The crystal structure of recombinant rabbit interferon-gamma was solved by the multiple isomorphous replacement technique at 2.7-A resolution and refined to a crystallographic R-factor of 26.2%. The interferon crystallizes with one-half of the functional dimer in the asymmetric unit, with the two polypeptide chains of the dimer related by a crystallographic 2-fold symmetry axis. The structure is predominantly alpha-helical with extensive interdigitation of the alpha-helical segments of the two polypeptide chains. 相似文献

Cellular receptors for IgG (Fc gamma R) mediate important protective functions. By using site-specific mutants of a chimeric antibody (mouse V H domain and L chain; human IgG3 C H domains), we have demonstrated that human Fc gamma RI interacts with a site in the lower hinge of human IgG (residues 234 to 237) and that this interaction dictates Fc gamma RI-mediated superoxide generation. Mutations at position 235 resulted in the most profound reductions in Fc gamma RI recognition. We have also mapped an interaction site for Fc gamma RII to the same region; however, mutations at position 234 and 237 resulted in the greatest reductions in Fc gamma RII recognition. The two receptors appear to recognize overlapping but nonidentical sites on the lower hinge of IgG. Deviations from the optimal motif 234-Leu-Leu-Gly-Gly-237 may then explain the human IgG subclass specificity profile for human Fc gamma RI and Fc gamma RII. 相似文献

To study the mechanisms of discrimination between various forms of vitamin E, four normal subjects, one patient with lipoprotein lipase deficiency, and three patients with abnormal apolipoprotein B-100 production were given an oral dose containing three tocopherols labeled with differing amounts of deuterium (2R,4'R,8'R-alpha-(5,7-(C2H3)2)tocopheryl acetate (d6-RRR-alpha-tocopheryl acetate), 2S,4'R,8'R-alpha-5-(C2H3)tocopheryl acetate (d3-SRR-alpha-tocopheryl acetate), and 2R,4'R,8'R-gamma-(3,4-2H)tocopherol (d2-RRR-gamma-tocopherol). The tocopherol contents of plasma, red cells, and lipoproteins were measured up to 76 h after the dose. In normal subjects all three tocopherols were absorbed and secreted in chylomicrons with equal efficiencies. Both d2-gamma- and d3-SRR-alpha-tocopherols peaked at similar concentrations in the other lipoprotein fractions, then decreased similarly, but 2-4 times more rapidly than did d6-RRR-alpha-tocopherol. A lipoprotein lipase-deficient patient and a patient with prolonged production of chylomicrons with absent apolipoprotein B-100 also demonstrated the lack of discrimination between tocopherols during absorption. Despite abnormal apolipoprotein B-100 production in two patients, the "VLDL" was preferentially enriched in d6-RRR-alpha-tocopherol. Our results show that there is no discrimination between the three tocopherols during absorption and secretion in chylomicrons, but subsequently there is a preferential enrichment of very low density lipoprotein (VLDL) with RRR-alpha-tocopherol. Catabolism of this VLDL results in the maintenance of plasma RRR-alpha-tocopherol concentrations. 相似文献

Two different crystal forms of recombinant rabbit IFN-gamma were obtained under different crystallization conditions. The first, a tetragonal form with space group P43212 or P41212, was obtained through vapor phase equilibration using the sitting drop rods technique with ammonium citrate as the major precipitating agent. The unit cell dimensions of this crystal form are a = b = 82.1 A and C = 116.3 A. These crystals diffract to 2.8 A resolution and contain a dimer in the asymmetric unit. A second crystal form was obtained by the batch method at pH 8.0 using sodium chloride as the precipitating agent. The crystals are hexagonal, space group P6122 or P6522, and with unit cell dimensions of a = b = 58.0 A and c = 169 A. This form contains monomer in the asymmetric unit and diffracts to greater than 2.7 A resolution. Both forms appear to be eminently suitable for further analyses and crystal structure solution. 相似文献

Larvae of the mayfly (Drunella grandis [Eaton]) from Diamond Fork Creek, Utah, were covered with a heavy growth of the sulfide-oxidizing bacterium Thiothrix. The bacterium did not seem to harm the mayfly, but the Thiothrix trichomes were parasitized by three morphologically distinct bacteria, two of which were cytoplasmic and one of which was probably periplasmic. At least two of the parasites destroyed the cytoplasmic contents of the Thiothrix sp., thus killing the host cell. Attempts to obtain the parasites in pure culture were unsuccessful. 相似文献

Mannosidosis: separation and characterization of two acid alpha-mannosidase forms in mutant fibroblasts 总被引:2,自引:0,他引:2
Two acidic forms of alpha-mannosidase activity, A and B, are separated and characterized in fibroblasts from controls and patients with mannosidosis. In normal cells, A and B differ in adsorption on DEAE-cellulose, electrophoretic mobility, stability at 70 degrees C and resistance to freezing at --20 degrees C. In 5 of 6 mutant cell lines, A and B both exhibit altered Km's for artificial substrates and decreased thermal stability. Zn2+ has no effect on A or B in mutant or normal cells. In contrast, Co2+ slightly inhibits B in normal cells but markedly enhances the activity of B in mutant cells. The mutation in mannosidosis clearly results in an alteration in the biochemical characteristics of both major acidic forms of alpha-mannosidase. 相似文献

Four benthic marine communities occur in the clastic facies of the prograding Upper Frasnian-Lower Famennian (Upper Devonian) Foreknobs Formation in the Central Appalachians along the Allegheny Front in Maryland, West Virginia, and Virginia. Deep-water, rapidly prograding environments were inhabited by the Ambocoelia-Chonetes Community, dominated by an epifauna of unattached brachiopods. Offshore bar environments were inhabited by the Cyrtospirifer-Camarotoechia Community, exhibiting adaptations to shallow-water, high-energy conditions and probably lowered salinities. Shallow-water, sublittoral environments were inhabited by the Atrypa-Cypricardella Community, a community in which existed a variety of life habitats and a diverse epifaunal and infaunal association of brachiopods and bivalve molluscs. The Leptodesma-Tylothyris Community flourished in nearshore bar-protected environments in the southern region of the study area, whereas in the north the Cyrtospirifer-Camarotoechia Community inhabited nearshore environments in conjunction with the onshore development of a large fluviodeltaic system. 相似文献

Lipids were extracted from fresh, field-grown coastal bermudagrass (Cynodon dactylon (L.) Pers.) and the fatty acids determined by gas chromatography. Total fatty acid levels (dry weight basis) increased during the day and reached a single maximum at sunset in 2-week-old grass; whereas, in older grass, the fluctuations in fatty acid levels showed two maxima. The first maximum occurred 4 h after sunrise and the second maximum occurred at sunset. Total fatty acid levels, based on dry weight, decreased during the first 6 weeks of growth and changed very little after an additional 4 weeks' growth in bermudagrass leaf blades. Chlorophyll levels (dry weight basis) continuously decreased during the entire growth period (10 weeks). Chlorophyll alb ratios increased at sunset in 2- and 6-week-old grass, but this ratio did not change during the day in subsequent growth stages. The results of these experiments show that stages of maturity affected fatty acid fluctuations during the day as well as total fatty acid and chlorophyll levels in Coastal bermudagrass leaf blades. Franz Boas spent several weeks at Fort Rupert, British Columbia, at the end of 1894, when he saw the Kwakiutl hamatsa ritual in situ for the first time. Soon after his return east Boas posed for a series of photographs in the U.S. National Museum for a diorama of the hamatsa dance.
