The ranges of arctic-alpine species have shifted extensively with Pleistocene climate changes and glaciations. Using sequence data from the trnH-psbA and trnT-trnL chloroplast DNA spacer regions, we investigated the phylogeography of the widespread, ancient (>3 million years) arctic-alpine plant Oxyria digyna (Polygonaceae). We identified 45 haplotypes and six highly divergent major lineages; estimated ages of these lineages (time to most recent common ancestor, T(MRCA)) ranged from ~0.5 to 2.5 million years. One lineage is widespread in the arctic, a second is restricted to the southern Rocky Mountains of the western United States, and a third was found only in the Himalayan and Altai regions of Asia. Three other lineages are widespread in western North America, where they overlap extensively. The high genetic diversity and the presence of divergent major cpDNA lineages within Oxyria digyna reflect its age and suggest that it was widespread during much of its history. The distributions of individual lineages indicate repeated spread of Oxyria digyna through North America over multiple glacial cycles. During the Last Glacial Maximum it persisted in multiple refugia in western North America, including Beringia, south of the continental ice, and within the northern limits of the Cordilleran ice sheet. Our data contribute to a growing body of evidence that arctic-alpine species have migrated from different source regions over multiple glacial cycles and that cryptic refugia contributed to persistence through the Last Glacial Maximum.  相似文献

The distributions of cell volumes of steady-state Escherichia coli ML30G cultures at various temperatures were measured. For cultures in a minimal medium, the distributions were indistinguishable at several temperatures between 15 and 30 C; at higher temperatures the cells were slightly smaller, and at lower temperatures they were slightly larger. For cultures in a complex medium, the cells were slightly larger at both high and low temperatures of growth. An abrupt change of temperature within the middle range led to a transient change in the distribution of cell volume, suggesting that the size of dividing cells is well regulated. No synchrony of division was induced by a change in temperature.  相似文献

Using preference-assessment tests with humans in conjunction with behavioral modification sessions has been a regular component of almost all operant conditioning programs with mentally challenged humans. This has been very effective in improving the efficiency of behavioral training in these settings and could be similarly effective in zoological and research environments. This study investigated the preferences of 9 captive orangutans for different food items. The study used a pairwise presentation to record each nonhuman animal's preferences for 5 different foods on 6 different occasions over the course of 6 months. Results of a Friedman's 2-way ANOVA indicated that the orangutans showed a clear overall preference for apple. However, there was significant variability among different orangutans in preference ranking for the 5 foods, as shown by a Kendall's tau. In addition, there was variability in preference rankings across time for each orangutan. Because the orangutans' preferences change over time and vary according to individual, regular assessments should identify items to be used as rewards in behavioral husbandry training or as part of feeding enrichment strategies.  相似文献

Testican is a putative extracellular heparan/ chondroitin sulfate proteoglycan of unknown function that is expressed in a variety of human tissues at widely different levels but is most abundant in the brain. In mice, testican mRNA has been detected only in brain and it is therefore likely to have an important function in the central nervous system. RNA blot analysis reveals the relative intensity of testican in various regions of the human brain. Levels of testican message are most pronounced in the thalamus, hippocampus, occipital lobe, nucleus accumbens, temporal lobe, and caudate nucleus, with somewhat lower levels in the cerebral cortex, medulla oblongata, frontal lobe, amygdala, putamen, spinal cord, substantia nigra, and cerebellum. In situ hybridization reveals the cellular distribution of the mRNA within these areas to be highest in neurons and in choroid plexus epithelium, and moderately lower in ependymal cells lining the ventricles and in vascular endothelial cells. Testican mRNA is not detected in oligodendrocytes or in most astrocytes. However, astrocytes in regions of reactive gliosis do express testican mRNA. These findings, along with a cysteine-rich pattern similarity to neurocan, brevican, versican, and other proteoglycans found in brain, suggest that testican may be a part of the specialized extracellular matrix of the brain.  相似文献

The natural separation maintained by microfluidic flows is employed as the basis of a particle/cell sorting device. This method of separating particulate suspensions exploits the inherent laminar nature of microscale fluid dynamics and incorporates applied fields and image cytometry to enable sorting based upon any visually identifiable difference between colloid-sized cells or particles. This technique may be used to easily isolate, separate, sort, or enrich virtually any suspension of microscale biological or colloidal particles within a microfluidic system. The entire footprint of the device described here is less than 0.01 mm(2), allowing it to be readily incorporated within highly integrated micro total analysis systems (microTAS).  相似文献

Abstract

Metabolic responses to auxin. VII. Effects on glycolysis. — The detection of changes the of level of respiratory substrates could provide an indication on the site of action of auxin. The effects of indoleacetic acid on the contents in several glycolysis intermediates in isolated segments from the growing part of the third internode of etiolated pea seadlings were thus investigated.

Hexose phosphates, triose phosphates, phospho-enol-pyruvate, pyruvate and ethanol were determined enzymatically in the neutralized perchloric extracts from groups of segments incubated for 3 hours in the presence or in the absence of 5 × 10 ?5 M indoleacetic acid. No significant difference appeared between the results on the perchloric extracts and those obtained after fractionation with barium and alcohol, according to Umbreit et al.

Acetaldehyde was measured as by Klotz. Pyruvate was determined also as a phenyl-hydrazone, according to Cavallini and Frontali; the same method was used for the determination of α-ketoglutarare and of an unknown compound, showing a chromatographic behavior similar to that of an uronic acid.

Auxin induced a modest but significant increase of hexose-6-phosphates (16%) and of fructose di-phosphate (38%). For glucose-1-phosphate and triose phosphates — present at very low concentration — no significant changes could be detected, α-ketoglutarate level appeared unchanged. The compound behaving as an uronic acid increased, in the auxin treated segments, by more than 100%.

A very definite response to auxin appeared at the level of pyruvate. acetaldehyde and alcohol. Pyruvate quite reproducibly increased by 60%, and acetaldehyde production was more than doubled. In contrast, alcohol decreased by about 40%.

These results are discussed in relation to the previous data on the effects of auxin on the oxidative metabolism, and, particularly, to the findings of an increased rate of CO-sensitive respiration and of the rise of the ATP, TPNH and red. glutathione levels in the auxin treated pea internode segments.  相似文献

Mediterranean-climate regions (med-regions) are global hotspots of endemism facing mounting environmental threats associated with human-related activities, including the ecological impacts associated with non-native species introductions. We review freshwater fish introductions across med-regions to evaluate the influences of non-native fishes on the biogeography of taxonomic and functional diversity. Our synthesis revealed that 136 freshwater fish species (26 families, 13 orders) have been introduced into med-regions globally. These introductions, and local extirpations, have increased taxonomic and functional faunal similarity among regions by an average of 7.5% (4.6–11.4%; Jaccard) and 7.2% (1.4–14.0%; Bray–Curtis), respectively. Faunal homogenisation was highest in Chile and the western Med Basin, whereas sw Cape and the Aegean Sea drainages showed slight differentiation (decrease in faunal similarity) over time. At present, fish faunas of different med-regions have widespread species in common (e.g. Gambusia holbrooki, Cyprinus carpio, Oncorhynchus mykiss, Carassius auratus, and Micropterus salmoides) which are typically large-bodied, non-migratory, have higher physiological tolerance, and display fast population growth rates. Our findings suggest that intentional and accidental introductions of freshwater fish have dissolved dispersal barriers and significantly changed the present-day biogeography of med-regions across the globe. Conservation challenges in med-regions include understanding the ecosystem consequences of non-native species introductions at macro-ecological scales.  相似文献
