These studies detail the altered structure-function relationships caused by oxidation of surfactant protein B (SP-B), a mode of damage thought to be important in acute respiratory distress syndrome (ARDS), a common and frequently fatal condition. An 18-residue fragment comprising the N-terminal helix of SP-B was investigated in oxidized and unmodified forms by solution and solid-state nuclear magnetic resonance (NMR), circular dichroism (CD), and molecular dynamics (MD) simulation. Taken together, the results indicate that tryptophan oxidation causes substantial disruptions in helical structure and lipid interactions. The structural modifications induced by tryptophan oxidation were severe, with a reduction in helical extent from approximately three helical turns to, at most, one turn, and were observed in a variety of solvent environments, including sodium dodecyl sulfate (SDS) micelles, dodecyl phosphocholine (DPC) micelles, and a 40% hexafluoro-2-propanol (HFIP) aqueous solution. The unmodified peptide takes on an orientation within lipid bilayers that is tilted approximately 30° away from an in-plane position. Tryptophan oxidation causes significant modifications to the peptide-lipid interactions, and the peptide likely shifts to a more in-plane orientation within the lipids. Interestingly, the character of the disruptions to peptide-lipid interactions caused by tryptophan oxidation was highly dependent on the charge of the lipid headgroup.  相似文献

We have developed a quantitative scintillation proximity assay (SPA) that reproduces the Raf/MEK/ERK signal transduction pathway. The components of this assay include human cRaf1, MEK1, and ERK2 and a biotinylated peptide substrate for ERK2. cRaf1 was expressed as a his-tagged protein in insect cells in an active form. MEK1 and ERK2 were expressed in Escherichia coli as glutathione S-transferase (GST)-fusion proteins in their inactive forms. ERK2 was removed from the GST portion of the fusion protein by cleavage with thrombin protease. When the purified components are incubated together, cRaf-1 phosphorylates and activates MEK1, MEK1 phosphorylates and activates ERK2, and ERK2 phosphorylates the peptide, biotin-AAATGPLSPGPFA. Phosphorylation of the peptide using [gamma-33P]ATP is detected following binding to streptavidin-coated SPA beads. The assay detects inhibitors of cRaf1, MEK1, or ERK2, and has been used to screen large numbers of compounds. The specific target of inhibition was subsequently identified with secondary assays described herein.  相似文献

Summary   Exotic predators are considered pests to wildlife and agriculture, requiring predator-control programs. Effective monitoring of predator-control operations is essential to justify their considerable cost, but often impossible in practice. The difficulties are especially severe if the target species is small and wide-ranging, and the area to be protected is inaccessible and/or extensive. A convenient model predator of this type, the feral Ferret ( Mustela furo ), is subject to control on farmland in New Zealand. We monitored the distribution of Ferrets over 2400 ha in the central North Island, before and throughout a standard control operation by professional trappers. We used 24 units of a new automated monitoring device, the Scentinel, set in a grid at 1 per 100 ha. Over 11 weeks (11 February to 29 April 2005, 1718 trap nights), we recorded 1559 visits by small mammals, including 198 by Ferrets. By the end of the 4th week, Ferrets had been detected at 17 of 24 sites. Removal of Ferrets from the study area by contractors began during the 5th week, and was reflected in significant declines in the number of Ferret visits recorded per day ( P  = 0.008) and the number of sites visited ( P  = 0.021). Analysis of our extensive repeat-survey data by site-occupancy methods confirmed these trends in greater detail, while also allowing for variation in detectability.  相似文献

Early embryonic swimming is widespread among marine invertebrates, but quantitative information about swimming behaviors is scarce. Swimming may affect encounters with predators, positioning in the water column, and nutrient absorption. Measured rates and patterns of swimming and sinking for blastulae of four eastern Pacific echinoid species show that sinking speeds equal or exceed swimming speeds. Swimming speed scaled negatively with embryo size, though sinking speed did not scale with size. Analysis of swimming paths of Strongylocentrotus franciscanus revealed a temperature dependency in swimming pattern that affected speed of upward movement. Sinking speeds were significantly greater at 10 degrees C than at 14 degrees C for blastulae of all four species examined. In Dendraster excentricus, killing the blastulae annulled this temperature effect, indicating an active density regulation by these embryos. Finally, measurements of particle velocities around sinking and swimming D. excentricus blastulae show that swimming creates a more localized disturbance than sinking. Embryonic swimming may therefore decrease rather than increase encounters with pelagic predators. Results from subsequent experiments in which embryos were reared in low-oxygen environments suggest that any oxygen-absorption advantages of swimming have little, if any, effect on the development of D. excentricus embryos.  相似文献

Phenylketonuria (PKU) is a metabolic disorder caused by phenylalanine hydroxylase deficiency leading to increased levels of phenylalanine in the brain. Hyperactivity is reportedly induced by a high level of orexin A, and therefore orexin A content was studied in the PKU mice. Hypothalamus and brain stem had higher levels of orexin A compared to cerebrum and cerebellum both in wild type and PKU mice brains as observed by radioimmunoassay method. Interestingly, all these regions of the brain in PKU mouse showed a higher level of orexin A compared to the wild type. Heart and plasma also had higher levels of orexin A in PKU compared to the wild type. Immunohistochemical analysis revealed an increased number of orexin A–stained cells in the brain and heart of PKU mouse compared to the wild type. This is the first report of increased level of orexin in the PKU mouse brain. Hyperactivity is commonly observed in children with PKU; thus these findings suggest that orexin A is a contributing factor for the hyperactivity.  相似文献

Marine invertebrate sperm proteins are particularly interesting because they are characterized by positive selection and are likely to be involved in prezyogotic isolation and, thus, speciation. Here, we present the first survey of interspecific and intraspecific variation of a bivalve sperm protein among a group of species that regularly hybridize in nature. M7 lysin is found in sperm acrosomes of mussels and dissolves the egg vitelline coat, permitting fertilization. We sequenced multiple alleles of the mature protein-coding region of M7 lysin from allopatric populations of mussels in the Mytilus edulis species group (M. edulis, M. galloprovincialis, and M. trossulus). A significant McDonald-Kreitman test showed an excess of fixed amino acid replacing substitutions between species, consistent with positive selection. In addition, Kolmogorov-Smirnov tests showed significant heterogeneity in polymorphism to divergence ratios for both synonymous variation and combined synonymous and nonsynonymous variation within M. galloprovincialis. These results indicate that there has been adaptive evolution at M7 lysin and, furthermore, show that positive selection on sperm proteins can occur even when postzygotic reproductive isolation is incomplete.  相似文献
