ABSTRACT. The responses of male Ceratitis capitata (Wiedemann) (Diptera; Tephritidae) to trimediure were investigated in a wind tunnel of novel design with disposable plastic walls. Trimediure was presented to the flies either in the form of a discrete 'plume' or uniformly dispersed in the air passing through the tunnel. The flies showed an increase in flight activity in response to the two forms of stimulation and performed a zig-zag anemotactic flight in response to the plume, reminiscent of the behaviour of moths responding to a sex pheromone. They performed a straight, upwind, anemotactic response to the uniformly dispersed odour. The design of wind tunnel used in these tests was found to have several advantages: air flow and turbulence can be easily varied and the plastic walls can be changed to eliminate contamination problems.  相似文献

African freshwater snails of the genus Bulinus act as intermediate hosts for schistosomes, trematode parasites responsible for medical and veterinary forms of schistosomiasis. The relationship between these snails and their parasites is an intricate one, with particular species of snail susceptible to infection only by certain species of schistosome. In common with other self-fertile hermaphrodite gastropods, Bulinus consists of a number of closely related species complexes with restricted gene flow between populations of each taxon. Consequently, despite their medical and veterinary importance as intermediate hosts, unambiguous identification and differentiation of planorbid snails such as these remains problematic, often confounding attempts to define the distribution and evolutionary relationships of conchologically similar taxa. Here we consider how morphological methods of discrimination can be used in conjunction with molecular based approaches to improve snail identification, thereby achieving a better understanding of the epidemiology of schistosomiasis. Data are presented from Central and East African taxa which illustrate how PCR-based methods have begun to be used in combination with traditional analyses in an integrated approach to characterize the genus Bulinus , specifically the B. forskalii species group. Particular emphasis is given to the analyses of Random Amplified Polymorphic DNA (RAPDs) and the mitochondrial gene cytochrome oxidase I (COI).  相似文献

The taxonomic affinity of Halosphaeria cucullata to Halosphaeria is reassessed based on a recent collection of this fungus. Halosphaeria cucullata is characterized by immersed, darkly coloured ascomata, clavate asci which deliquesce very early in development, and cylindrical ascospores with or without a polar cap-like appendage at one end. In a phylogenetic analysis of the LSU rDNA sequences from members of the Halosphaeriaceae, H. cucullata did not form a monophyletic clade with H. appendiculata , the type species of the genus. These results suggest that H. cucullata should not be included in Halosphaeria . Spatial patterns of spread were compared between strains of Bean yellow mosaic virus (BYMV) that differ in causing systemic necrotic (hypersensitive) or non‐necrotic symptoms in narrow‐leafed lupin (Lupinus angustifolius). Both types of BYMV were spread naturally by aphids from adjacent infected pasture into a large lupin block (‘natural spread site’), or from clover plants introduced as virus sources into two field experiments with lupin. Cumulative spatial data for plants with disease symptoms from a range of times in the growing period were assessed using Spatial Analysis by Distance IndicEs (SADIE). At the‘natural spread site’, with non‐necrotic BYMV, the extent of clustering of plants with symptoms increased gradually over time, while with necrotic BYMV there was less clustering and no increase over time. In both experiments, for the type of BYMV that was introduced into a plot, there was a gradual increase in clustering, but with this being greater with non‐necrotic BYMV. In the second experiment, there was also significant clustering of plants with symptoms of non‐necrotic BYMV in plots without introduced non‐necrotic foci but not for necrotic BYMV in plots without introduced necrotic foci. When clustering data for plants with newly recorded symptoms was tested for spatial association between successive assessment dates, association was positive for both BYMV types though stronger for the non‐necrotic type, declining as the temporal lag increased. Generally, association was strongest for assessments 2–3 wk apart, corresponding approximately to the period for BYMV to move systemically in plants and for obvious symptoms to appear in shoot tips. Contour maps for local association between dates showed that the strongest spatial associations were from coincidence of infection gaps rather than infection patches. The combination of information from clustering and association analysis showed that spread of non‐necrotic BYMV is less diffuse, with considerably more localised infection surrounding the infection sources. This work demonstrates how spatial virus spread can be diminished when hypersensitive (necrotic) resistance is deployed, and the limitations associated with employing hypersensitivity that is strain specific.  相似文献

