Biological systems with asexual reproduction have often attracted research on parasites and host immune defence, because parasites are expected to be better able to exploit genetically less diverse populations. In addition, maternally inherited parasitic microorganisms such as Wolbachia can directly alter the reproductive systems of their hosts and induce parthenogenesis. In the freshwater ostracod Eucypris virens, both sexual and asexual reproduction is known, and we speculated that parasite pressures might help to explain their co‐existence. This species complex inhabits shallow, often eutrophic temporary water bodies, conditions that should provide ample opportunities for parasite infections. We surveyed natural populations of E. virens throughout its Europe‐wide range for natural parasites, and particularly tested for the presence of intracellular Wolbachia bacteria. Surprisingly, the results indicate that very few E. virens populations support parasite infections. We also found no evidence for the presence of Wolbachia in the populations screened. Female mate choice is considered an important evolutionary agent, but there has been an ongoing debate over the fitness consequences it produces, especially in species that have a resource‐free mating system. We examined a potential fitness benefit resulting from the pre‐spawning mate preference in Arctic charr Salvelinus alpinus, a salmonid fish with no parental care. The females were first allowed to discriminate behaviourally between two males presented to them in a free choice test. We then tested with controlled fertilizations whether the females would accrue indirect genetic benefits for their offspring, as measured by embryonic viability, if they had mated with the male they preferred. Both parental identities influenced offspring survivorship, but the females did not consistently prefer the male which gave her the higher reproductive success. Neither was the degree of male red breeding coloration associated with female preference or the observable genetic quality. In contrast, there was a negative relationship between female coloration and her offspring survivorship, suggesting a significant trade‐off in resource investment between sexual ornamentation and reproduction. To conclude, the potential indirect fitness consequences arising from females' pre‐spawning mate preference seem to be negligible in early stages of development of Arctic charr. © 2011 The Linnean Society of London, Biological Journal of the Linnean Society, 2011, 103 , 602–611.  相似文献

1. The concepts of community assembly and succession are closely related, yet their foci differ slightly. Succession describes the trajectory of species replacements during the temporal development of the community, while assembly also allows that a locality can harbour different communities depending on the events in the near past of community development. 2. The aims of this study were (i) to examine the year‐to‐year variation in phytoplankton community assembly among basins of different trophy and disturbance in the large boreal Lake Hiidenvesi and (ii) to assess community persistence and diversity among basins in relation to prevailing environmental factors. 3. The results showed that the assembly did not follow similar trajectories each year. According to mean similarity analyses, there was a large degree of variability especially among the groups of samples collected in the same months of different years. Similarity between pairs of consecutive samples was highest in a cold year (1998) in all basins. Community assembly was most unpredictable in the basin of highest productivity, perhaps implying that the number of alternative stable states increased towards higher productivity. Our data also showed a strong unimodal relationship between phytoplankton species richness and grazing by cladoceran zooplankton in the basin of highest trophy. 4. This study showed that phytoplankton community assembly exhibited large variability among the years. This implies that different environmental conditions might be the strongest mechanism behind this pattern, given that the degree of community similarity paralleled the year‐to‐year variation in mean temperature. Unravelling the patterns in community assembly has a number of important implications, especially for the monitoring of ecological impacts based only on snapshots of biological assemblages.  相似文献

1. Most Finnish streams were channelised during the 19th and 20th century to facilitate timber floating. By the late 1970s, extensive programmes were initiated to restore these degraded streams. The responses of fish populations to restoration have been little studied, however, and monitoring of other stream biota has been negligible. In this paper, we review results from a set of studies on the effects of stream restoration on habitat structure, brown trout populations, benthic macroinvertebrates and leaf retention. 2. In general, restoration greatly increased stream bed heterogeneity. The cover of mosses in channelised streams was close to that of unmodified reference sites, but after restoration moss cover declined to one‐tenth of the pre‐restoration value. 3. In one stream, densities of age‐0 trout were slightly lower after restoration, but the difference to an unmodified reference stream was non‐significant, indicating no effect of restoration. In another stream, trout density increased after restoration, indicating a weakly positive response. The overall weak response of trout to habitat manipulations probably relates to the fact that restoration did not increase the amount of pools, a key winter habitat for salmonids. 4. Benthic invertebrate community composition was more variable in streams restored 4–6 years before sampling than in unmodified reference streams or streams restored 8 years before sampling. Channelised streams supported a distinctive set of indicator species, most of which were filter‐feeders or scrapers, while most of the indicators in streams restored 8 years before sampling were shredders. 5. Leaf retentiveness in reference streams was high, with 60–70% of experimentally released leaves being retained within 50 m. Channelised streams were poorly retentive (c. 10% of leaves retained), and the increase in retention following restoration was modest (+14% on average). Aquatic mosses were a key retentive feature in both channelised and natural streams, but their cover was drastically reduced through restoration. 6. Mitigation of the detrimental impacts of forestry (e.g. removal of mature riparian forests) is a major challenge to the management of boreal streams. This goal cannot be achieved by focusing efforts only on restoration of physical structures in stream channels, but also requires conservation and ecologically sound management of riparian forests.  相似文献

