Mapping of citrullinated fibrinogen B-cell epitopes in rheumatoid arthritis by imaging surface plasmon resonance

Introduction  

Rheumatoid arthritis (RA) frequently involves the loss of tolerance to citrullinated antigens, which may play a role in pathogenicity. Citrullinated fibrinogen is commonly found in inflamed synovial tissue and is a frequent target of autoantibodies in RA patients. To obtain insight into the B-cell response to citrullinated fibrinogen in RA, its autoepitopes were systematically mapped using a new methodology.     

Flux Analysis of Glucose Metabolism in Rhizopus oryzae for the Purpose of Increasing Lactate Yields

A flux analysis of glucose metabolism in the filamentous fungus Rhizopus oryzae was achieved using a specific radioactivity curve-matching program, TFLUX. Glycolytic and tricarboxylic acid cycle intermediates labeled through the addition of extracellular [U-14C]glucose were isolated and purified for specific radioactivity determinations. This information, together with pool sizes and the rates of glucose utilization and end product production, provided input for flux maps of the metabolic network under two different experimental conditions. Based upon the flux analysis of this system, a mutant of R. oryzae with higher lactate and lower ethanol yields than the parent was sought for and found.     

Stimulation by thyrotropin of horse thyroid plasma membranes adenylate cyclase: evidence of cooperativity

Horse thyroid plasma membranes were prepared by partition in an aqueous two phases system. The membrane fraction was enriched in adenylate cyclase and only slightly contaminated with mitochondria and lysosomes. Adenylate cyclase activity was stimulated by TSH and PGE₁. The TSH stimulatory effect was nearly immediate and occured in the same range of concentrations that activates intact cell metabolism. It was potentiated by GTP and ITP. A quantitative analysis of the data suggests that activation of thyroid adenylate cyclase by TSH is a cooperative process.
