When [l-14C]-malonate was supplied to discs cut from matureleaves of Coffea arabica, 14CO2 was released (approximately12% of the total CO2 respired) and organic acids of the Krebscycle, uronic acids, sugars and amino acids became radioactive.There was no incorporation of MC into either lipids or phenoliccompounds. The formation of glucose from malonate has not beenobserved in other studies with plant tissues. The synthesisof labelled glucose together with an active pentose phosphatepathway that is stimulated by malonate explains the accumulationof radioactive phosphogluconate in the leaf discs. 1. The southern monarch, Danaus erippus, uses mainly Asclepias curassavica as its host in the Neotropics, a plant species bearing articulated anastomosing laticifers. When artificially severed, A. curassavica has been shown to release significantly less latex than other Asclepias species. 2. The present study tested the hypothesis that sabotaging behaviour changes during the ontogeny of D. erippus and recorded latex outflow of A. curassavica during sabotaging and feeding. Larvae displayed vein‐cutting behaviour, which was initially observed in the second instar, became more pronounced in the third and fourth instars, and less frequent in the fifth instar. When present, latex outflow was never more than 1 µl at a time during either vein cutting or feeding, regardless of the instar. 3. Mandibular and midrib morphometrics revealed that larvae selected thicker midrib sites for severing as instars progressed; however, no correlation between mandibular size and midrib size severed was found within instars. 4. Costs of sabotaging behaviour and the effects of A. curassavica latex outflow on D. erippus larvae were also examined. Sabotaging behaviour did not incur growth costs for larvae, and only latex exudation volumes at least 10‐fold greater than those observed due to D. erippus sabotaging or feeding, caused significantly higher larval mortality than controls. 5. Since latex outflow is minimal or non‐existent in A. curassavica, sabotaging behaviour in D. erippus is mostly limited by morphological constraints and is probably driven by chemical stimulants rather than latex defence. In turn, latex does not constitute a major defence of A. curassavica against D. erippus.  相似文献

The present study first addressed the question of whether developmental time (DT) and viability (VT) vary clinally along latitudinal and altitudinal gradients in Drosophila buzzatii, an autochthonous specialist and the generalist invasive Drosophila melanogaster. Coincident and positive altitudinal clines across species and, direct and inverse latitudinal clines were observed for DT in D. melanogaster and D. buzzatii, respectively. Opposing latitudinal and altitudinal clines were detected for VT only in D. melanogaster. The patterns observed along altitudinal gradients prompted us to investigate whether flies living at lowland and highland environments may respond differentially to thermal treatments consisting of regimes of constant and alternating temperatures. Flies reared at higher mean temperature developed faster than at lower mean temperature in both species. By contrast, the response in VT differed greatly between species. Highland D. melanogaster were more viable than lowland regardless the treatment, whereas, in D. buzzatii, highland flies were more viable than lowland in alternating thermal regimes and the reverse was true in treatments of constant temperature. Iberus gualtieranus is a species complex of land snails that is endemic to the Iberian Peninsula. The species taxonomy of the group is based merely on the basis of shell morphology, but validity of the existing taxonomy is uncertain. Using mitochondrial DNA (mtDNA) data (cytochrome oxidase I and 16S rRNA sequences) we were able to validate the observed phylogenetic taxa within the I. gualtieranus s.l. complex by means of the analysis of specimens of the different morphospecies, together with the study of topotypes. Strong incongruences were obtained between morphology and molecular data. The Iberus alonensis morphospecies comprised several genetically divergent but morphologically cryptic lineages. Considering (1) the allopatric distribution of the operational taxonomic units (OTUs), (2) the morphological differentiation, (3) the possible occurrence of hybridization among the different lineages, and (4) the strong differentiation of the mtDNA phylogroups, we suggest the main lineages obtained, for the time being, may be treated as evolutionary species. The robust phylogenetic reconstruction obtained allows us to consider I. alonensis s.s., Iberus campesinus, Iberus carthaginiensis, and Iberus gualtieranus s.s. as valid species. Two additional unnominated taxa of the alonensis shell type have also been identified. The present study aimed to infer evolutionary scenarios for Vipera latastei and Vipera monticola in the Iberian Peninsula and the Maghreb through the identification of spatial patterns in morphological character variation and biogeographic patterns in morphological variability distribution. Ten morphological traits from 630 vipers were analysed with geostatistic and ecological niche modelling in a geographical information system. Interpolation by Kriging was used to generate surfaces of morphological variation, which were combined with spatial principal components analysis (SPCA). Putative morphological differentiated groups generated by SPCA maps were tested with discriminant function analysis (DFA). Maximum entropy modelling and nine environmental variables were used to identify factors limiting the distribution of groups and areas for their potential occurrence. Groups supported by DFA were: Western Iberia, Eastern Iberia, Rif plus Middle Atlas, Algeria, and High Atlas. Their distribution is influenced by common environmental factors such as precipitation. Areas of probable sympatry between Iberian groups matched the morphological clines observed by geostatistics tools. Geographic variation patterns in V. latastei‐monticola are probably due to vicariant separation of Iberian and African populations during the opening of the Strait of Gibraltar, and population refugia during the Quaternary glaciations with secondary contact. The taxonomic status of northern Morocco and Algerian groups should be further investigated. Abstract: New multituberculate mammals from the Hauterivian/Barremian transition of Europe are described. They were found in the late Hauterivian‐early Barremian fossiliferous locality of La Cantalera (Josa, Teruel, Spain), one of the Early Cretaceous sites in the Aragonese branch of the Iberian Ranges, in northeastern Iberia. The fossils have been assigned to at least three taxa on the basis of nine isolated teeth: a new pinheirodontid taxon, Cantalera abadi gen. et sp. nov.; a representative of the eobaatarid Eobaatar; a taxon described as Plagiaulacidae or Eobaataridae gen. et sp. indet.; and other as Plagiaulacida indet. These fossils have increased the resolution of European Early Cretaceous multituberculate mammalian biostratigraphy and palaeobiogeography: the oldest representative of Eobaatar is described here; a taxon is assigned to ?Plagiaulacidae, in which case it would be the first of this family in the Iberian Peninsula; and the discovery of a new late Hauterivian pinheirodontid taxon demonstrates greater biodiversity and a wider distribution for these multituberculates than was previously known. The mutituberculate fauna of La Cantalera consists of endemic taxa (Pinheirodontidae), which were restricted to what is now Western Europe, and others (Eobaataridae) which have also been described in Asia. Consistent with the Iberian record of late Barremian gobiconodontid mammals, the presence of Eobaatar in Iberia with representatives from the late Hauterivian to late Barremian, as well as in the Aptian or Albian of Mongolia, indicates that faunal exchanges between Europe and Asia could have existed for most of the Early Cretaceous, either sporadically or constantly.  相似文献