The Siberian Tit Parus cinctus population of Finland, and probably of the whole of Fennoscandia, has declined dramatically during this century. Understanding its population dynamics is essential for its conservation. We studied annual survival rate and dispersal distance of both breeding and fledgling Siberian Tits during 1989-97 in a moderately managed forest habitat near the southern border of its range in Kuusamo, northeastern Finland. Breeding density was low, averaging 0.51 pairs/km in a nestbox area of about 42 km2. This was probably an underestimate, because we did not search for nests in natural holes. However, following its population decline, overall densities at the southern border of the range are low. The study area was, however, suitable breeding habitat as reflected by their high nesting success. Brood size at fledging averaged 7.45 young, and reproductive output was 6.16 young per breeding pair, including failed nests. Clutch size adjustment was successful among pairs producing fledglings because, on average, 92.6% of the eggs laid produced young in successful broods. Median distance between consecutive breeding attempts was 100 m for both males and females (range 0–2500 m and 0–6000 m, respectively). Natal dispersal distance was significantly longer in females than in males. We applied Cormack-Jolly-Seber modelling to estimate survival and recapture probabilities separately. Survival of breeding birds was not sex-related, averaging 0.69 annually. Some of the ringed fledglings (3.0%) were later captured as breeders in the area. This is an underestimate of local survival rate due to incomplete recapturing of breeding birds. The implications of these results with respect to the conservation status of the species are discussed.  相似文献

Recent papers have promoted the view that model‐based methods in general, and those based on Approximate Bayesian Computation (ABC) in particular, are flawed in a number of ways, and are therefore inappropriate for the analysis of phylogeographic data. These papers further argue that Nested Clade Phylogeographic Analysis (NCPA) offers the best approach in statistical phylogeography. In order to remove the confusion and misconceptions introduced by these papers, we justify and explain the reasoning behind model‐based inference. We argue that ABC is a statistically valid approach, alongside other computational statistical techniques that have been successfully used to infer parameters and compare models in population genetics. We also examine the NCPA method and highlight numerous deficiencies, either when used with single or multiple loci. We further show that the ages of clades are carelessly used to infer ages of demographic events, that these ages are estimated under a simple model of panmixia and population stationarity but are then used under different and unspecified models to test hypotheses, a usage the invalidates these testing procedures. We conclude by encouraging researchers to study and use model‐based inference in population genetics.  相似文献

A non‐vented non‐steady state flow‐through chamber and a non‐vented non‐steady state non‐flow‐through chamber technique were used to measure CO2 efflux of a young Scots pine forest on a fertile till soil in southern Finland. Soil temperature, soil moisture and soil CO2 concentration were measured concurrently with CO2 efflux for two and a half successive years. The CO2 efflux showed a seasonal pattern, effluxes ranging from low 0.0–0.1 g CO2 m ? 2 h ? 1 in winter to peak values of 2.3 g CO2 m ? 2 h ? 1 occurring in late June and in July. The daily average effluxes in July measured by flow through chambers were 1.23 and 0.98 g CO2 m ? 2 h ? 1 in 1998 and 1999, respectively. The annual accumulated CO2 efflux was 3117 and 3326 g CO2 m ? 2 in 1998 and 1999, respectively. The spatial variation in CO2 efflux was high (CV 0.18–0.45) and increased with increasing efflux. Soil air CO2 concentration showed similar seasonal pattern the peak concentrations occurring in July–August. The CO2 concentrations ranged from 580 to 780 µ mol mol ? 1 in the humus layer to 13 620–14 470 µ mol mol ? 1 in the C‐horizon. In winter the soil air CO2 concentrations were lower, especially in deeper soil layers. Drought decreased CO2 efflux and soil air CO2 concentration. The in situ comparison on forest soil between the chamber methods showed the non‐flow‐through chamber to give ~~50% lower efflux values than that of the flow‐through chamber. When calibrated against known CO2 efflux ranging from 0.4 to 0.8 g CO2 m ? 2 h ? 1 generated with a diffusion box method developed by Widén and Lindroth [Acta Universitatis Agriculturae Suecia Silvestria, 2001], the flow‐through chamber gave equal effluxes at the lower end of the calibration range, but overestimated high effluxes by 20%. We studied habitat choice, distribution and reproductive success in the Pied Flycatcher Ficedula hypoleuca in relation to the spatial structure of a fragmented forest area. Two hierarchical levels of forest habitat structure were used: (1) forest patch size (macrohabitat) and (2) vegetative structure within forest patches (microhabitat). In spring, both males and females settled preferentially in large and medium-sized forest stands (>1 ha) where breeding density was also higher than in small stands (<1 ha). Stands <5 ha were occupied later, and the proportion of unpaired males was higher there than in large stands (>5 ha). The known age distributions of breeding birds and breeding success were independent of forest patch size. Nest predation rate was not associated with stand size or nest distance from the forest edge. The preference of breeding birds for large forest patches was presumably related to the higher amount of resources (nest sites, mates, food) a large patch can offer for reproduction compared with a small patch. At the microhabitat level, territories of old males were characterized by relatively more deciduous trees that contained more invertebrate food than coniferous tree-dominated territories of yearling males. The reproductive success of old males, as measured by the number of fledged young per male, was higher than that of yearling males. This suggests that the larger body size and blacker plumage of old males possibly contributed to male dominance and that old males excluded younger males from preferred habitats through territorial behaviour. Our results suggest that habitat choice of the Pied Flycatcher was affected by both micro- and macrohabitat. On the basis of settlement pattern, density and reproductive success, the distribution of Pied Flycatchers across forest stands of different sizes followed the ideal-free distribution model, whereas at the microhabitat level, age-related unequal distribution of males followed the ideal-despotic model of Fretwell and Lucas.  相似文献

