Leishmania donovani promastigotes in late-stationary phase incorporated label from [2-14C]acetate and [1-14C]laurate into the mannose residues of mannan, thus confirming the presence of a functional glyoxylate bypass in these parasitic protozoa. Isolated, washed calls also incorporated label from [2-14C]acetate and [1-14C]laurate into mannan during a 1-hr incubation in buffer. Glucose had no effect on label incorporation into mannan, but glutamate caused over a four-fold increase in incorporation from [2-14C]acetate and a 2.4-fold increase from [1-14C]laurate. Staurosporine, a protein kinase inhibitor that inhibits glutamate and alanine oxidation, did not inhibit label incorporation from [2-14C]acetate into mannan. Hyperosmolality caused about a 33% inhibition of label incorporation into mannan. These results show the glyoxylate cycle and/or the subsequent biosynthetic pathway from fructose-6-phosphate to mannan are subject to regulation.  相似文献

SUMMARY. 1. The vertical distribution within the substrate of three lotic mayflies (Rhithrogena sp., Paraleptophlebia sp. and Baetis tricaudalus Dodds) was examined in laboratory streams to determine if diel patterns of vertical movement across substrate surfaces existed and whether light intensity, temperature, substrate composition or food availability influenced the observed movement patterns.
2. Rhithrogena sp. and B. tricaudatus exhibited significant diel movement from lower substrate surfaces during the day to upper surfaces at night, although >64% of B. tricaudalus occupied the upper surfaces throughout the 24 h period. In contrast, Parakplophlebia sp. larvae did not change their vertical distribution over the 24 h period.
3. The proportion of mayflies occupying substrate surfaces was influenced by the amount of upper and lower surface area available in the substrate and, possibly, crevice sizes associated with lower surfaces.
4. The movement of Rhiihrogena sp. from lower surfaces during the day to the upper surfaces during the night was influenced by changes in light intensity, but not by changes in food availability or water temperature.
5. The diel patterns of vertical movement exhibited by lotic mayflies in this investigation raise the possibility that at least some biotic interactions may vary over a diel cycle. These could include competitive interactions or temporal cycling of prey availability to predators.  相似文献

Several lines of experimental evidence are presented suggesting that the L antigens in low potassium (LK) sheep red cells are associated with separate Na(+)K(+) pump flux is distinct from the action of anti-L(l) on K(+) leak flux, implying that K(+) leak transport sites may not be converted into active pumps by the L antiserum. Treatment of LK red cells with trypsin completely abolished both the stimulation of K(+) pump flux and the enhancement of the rate of ouabain binding brought about by anti- L. That this effect is due to a total destruction of the L(p) determinant associated with the LK pump was evident from the complete failure of anti-L(p) to bind to trypsinized LK red cells. The L(p) antigen can be effectively protected against the trypsin attack by prior incubation with anti-L, indicating that the sites for antibody binding and trypsin action may be closely adjacent at the structural level. Trypsin treatment, however, did not interfere with anti-L(l) reducing ouabain insensitive K(+) leak influx, nor did it prevent binding of anti-L(ly), the hemolytically active L antibody which is probably identical with anti-L(l). The functional independence of the L(p) and L(l) sites was documented by the observation that anti-L(l) still reduced K(+) leak influx in LK cells with experimentally induced high potassium concentrations, at which K(+) pump flux is fully suppressed, whether or not anti-L(p) was binding to the L(p) antigen associated with the LK pump.  相似文献

SYNOPSIS. Mitochondria were isolated from Euglena gracilis strain Z by pressure-breakage of the cells and sucrose-cushion centrifugation. Multiple peaks (2-4) were observed in the rate of phosphorylation with Mg-ADP-phosphate concentration curves. The phosphorylative and oxidative activities were highest with NADH as the substrate, moderate with succinate, and lowest with glutamate. Inhibition of phosphorylation with 2,4-dinitrophenol and carbonyl cyanide, m-chlorophenylhydrazone gave sigmoidal concentration curves, with the extent of inhibition by DNP depending on the substrate used. Inhibition of phosphorylation by valinomycin, atractyloside, or carboxyatractyloside was only ~ 60%. Oligomycin inhibited phosphorylation in 2 phases at low and high concentrations; it inhibited Mg-ATPase in a sigmoidal fashion. Both phosphorylation and oxidation had discontinuities in Arrhenius plots at 34 C and 18 C. The relative Mg2+-dependent nucleoside triphosphatase activity was: 1 for ATP and GTP, 0.6 for ITP, 0.15 for CTP and and UTP; with Ca2+ in place of Mg2+ this activity was 0.35. Both DNP and CCCP stimulated the Mg-ATPase 50-200%. The optimal pH for the stimulation was ~ 7 regardless of the uncoupler used, and ~ 8 without the uncouplers. The few differences observed between mitochondria from Euglena and those from other sources are probably due to the fragmentation of the reticular mitochondrial structure during isolation and not to unique characteristics of these mitochondria.  相似文献