The genetic structure and morphometric differentiation of mangrove crab Perisesarma guttatum populations were examined among shelf connected locations along a latitudinal gradient on the East African coast. Over 2200 specimens were sampled from 23 mangrove sites for geometric morphometrics analysis. Population genetic analyses of mitochondrial cytochrome c oxidase subunit I (COI) DNA sequences were used to evaluate connectivity among populations. A total of 73 haplotypes were detected, and almost no haplotypes were found in common between two highly supported phylogeographic clades: southern Mozambique (Inhaca Island and Maputo Bay) and a northern clade that included north Mozambique, Tanzania and Kenya. These two clades were identified based on the species' populations pairwise genetic differentiation and geographical location. ΦST values were considerably high between the two clades, indicating the presence of significant population genetic structure between Kenya and South Mozambique. However, each clade was composed of genetically similar populations along the latitudinal gradient, and no significant population structure was found within each clade because the Φst values were not significant. The morphometric analysis corroborated the division into two clades (i.e. Inhaca Island/Maputo Bay and northern populations) and also detected less shape variation among populations that were few kilometres apart. The significant spatial genetic structuring between the southern and the northern populations of P. guttatum along the geographic gradient under study, combined with morphological differences, suggests that these populations may be considered as cryptic species. © 2010 The Linnean Society of London, Biological Journal of the Linnean Society, 2010, 99 , 28–46.  相似文献

Soil salinity restricts plant growth and productivity. Na+ represents the major ion causing toxicity because it competes with K+ for binding sites at the plasma membrane. Inoculation with arbuscular mycorrhizal fungi (AMF) can alleviate salt stress in the host plant through several mechanisms. These may include ion selection during the fungal uptake of nutrients from the soil or during transfer to the host plant. AM benefits could be enhanced when native AMF isolates are used. Thus, we investigated whether native AMF isolated from an area with problems of salinity and desertification can help maize plants to overcome the negative effects of salinity stress better than non‐AM plants or plants inoculated with non‐native AMF. Results showed that plants inoculated with two out the three native AMF had the highest shoot dry biomass at all salinity levels. Plants inoculated with the three native AMF showed significant increase of K+ and reduced Na+ accumulation as compared to non‐mycorrhizal plants, concomitantly with higher K+/Na+ ratios in their tissues. For the first time, these effects have been correlated with regulation of ZmAKT2, ZmSOS1 and ZmSKOR genes expression in the roots of maize, contributing to K+ and Na+ homeostasis in plants colonized by native AMF.  相似文献

Chronic Chagasic patient immunoglobulins (CChP-IgGs) recognize an acidic amino acid cluster at the second extracellular loop (el2) of cardiac M2-muscarinic acetylcholine receptors (M2AChRs). These residues correspond to a common binding site for various allosteric agents. We characterized the nature of the M2AChR/CChP-IgG interaction in functional and radioligand binding experiments applying the same mainstream strategies previously used for the characterization of other allosteric agents. Dose-response curves of acetylcholine effect on heart rate were constructed with data from isolated heart experiments in the presence of CChP or normal blood donor (NBD) sera. In these experiments, CChP sera but not NBD sera increased the efficacy of agonist action by augmenting the onset of bradyarrhythmias and inducing a Hill slope of 2.5. This effect was blocked by gallamine, an M2AChR allosteric antagonist. Correspondingly, CChP-IgGs increased acetylcholine affinity twofold and showed negative cooperativity for [3H]-N-methyl scopolamine ([3H]-NMS) in allosterism binding assays. A peptide corresponding to the M2AChR-el2 blocked this effect. Furthermore, dissociation assays showed that the effect of gallamine on the [3H]-NMS off-rate was reverted by CChP-IgGs. Finally, concentration-effect curves for the allosteric delay of W84 on [3H]-NMS dissociation right shifted from an IC50 of 33 nmol/L to 78 nmol/L, 992 nmol/L, and 1670 nmol/L in the presence of 6.7 × 10? 8, 1.33 × 10? 7, and 2.0 × 10? 7 mol/L of anti-el2 affinity-purified CChP-IgGs. Taken together, these findings confirmed a competitive interplay of these ligands at the common allosteric site and revealed the novel allosteric nature of the interaction of CChP-IgGs at the M2AChRs as a positive cooperativity effect on acetylcholine action.  相似文献
