When mature Protosiphon cells were placed in darkness, zoospore production was more extensive and was completed in a shorter time at a temperature of 27 C than at 22 or 15 C. Cool-white fluorescent (Sylvania) light inhibited the process measurably at a radiation intensity of 0.6±103 ergsjcm2-sec; inhibition was 96% complete at 14±103 ergs/cm2-sec. For mature cells previously grown under repeated 12-12 hr light-dark cycles, a dark period of approximately 2 hr at 22 C allowed cell division to proceed to a stage such that reillumination did not inhibit continued development of zoospores. Monochromatic light from 402 to approximately -494 nm, as compared to darkness, inhibited zoospore formation; maximal inhibition was at 432-461 nm. In contrast, monochromatic light from 522 to 726 nm stimulated zoospore formation relative to darkness. Synchronous zoospore production was obtained using the following regimes: (A) 12 hr cool-white alternated with 12 hr yellow, (B) 12 hr cool-white alternated with 12 hr blue. Under regime A synchronous zoospore release (following synchronous production) occurred near the end of the yellow irradiation period, while under regime B it occurred near the end of the cool-white irradiation period. Experiments have explored the possible relationships between the flagellar surface motility of chlamydomonas, visualized as translocation of polystyrene beads by paralyzed (pf) mutants (Bloodgood, 1977, J. Cell Biol. 15:983-989), and the capacity of gametic flagella to participate in the mating reaction. While vegetative and gametic flagella bind beads with equal efficiencies and are capable of transporting them along entire flagellar lengths, beads on vegetative flagella are primarily associated with the proximal half of the flagella whereas those of gametic flagella exhibit no such preference. This difference may relate to the "tipping" response of gametes during sexual flagellar agglutination (Goodenough and Jurivich, 1978, J. Cell Biol. 79:680-693). Colchicine, vinblastine, chymotrypsin, cytochalasins B and D, and anti-β-tubulin antiserum are all able to inhibit the binding of beads to the flagellar suface. Trysin digestion and an antiserum directed against whole chlamydomonas flagella have no effect on the ability of flagella to bind beads, but the beads remain immobile. These results suggest that at least two flagellar activities participate in surface motility: (a) bead binding, which may involve a tubulin-like component at the flagellar surface; and (b) bead translocation, which may depend on a second component (e.g. an ATPase) of the flagellar surface. Surface motility is shown to be distinct from gametic adhesiveness per se, but it may participate in concentrating dispersed agglutinins, in driving them toward the flagellar tips, and/or in generating a signal-to-fuse from the flagellar tips to the cell body. Directly supporting these concepts is the observation that bound beads remain immobilized at the flagellar tips during the "tip-locking" stage of pf x pf matings, and the observation that bound ligands such as antibody fail to be tipped by trypsinized flagella. 相似文献

The influence of environmental heterogeneity on components of male and female fitness is examined using Atriplex canescens growing on steep slopes and alluvium at the slope base as a model system. Female fitness is estimated as the grams of fruit produced per plant and the grams of fruit per gram leaf tissue. Male fitness is estimated as the grams of stamens produced, the number of pollen grains dispersed to a given distance, and the potential number of grams of fruit sired taking into consideration the number and distribution of mates and competing pollen donors. The influence of increased plant size on male and female fitness components, the cost of reproduction (as measured by biomass, joules and nitrogen) on a gross level and a per offspring basis are also examined. The results indicate that the female function is more limited on the slope than the male function. The efficiency of pollen dispersal (the number of pollen grains per unit donor plant volume dispersed to a given distance) is enhanced by growing on slopes. Males become less efficient at dispersing pollen as they increase in size, while the efficiency of female reproduction (grams of fruit per volume or gram leaf tissue) is unaffected by increasing plant size. The cost to a male of siring a gram of fruit is about the same as the cost to a female of producing the gram of fruit. Implications for the evolution of sexual lability and dioecy are discussed. 相似文献