Melanophores, xanthophores, and iridophores are fundamentallydistinct chromatophores in their appearance, composition, andfunction. All migrate from their neural crest site of originto populate the integument. Their respective pigments, melanins,ptendines, and purines are found in organelles designated respectivelyas melanosomes, pterinosomes and reflecting platelets. Theseorganelles are all derived from an endoplasmic reticular vesicle.This is in keeping with a hypothesis about the common originof pigment cells from a stem cell containing a primordial organellewith the potential of becoming any of the circumscribed pigmentaryorganelles. It is believed that chromatoblasts may not be specificallydetermined until they reach a final destination where they willdifferentiate in accordance with a pattern already specifiedin the integument. Antarctica supported fossil ichthyofaunas during the Devonian,Jurassic, Cretaceous and Eocene/Oligocene. These faunas arenot ancestral to each other, nor are they related to any componentof the modern fauna. About one hundred species of notothenioidsdominate a modern fauna of over 200 species of bottom fishes.This highly endemic perciform suborder is not representedinthe fossil record of Antarctica. Notothenioids may have evolvedin situ on the margins of the Antarctic continent while graduallyadapting to cooling conditions during the Tertiary. Cladisticstudies indicate that notothenioids are a monophyletic group,but a sister group has not been identified among perciform fishes.With relatively few non-notothenioid fishes in Antarctic waters,notothenioids fill ecological roles normally occupied by taxonomicallydiverse fishes in temperate waters. There are six notothenioidfamilies: Bovichtidae, Nototheniidae, Harpagiferidae, Artedidraconidae,Bathydraconidae and Channichthyidae. Aspects of theirbiologyare briefly considered with emphasis on the Nototheniidae, themost speciose family. Evolutionary diversification within thisfamily allows recognition of species which are pelagic, cryopelagic,benthopelagic and benthic.  相似文献

ABSTRACT. The ability of Giardia strains of the duodenalis type to grow in Keister's modified TYI-S-33 medium varies with serum lot. Recently, strains of Giardia including MR4, WB, and Human-1-Portland, have been cultivated in Keister's modified TYI-S-33 medium containing the serum substitute Ultroser G and have been cultured serially at least 40 times. An optimal concentration of 8% Ultroser G promotes maximal growth in Keister's modified TYI-S-33 medium for all three strains. This concentration of Ultroser G will produce a two-log increase in the number of trophozoites in approximately three days post-inoculation. Generation times for the trophozoites ranging from 6 to 11 h have been achieved in Keister's modified TYI-S-33 containing 10% adult bovine serum and from 8 to 13 h in Keister's modified TYI-S-33 with 8% Ultroser G. Despite the excellent growth of Giardia strains in medium containing Ultroser G, the maximum trophozoite density is only about half of that achieved in Keister's modified TYI-S-33 medium supplemented with 10% adult bovine serum. Comparisons of trophozoites grown with serum or the serum substitute reveal no discernable differences in morphology and motility. Additionally, these strains have been successfully cryopreserved and revived in Keister's modified TYI-S-33 medium supplemented with Ultroser G. Because Ultroser G is a characterized mixture of six main groups of ingredients (growth factors, adhesion factors, mineral trace elements, hormones, binding proteins, and vitamins), the variability in cell proliferation that may occur when changing serum lots should be minimized when using this product.  相似文献

When grown under conditions of low relative humidity, the C3–C4intermediate Panicum milioides, as well as the C3 grasses Triticumaestivum and Poa pratense, exhibited 13C values which were upto 2–7%o less negative than the 13C values of the correspondingplants grown at high relative humidity. Along the coastal plain of Israel, shell darkness of the polymorphic land snail Xeropicta vestalis is positively related to the extent of perennial vegetation. It is not related to rain, temperature, geographic position or darkness of the ground.
White shells reflect more radiation than dark ones do, and therefore in Israel's coastal plain, where solar radiation is very strong, they are favoured. Perennial vegetation, where it occurs, shields the snails from the sun by absorbing radiation, so that snails amongst them can afford to be darker, and thus more cryptic. Hence, the more perennial vegetation in the habitat, the darker the shells.
Also during the Pleistocene, when temperatures were 5–10°C lower than today, the shells of the coastal plain were as pale as recent ones are. The distribution of a snail-predator, Gerbillus allenbyi , which is restricted to sandy biotopes where perennial bushes occur, and which was absent from Israel during the Pleistocene, could perhaps explain the distribution of X. vestalis morphs, both today and in the past.  相似文献