Background

Mesenchymal stromal cell (MSC)–based therapy has great potential to modulate chronic inflammation and enhance tissue regeneration. Crosstalk between MSC-lineage cells and polarized macrophages is critical for bone formation and remodeling in inflammatory bone diseases. However, the translational application of this interaction is limited by the short-term viability of MSCs after cell transplantation.

Methods

Three types of genetically modified (GM) MSCs were created: (1) luciferase-expressing reporter MSCs; (2) MSCs that secrete interleukin (IL)-4 either constitutively; and (3) MSCs that secrete IL-4 as a response to nuclear factor kappa-light-chain-enhancer of activated B cell (NFκB) activation. Cells were injected into the murine distal femoral bone marrow cavity. MSC viability and bone formation were examined in vivo. Cytokine secretion was determined in a femoral explant organ culture model.

Results

The reporter MSCs survived up to 4 weeks post-implantation. No difference in the number of viable cells was found between high (2.5?×?106) and low (0.5?×?106) cell-injected groups. Injection of 2.5?×?106 reporter MSCs increased local bone mineral density at 4 weeks post-implantation. Injection of 0.5?×?106 constitutive IL-4 or NFκB-sensing IL-4–secreting MSCs increased bone mineral density at 2 weeks post-implantation. In the femoral explant organ culture model, LPS treatment induced IL-4 secretion in the NFκB-sensing IL-4–secreting MSC group and IL-10 secretion in all the femur samples. No significant differences in tumor necrosis factor (TNF)α and IL-1β secretion were observed between the MSC-transplanted and control groups in the explant culture.

Discussion

Transplanted GM MSCs demonstrated prolonged cell viability when transplanted to a compatible niche within the bone marrow cavity. GM IL-4–secreting MSCs may have great potential to enhance bone regeneration in disorders associated with chronic inflammation.  相似文献

Fens, which extend over vast areas in the Northern hemisphere, are sources of the greenhouse gas CH4. Climate change scenarios predict a lowering water table (WT) in mires. To study the effect of WT drawdown on CH4 dynamics in a fen ecosystem, we took advantage of a WT drawdown gradient near a ground water extraction plant. Methane fluxes and CH4 production and oxidation potentials were related to microbial communities responsible for the processes in four mire locations (wet, semiwet, semidry, and dry). Principal component analyses performed on the vegetation, pH, CH4, and WT results clearly separated the four sampling locations in the gradient. Long‐term lowering of WT was associated with decreased coverage of Sphagnum and aerenchymatic plants, decreased CH4 field emissions and CH4 production potential. Based on mcrA terminal restriction fragment length polymorphism the methanogen community structure correlated best with the methane production and coverage of aerenchymatic plants along the gradient. Methanosarcinaceae and Methanocellales were found at the pristine wet end of the gradient, whereas the Fen cluster characterized the dry end. The methane‐oxidizing bacterial community consisted exclusively of Methylocystis bacteria, but interestingly of five different alleles (T, S, R, M, and O) of the particulate methane monooxygenase marker gene pmoA. The M allele was dominant in the wet locations, and the occurrence of alleles O, S, and T increased with drainage. The occurrence of the R allele that characterized the upper peat layer correlated with CH4 oxidation potential. These results advance our understanding of mire dynamics after long‐term WT drawdown and of the microbiological bases of methane emissions from mires.  相似文献
