Pseudopregnancy was induced in rabbits by injection of 50 i.u. hCG into the lateral ear vein. Blood was collected on Days 0, 1, 2 and 6 after hCG from the lateral ear vein and, in some studies, from the ovarian vein as well. Ovaries were collected on Days 0, 1, 2 or 6 after hCG injection, and follicles (greater than 0.8 mm diam.) were obtained by microdissection. Concentrations of oestradiol-17 beta, testosterone and progesterone in blood and follicular homogenates were measured by RIA. Follicular steroidogenesis was increased significantly on Day 6 after ovulation, at the time when the corpus luteum transforms into an oestrogen-dependent tissue. The ability of developing post-ovulatory follicles to secrete steroids in vitro in response to a gonadotrophic stimulus also increased over this same time interval. Follicular oestradiol production on Days 1 and 2 of pseudopregnancy may synergize with the post-ovulatory secretion of FSH to promote further follicular growth in the pseudopregnant rabbit. 相似文献

It proved possible to determine the levels of Sr-90 in the opercular bones of individual pike, Esox Iucius , and in pooled samples of bones from perch, Perca fluviatilis . Results from both species from Windermere demonstrated that Sr-90 levels rose from below the detection limits in the 1940s to a peak in the 1960s, followed by a decline in the subsequent two decades. This decline was slower than would have been expected from the decline in northern hemisphere Sr-90 fallout, indicating the likelihood of recycling within the environment. Sr-90 levels were consistently lower in pike than in perch, their main prey fish. Thus, there is no concentration of Sr-90 up this part of the aquatic food chain. Tracking Sr-90 in bones taken in successive years from ages 3 to 8 for a single cohort of pike showed that the quantity of Sr-90 was closely related to opercular bone (and hence fish) weight. Hurricane Danny resulted in the rapid deposition of 10cm of oxidized, acidic sediment in the Contrary Creek arm of Lake Anna, Virginia. Several biological and geochemical parameters were monitored with time to ascertain how long it took the newly-deposited lake sediments to attain the anaerobic, circumneutral, actively sulfate-reducing state normally observed in this portion of the lake. The sediment platinum-electrode potential dropped from 350 mV to 100 mV within the first week after the storm. The pH of the pore water increased from 4.5 to 5.8 within three weeks, and titratable alkalinity was detected within two weeks and three weeks at 3 cm and 1 cm depths, respectively. Accumulation of reduced products of sulfate reduction (acid volatile sulfide) began by three to four weeks after the storm event. Both methanogens and sulfate reducers were present in high and approximately equal numbers in the freshly deposited material. The rapid neutralization of the acidity in the fresh sediment prior to the onset of sulfate reduction suggests that reactions other than sulfate reduction caused the initial increase in pH and alkalinity in this system. 相似文献

Dissecting the link between genetic variation and adaptive phenotypes provides outstanding opportunities to understand fundamental evolutionary processes. Here, we use a museomics approach to investigate the genetic basis and evolution of winter coat coloration morphs in least weasels (Mustela nivalis), a repeated adaptation for camouflage in mammals with seasonal pelage color moults across regions with varying winter snow. Whole-genome sequence data were obtained from biological collections and mapped onto a newly assembled reference genome for the species. Sampling represented two replicate transition zones between nivalis and vulgaris coloration morphs in Europe, which typically develop white or brown winter coats, respectively. Population analyses showed that the morph distribution across transition zones is not a by-product of historical structure. Association scans linked a 200-kb genomic region to coloration morph, which was validated by genotyping museum specimens from intermorph experimental crosses. Genotyping the wild populations narrowed down the association to pigmentation gene MC1R and pinpointed a candidate amino acid change cosegregating with coloration morph. This polymorphism replaces an ancestral leucine residue by lysine at the start of the first extracellular loop of the protein in the vulgaris morph. A selective sweep signature overlapped the association region in vulgaris, suggesting that past adaptation favored winter-brown morphs and can anchor future adaptive responses to decreasing winter snow. Using biological collections as valuable resources to study natural adaptations, our study showed a new evolutionary route generating winter color variation in mammals and that seasonal camouflage can be modulated by changes at single key genes. 相似文献