Previous studies have indicated that the common European pipistrelle bat ( Pipistrellus pipistrellus ) comprises two cryptic species, P. pipistrellus and Pipistrellus pygmaeus , which differ in echolocation call frequency and mitochondrial DNA sequence. However, levels of divergence based on nuclear markers have not been examined, and hence the potential for male-mediated gene flow between the species cannot be discounted. Moreover, little is known about population structure and migration patterns in either species. Here, we describe the use of microsatellites to investigate nuclear DNA differentiation between, and the pattern of population genetic structure within, the two cryptic pipistrelle species. In total, 1300 individuals from 82 maternity colonies were sampled across the British Isles and Continental Europe. We show, using multivariate analyses, that colonies of the same species are generally genetically more similar to each other than to those from the other species regardless of geographical location. Our findings support the hypothesis that the species are reproductively isolated. Significant patterns of genetic isolation by distance were identified in both species, indicating that mating may occur before any long-distance autumnal migration. The presence of a sea channel does not confer higher levels of genetic differentiation among colonies over and above distance alone in either species. Differences in genetic population structure were identified between the species, with P. pipistrellus showing a wider range of levels of genetic differentiation among colonies and a stronger relationship between genetic and geographical distance than P. pygmaeus . Using open-top chambers, four prominent species (Lolium perenne,Cynosurus cristatus, Holcus lanatusandAgrostis capillaris) ofIrish neutral grasslands were grown at ambient and elevated(700 µmol mol-1) atmospheric CO2for a period of 8 months.The effects of interspecific competition on plant responsesto CO2enrichment were investigated by growing the species ina four-species mixture. The results indicate that the speciesdiffer in their ability to respond to elevated CO2. CO2-enrichmenthad the largest effect on the biomass production ofH. lanatus,but substantial stimulations in biomass production were alsofound for the other three species. The CO2-stimulation of biomassproduction forH. lanatuswas accompanied by increased tillering.In addition, reductions in specific leaf area were found forall species. Exposure to elevated CO2increased the communitybiomass of the four-species mixture. This increase can be mainlyattributed to a significant increase in the biomass ofH. lanatusatelevated CO2. No statistically-significant changes in speciescomposition of community biomass were found. However,H. lanatusdidincrease its share of community biomass at each of the harvests,with the other three species, mainlyL. perenne, suffering lossesin their shares at elevated CO2. IT seems to have been widely assumed that autonomic neurones respond to axonal damage much as do somatic motoneurones. From an extensive series of experiments on the albino rat, however, we have obtained evidence that autonomic preganglionic neurones are much more seriously affected. Thus, when the cervical vagus on one side was either crushed or cut, we consistently found in more than 100 animals that changes in the cells of the dorsal motor nucleus of the vagus were more severe and more protracted than in those of the hypoglossal nucleus when its nerve was similarly treated. The changes are seen particularly well in histochemical preparations designed to show the distribution of various metabolic enzymes. In the work described here attention was concentrated on alterations in the cholinesterases and in acid phosphatase.  相似文献

Thin sections of diseased raspberry (Rubus idaeus) were examined by electron microscopy. Plants of the cv. Baumforth's B and of an aphid (Amphorophora rubi)-resistant breeding selection (6820/54), both infected with raspberry vein chlorosis virus (RVCV) but not with other detectable viruses, contained large bacilliform particles c. 430 × 65 nm. Particles occurred in the cytoplasm and perinuclear space of a small proportion of xylem parenchyma cells. They had an inner core c. 25–30 nm in diameter with cross-banding of periodicity 4·5 nm, and were bounded by an outer membrane. They are probably the particles of RVCV. Plants of cv. Mailing Jewel and of a selection (M14) both showing symptoms of raspberry mosaic (veinbanding) disease contained smaller bacilliform particles c. 125 × 30 nm, which occurred singly or in clusters in the cytoplasm of a small proportion of vascular parenchyma cells. It is not known which, if any, of the viruses associated with raspberry mosaic are represented by the particles.  相似文献

The trapping of alate aphids in emergence cages each 1 yd2 (0–83 m2) over cereal crops from mid-June to the end of July, 1964 to 1971, always revealed colonies of cereal aphids within the crop. Four species, Sitobion avenae, S. fragariae, Metopolophium dirhodum and Rhopalosiphum padi occurred every year in different proportions. Alate aphids from winter wheat were most numerous in 1968 and fewest in 1967. Alatae developed slightly earlier in cages than in the field and peak catches were a few days earlier than in a nearby 12-2 m suction trap. Cereal aphid colonies were adversely affected by bad weather in May, e.g. in 1969, and by predators. Coccinellidae (chiefly Propylea 14-punctata) were the dominant predators in 1971 and 1968, Syrphidae in 1966, 1971 and 1968 and Chrysopidae in 1970. Parasites belonging mainly to the genus Aphidius were numerous every year. When hyperparasites such as Asaphes vulgaris, Lygocerus sp., Conostigmus sp. and Phaenoglyphis sp. were abundant as in 1967, they affected numbers of aphids in the current year and increased them in the following year (1968), possibly by hindering early, heavy parasitism. Hyperparasites could have an important influence in fluctuations of cereal aphid populations from year to year. Aphids of one species or another are always present in cereal crops in sufficient numbers during the summer months to provide copious quantities of honey dew, and this is unlikely to be a limiting factor in the biology of the wheat bulb fly, Leptohylemyia coarctata.  相似文献
