A Note on the Enumeration of Epiphytic Bacteria by Microscopic Methods with Particular Reference to Two Freshwater Plants 总被引:6,自引:5,他引:1
Bacteria on plant surfaces were examined using epifluorescence, bright-field microscopy and an impression technique. Staining bacteria directly on the plant surface with phenolic aniline blue was found to be the best method to use for the determination of bacterial density. The effect on the estimation of population density of pretreatment of the plant with agents such as methanol and eosin yellowish was investigated. The average sizes of the bacterial populations on two freshwater plants, Rorippa and Lemna , estimated after staining by this method, were 5 times 106 and 9 times 106 bacteria cm-z respectively. 相似文献

Mitotic PtK1cells were treated both during mid-anaphase and at furrow initiation with the potent microtubule (MT) stabilizing agent, taxol, to determine the role of MTs in the rate of cytokinetic events. Rates of cytokinesis (μm/min) were measured by changes in furrow diameter. Incubation of PtK1cells during mid-anaphase with 5 μg/ml taxol slows the rate of cytokinesis by an average of 43%. Instead of furrow initiation to midbody formation taking an average of 10.7 min (1.6 μm/min), furrowing to midbody formation was completed in an average of 19.0 min (0.9 μm/min), which does not include the 7-min period between taxol application in mid-anaphase and furrow initiation. Application of 5 μg/ml taxol to cells at furrow initiation had a reduced effect on decreasing the rate of cytokinesis and midbody formation; furrowing to midbody formation took an average of 14.6 min (1.2 μm/min). These data suggest that delays in the rate of cytokinesis is dependent on the mitotic stage at which taxol is applied. Ultrastructural analysis shows that taxol treatment of anaphase cells prevents midbody formation during early G1, yet MT number and organization in the furrowed region is not significantly altered from untreated cells. There is little change in the organization and amount of contractile ring microfilaments, yet filaments are also found parallel to midbody MTs. Our results may be explained by the fact that taxol tends to stabilize MTs which probably affects the rate at which they depolymerize in the terminal phases of cytokinesis. 1. Benthic macroinvertebrates were sampled for 1 year to assess functional and taxonomic differences in invertebrate biomass and production with respect to habitat types, reaches and catchments in Wine Spring Basin, western North Carolina. Quantitative samples were collected from depositional, cobble-riffle and bedrock outcrop habitats at four stream reaches (two headwater sites, one second order, and one third order). Other measures included physical parameters, periphyton and organic matter standing crops. Invertebrate data from the Wine Spring catchment were also compared with data from another catchment (Ball Creek) within the same region. 2. The three habitat types had different current velocities and mean substratum particle sizes; both measures were greatest in bedrock outcrop habitats and lowest in depositional habitats. Organic matter standing crops, invertebrate functional group productivity and biomass also differed significantly with respect to habitat type. Cobble-riffle areas had the lowest standing crops of organic matter, invertebrate productivity and biomass. 3. Both invertebrate communities and organic matter standing crops differed significantly between the two headwater reaches. First- to third-order reaches differed in taxonomic composition at the genus level, yet had similar relative functional group productivity and biomass. 4. Annual mean invertebrate biomass and secondary production were greater in the Wine Spring Basin than in Ball Creek. Sites in both the Wine Spring and Ball Creek catchments, however, exhibited similar functional group distributions per habitat type. 5. Local geomorphology and related physical parameters influenced the structure of invertebrate functional group composition, and the distribution of organic matter standing crops. SYNOPSIS. Cuticular proteins show specificity for stage, age,and anatomical region. Analysis of the cuticular proteins ofsecond pupae created by application of juvenile hormone demonstratesthat the hormone prevents the onset of new sequences of synthesesand favors repetition of the region-specific, temporal patternof syntheses used in the previous stage. The argument made isthat juvenile hormone might exert this "status quo" action bypreventing alterations in chromatin configuration. Evidencefrom a wide variety of systems shows that polyamines might beinvolved in reprogramming chromatin. Ecdysterone induces ornithinedecarboxylase, the rate limiting enzyme in animal polyaminesynthesis. I suggest that juvenile hormone might be exertingits status quo action by inhibiting this induction. Preliminarystudies of ornithine decarboxylase induction support this specifichypothesis; experiments with an inhibitor of this enzyme, -difluoromethylornithine, however, do not show the expected juvenile hormonemimicry. The early Miocene sites of Moruorot and Kalodirr (Kenya, 17.5 Myr) have yielded a rich collection of mammals. New listriodont material from these localities, including a complete skull and a partial mandible, provide long awaited information on cranial features of early bunodont Listriodontinae. The evolution and systematics of the group are highly debated, especially regarding its first representatives. The new material described here sheds light on the differentiation of bunodont Listriodontinae in Africa and clarifies the systematics of the group. The first phylogenetic analysis of the Listriodontinae is here performed and supports close relationships between Kubanochoerus and a clade (Eurolistriodon, Listriodon). Lopholistriodon is the most basal representative of the listriodontine clade.
