaf extracts or chloroplasts. Moreover, NtHXK1 was able to complement the glucose‐insensitive Arabidopsis mutant gin2‐1 suggesting that NtHXK1 can take over glucose sensing functions. RNAi lines of NtHXK1 showed severely damaged leaf and chloroplast structure, coinciding with an excess accumulation of starch. We conclude that NtHXK1 is not only essential for maintaining glycolytic activity during respiration but also for regulating starch turnover, especially during the night.  相似文献

1. Agricultural intensification has caused dramatic biodiversity loss in many agricultural landscapes over the last century. Here, we investigated whether new types of farm ponds (made of artificial substrata) in intensive systems and natural‐substratum ponds in traditional farming systems differ in their value for aquatic biodiversity conservation. 2. We analysed the main patterns of environmental variation, compared α‐, β‐ and γ‐diversity of macroinvertebrates between ponds types and evaluated the role of submerged aquatic vegetation (SAV). Generalised additive models (GAM) were used to analyse the relationships of α‐ and β‐diversity with environmental predictors, and variation partitioning to separate the effect of environmental and spatial characteristics on the variation in macroinvertebrate assemblages. Moran’s eigenvector maps (MEMs) were used to define spatial variables. 3. A principal coordinate analysis (PCoA) detected a primary environmental gradient that separated nutrient‐rich ponds from those dominated by SAV; a secondary morphometric gradient distinguished natural‐substratum ponds, with large surface area and structural complexity, from artificial‐substratum ponds with steeper slopes. Natural‐substratum ponds had almost twice the α‐ and γ‐diversity of artificial‐substratum ponds, and diversity significantly increased when SAV was present, particularly in artificial‐substratum ponds. Total phosphorus (TP) strongly contributed to explain the patterns in diversity, while SAV was a significant predictor of assemblage composition and diversity. GAMs revealed optima of both α‐diversity at intermediate SAV covers and β‐diversity at intermediate–high TP concentrations. 4. These findings have important implications for conservation planning. Adaptation of artificial‐substratum ponds by adding natural substratum and smoothing the gradient of pond margins would improve their conservation value. Development of SAV with occasional harvests and certain cautionary measures to control nutrient levels may also improve both the agronomical and environmental function of ponds.  相似文献

SUMMARY. 1 Mechanisms which might have caused a switch from submerged plant dominance to phytoplankton dominance in a series of shallow lakes, the Norfolk Broads, during the 1950s and 1960s, are reviewed. It is argued that a likely mechanism was a poisoning of the community of Cladocera which graze on algae and are associated with the plants. This allowed phytoplankton to take advantage of the increased nutrient loadings and to increase. The 1950s and 1960s were periods of liberal organochlorine pesticide use. Cladocera are particularly susceptible to organochlorine toxicity.
2. Sediment cores were taken from Hoveton Great Broad, in which the switch to phytoplankton has occurred, and from two Broads (Upton and Martham South Broads) in which submerged plants are still dominant. Analyses were made for cladoceran remains, diatoms and molluscs. Cross dating with previously taken dated cores allowed time-scales to be applied.
3. In Hoveton Great Broad, a switch from clear-water-associated chydorids to turbid-water forms was found to coincide with the loss of aquatic plants in the 1950s. No Daphnia remains were found. In the other two Broads, clear-water chydorids, mostly plant-associated, were found throughout the cores.
4. Residues of dieldrin (HEOD), DDD and TDE were found in the Hoveton Great Broad cores, with the DDT derivatives particularly associated with the end of the phase of submerged plant dominance and the beginning of that of phytoplankton dominance. Bosmina remains became more abundant after this point. Pesticide residues were scarce in the cores from the other two Broads and did not form any particular pattern. Residues of polychlorinated biphenyls were widespread in all cores.
5. Calculation of DDT concentration in the water at the time the sediments with greatest concentrations of residue were laid down suggests that concentrations above typical LC50 values for Daphnia species could have been present.  相似文献