A novel and patented procedure is described for the sonochemical fabrication of a new class of microelectrode array based sensor with electrode element populations of up to 2 x 10(5) cm(-2). For some years it has been accepted that microelectrode arrays offer an attractive route for lowering minimum limits of detection and imparting stir (convectional mass transport) independence to sensor responses; despite this no commercial biosensors, to date, have employed microelectrode arrays, largely due to the cost of conventional fabrication routes that have not proved commercially viable for disposable devices. Biosensors formed by our sonochemical approach offer unrivalled sensitivity and impart stir independence to sensor responses. This format lends itself for mass fabrication due to the simplicity and inexpensiveness of the approach; in the first instance impedimetric and amperometric sensors are reported for glucose as model systems. Sensors already developed for ethanol, oxalate and a number of pesticide determinations will be reported in subsequent publications. 相似文献

Most organisms form Cys-tRNA(Cys), an essential component for protein synthesis, through the action of cysteinyl-tRNA synthetase (CysRS). However, the genomes of Methanocaldococcus jannaschii, Methanothermobacter thermautotrophicus, and Methanopyrus kandleri do not contain a recognizable cysS gene encoding CysRS. It was reported that M. jannaschii prolyl-tRNA synthetase (C. Stathopoulos, T. Li, R. Longman, U. C. Vothknecht, H. D. Becker, M. Ibba, and D. S?ll, Science 287:479-482, 2000; R. S. Lipman, K. R. Sowers, and Y. M. Hou, Biochemistry 39:7792-7798, 2000) or the M. jannaschii MJ1477 protein (C. Fabrega, M. A. Farrow, B. Mukhopadhyay, V. de Crécy-Lagard, A. R. Ortiz, and P. Schimmel, Nature 411:110-114, 2001) provides the "missing" CysRS activity for in vivo Cys-tRNA(Cys) formation. These conclusions were supported by complementation of temperature-sensitive Escherichia coli cysS(Ts) strain UQ818 with archaeal proS genes (encoding prolyl-tRNA synthetase) or with the Deinococcus radiodurans DR0705 gene, the ortholog of the MJ1477 gene. Here we show that E. coli UQ818 harbors a mutation (V27E) in CysRS; the largest differences compared to the wild-type enzyme are a fourfold increase in the K(m) for cysteine and a ninefold reduction in the k(cat) for ATP. While transformants of E. coli UQ818 with archaeal and bacterial cysS genes grew at a nonpermissive temperature, growth was also supported by elevated intracellular cysteine levels, e.g., by transformation with an E. coli cysE allele (encoding serine acetyltransferase) or by the addition of cysteine to the culture medium. An E. coli cysS deletion strain permitted a stringent complementation test; growth could be supported only by archaeal or bacterial cysS genes and not by archaeal proS genes or the D. radiodurans DR0705 gene. Construction of a D. radiodurans DR0705 deletion strain showed this gene to be dispensable. However, attempts to delete D. radiodurans cysS failed, suggesting that this is an essential Deinococcus gene. These results imply that it is not established that proS or MJ1477 gene products catalyze Cys-tRNA(Cys) synthesis in M. jannaschii. Thus, the mechanism of Cys-tRNA(Cys) formation in M. jannaschii still remains to be discovered. 相似文献

Large‐scale restoration efforts are underway globally to mitigate the impact of decades of land degradation by returning functional and biodiverse ecosystems. Revegetation is a heavily relied upon restoration intervention, and one that is expected to result in associated biodiversity returns. However, the outcome of such restoration interventions rarely considers recovery to the soil microbiome, a mega‐diverse and functionally important ecosystem component. Here we examine the archaeal component of the soil microbiome and track community change after a decade of eucalypt woodland restoration in southern Australia. We employed DNA metabarcoding to show that archaeal community composition, richness, and diversity shifted significantly, and towards a restored state 10 years after the restoration intervention. Changes in soil pH and nitrate associated with changes to the archaeal community, potentially relating to the pH responsive properties and close relationship with the nitrogen cycle of some archaea. Our study helps shed light on archaeal community dynamics, as no other study has used DNA metabarcoding to study archaeal responses across a restoration chronosequence. Our results provide great promise for the development of molecular monitoring of the soil microbiome as a future restoration monitoring tool. 相似文献
