Light and electron microscopy was used to study the ultrastructural effects of June Yellows (JY) in leaves of strawberry. Four cultivars of strawberry, affected to different extents by JY, were compared with healthy (JY-free) cv. Cambridge Favourite and with strawberry infected with strawberry crinkle rhabdovirus, Fragaria vesca infected with strawberry mottle virus (SMotV), raspberry and black currant showing virus-induced yellowing and with strawberry and raspberry showing chaemeric yellow sectors in the leaves. Except for isometric virus-like particles detected in SMotV-infected F. vesca, no virus-like particles or structures of other pathogenic agents were found in any of the tissues examined. Leaf cells of JY-affected strawberry showed severe disruption of chloroplasts and plasmalemma, whorls of membranous vesicles and decreased vacuole size. The extent and severity of these abnormalities increased with increased severity of JY symptoms but, even in leaves with mild JY symptoms, chloroplast abnormalities were obvious. In the most severely affected leaves, the cells lacked discrete vacuoles and extensive hypertrophy was seen in other organelles such as nuclei and mitochondria. Few, if any, ultrastructural abnormalities were observed in virus-infected strawberry or F. vesca, or in chaemeric leaves of strawberry and raspberry. By contrast, in raspberry and black currant with yellowed leaves caused by virus infection, the cells showed enlarged chloroplasts, decreased vacuole size and vesicle formation. However, chloroplast enlargement and disruption in this material seemed due to increased size of starch grains which were largely absent from swollen chloroplasts of JY-affected strawberry. The ultrastructural abnormalities observed in JY-affected strawberry are, therefore, not inconsistent with the possibility that a pathogenic agent may be involved in the condition.  相似文献

Changes in the leaf solute composition of a number of perennialTriticeae in response to salinity are described. There was variationboth within and between species in the extent of Na and Cl accumulationin the leaves. In almost all cases leaf nitrate concentrationsdecreased substantially and orthophosphate concentrations increasedin response to salinity. There was no overall decrease in arange of soluble nitrogen-containing compounds nor in totalnitrogen. In Thinopyrum scirpeum there was an inverse relationshipbetween proline and glycinebetaine contents of different tissues,with proline levels higher in older leaves. Abstract. 1. Euedaphic collembola alter their soil distribution in response to above‐ground aphid herbivory of Poa annua L. Graminae, a host grass. 2. Two mechanisms potentially underpin this effect. Carbon‐rich aphid honeydew falling onto the soil surface may affect mycophagous collembola; alternatively aphid‐induced changes in root biomass may be necessary to produce changes in collembola abundance. 3. When compared to a plant‐only control, aphid herbivory increased the number of collembola in the top 5 cm of soil, reduced both foliar and root biomass, and increased shoot/root ratio. Honeydew addition had no effect on collembola numbers or any recorded host‐plant parameter. 4. Honeydew deposition is not responsible for the increased numbers of collembola found in the upper soil after aphid herbivory; aphid‐induced reductions in root biomass may be the most important factor explaining knock‐on effects of aphid herbivory on soil fauna.  相似文献

Abstract 1. The leaf‐cutting ants practise an advanced system of mycophagy where they grow a fungus as a food source. As a consequence of parasite threats to their crops, they have evolved a system of morphological, behavioural, and chemical defences, particularly against fungal pathogens (mycopathogens). 2. Specific fungal diseases of the leaf‐cutting ants themselves have not been described, possibly because broad spectrum anti‐fungal defences against mycopathogens have reduced their susceptibility to entomopathogens. 3. Using morphological and molecular tools, the present study documents three rare infection events of Acromyrmex and Atta leaf‐cutting ants by Ophiocordyceps fungi, agenus of entomopathogens that is normally highly specific in its host choice. 4. As leaf‐cutting ants have been intensively studied, the absence of prior records of Ophiocordyceps suggests that these infections may be a novel event and that switching from one host to another is possible. To test the likelihood of this hypothesis, host switching was experimentally induced, and successfully achieved, among five distinct genera of ants, one of which was in a different sub‐family than the leaf‐cutter ants. 5. Given the substantial differences among the five host ants, the ability of Ophiocordyceps to shift between such distant hosts is remarkable; the results are discussed in the context of ant ecological immunology and fungal invasion strategies.  相似文献

Abstract: Condensed tannins (CT) can reduce digestibility of forages for white-tailed deer (Odocoileus virginianus), potentially confounding estimates of diet quality and nutritional carrying capacity. We collected 143 spring and 142 summer samples of 8 important deer forage species from 22 properties in Mississippi, USA, and tested for CT content using a modified butanol-HCl assay. Three species (partridge pea [Chamaecrista fasciculata], southern dewberry [Rubus trivialis], and roundleaf greenbrier [Smilax rotundifolia]) contained CT, ranging from 0.11% to 6.46% dry weight. Summer CT concentration was greater than in spring for 2 species. We ranked soil samples from least to most fertile using 8 chemical characteristics and found a positive correlation between fertility and CT concentration for 1 species and no correlation for 2 species. We tested effects of CT concentration on in vitro dry matter disappearance (IVDMD) and in vitro protein digestibility using samples of partridge pea and roundleaf greenbrier and rumen fluid from 3 free-ranging deer. Average IVDMD was reduced 1.9% for each 1% increase in CT concentration. In vitro protein digestibility was reduced 2.5% for each 1% increase in CT concentration. Assuming that our methods reflect the effects of CT on in vivo digestibility, maximum loss of available crude protein (CP) in our samples was 3.0 g/100 g dry-weight forage, and only 13 of the 112 CT-containing forage samples (12%) would have decreased available CP by >1 g/100 g dry-weight forage. Deer consuming equal portions of sampled forages would lose <1% of dietary CP to CT. Comparisons of foraging area quality using crude protein estimates should be unaffected by CT under reasonable restrictions of similar habitat types, soil fertility, and time. Given the ability of deer to forage selectively and the abundance of alternative forages in Mississippi, the potential for CT to substantially affect spring or summer diet quality of deer appears minimal.  相似文献

Abstract To characterize the structure and species density of termite assemblages in tropical forests, we developed a sampling protocol based on a single large quadrat (belt transect) that standardises sampling effort and area. Emphasizing the estimation of species richness, Roisin and Leponce (2004, Austral Ecology, 29 : 637–46) recommend sampling numerous small quadrats, and varying the number of quadrats depending on local species density. However, their method would result in the loss of standardisation and speed, taking up to four times longer to collect and process the samples. We show that even for a subtropical forest with low species density, our protocol produces robust results.  相似文献