Transposon tagging with modified maize DsGUS constructswas used to isolate genes induced by oxygen deprivation in Arabidopsisthaliana. Seedlings of 800 gene-trap (DsG) and 600 enhancer-trap(DsE) lines were grown on vertically positioned plates for 1 week,oxygen deprived for up to 24 h and stained for GUS activity.Oxygen deprivation induced intricate patterns of gene expressionin seedlings of 65 lines. The insertion site and phenotypesof 15 lines were examined. Surprisingly, none of the insertionswere into genes that encode known anaerobic polypeptides. Insertionswere identified within or adjacent to genes encoding proteinsof regulatory, enzymatic, mitochondrial protein import and unknownfunction, as well as adjacent to genes encoding a putative receptor-likekinase and putative sensor-histidine kinase. Four lines hadsignificantly lower ADH activity after 24 h of oxygen deprivationand three of these showed reduced stress tolerance. Two lineswith wild-type levels of ADH were low-oxygen intolerant. Paradoxically,several lines had significantly higher ADH activity after 12 hof oxygen deprivation but reduced stress tolerance. Caffeinetreatment, which increased ADH specific activity in wild-typeseedlings under aerobic conditions, was sufficient to increaseGUS staining in seven of the 15 lines, providing evidence thatthese genes may be regulated by cytosolic calcium levels. Theseresults demonstrate the effectiveness of the Ds–GUS taggingsystem in the identification of genes that are regulated inresponse to oxygen deprivation and a calcium second messenger.  相似文献

1. Biological invasions are widely recognised as a significant component of human‐caused environmental change and a primary threat to native biodiversity. The negative impacts of species invasions are particularly evident for freshwater crayfish faunas. 2. This study provides novel insight into the ecological effects of native and non‐native crayfish on zoobenthic communities (with emphasis on the non‐native rusty crayfish, Orconectes rusticus) across broad scales by combining a meta‐analysis of small‐scale experimental studies with a long‐term observational study conducted over a 24 year period in Sparkling Lake, Wisconsin, U.S.A. (46°00′N, 89°42′W). 3. The meta‐analysis summarised quantitatively the results of cage experiments for seven species of crayfish spanning four continents. We found that total zoobenthos densities (primarily Gastropoda and Diptera) were significantly lower in treatments containing crayfish relative to controls; a result that was significant for non‐native crayfish but not for crayfish in their native range, perhaps owing to a small sample size. In contrast to other species, rusty crayfish were also negatively associated with Ephemeroptera. 4. Results from the time series analysis comparing temporal trends in rusty crayfish and invertebrate abundances from Sparkling Lake were consistent with the findings from the meta‐analysis. Rusty crayfish were negatively correlated with the abundance of total zoobenthos, Diptera, Ephemeroptera and Odonata, as well as families of Trichoptera. 5. By coupling the results from short and long‐term research, our study offers greater insight into the nature of crayfish‐invertebrate interactions in aquatic systems, revealing consistent effects of invasive crayfish on native fauna. The control and management of invasive species is facilitated by the knowledge that well executed small‐scale studies may be extrapolated to understand larger‐scale ecological interactions.  相似文献

Air flow over a Populus leaf was investigated using a hot-wireanemometer. When the air flow in the wind tunnel was laminar,the boundary layer was often turbulent at a wind speed of only1.5 m s–1, particularly when encouraged by uneven topographyand roughness of the surface, as on the lower side of the leaf.The smoother upper surface behaved in a similar way to a flatplate when at low wind speeds, and the profiles of wind speedcould be shown to be equivalent to those expected from laminarboundary layer theory. Nevertheless, the boundary layer becameturbulent at Reynolds numbers much lower than those requiredto cause the transition to turbulence in a flat plate. Turbulentair flow in the wind tunnel greatly increased boundary layerturbulence but had only a small effect on evaporation from amodel of the leaf. The evaporation rates observed were 2.5 timeshigher than expected from theory, irrespective of the turbulenceregime.  相似文献
