Abstract: Because of increasing concerns about puma (Puma concolor) attacks on people and the desire to minimize dangerous puma-human encounters while conserving puma populations, we examined spatial and temporal relationships between pumas and people that used Cuyamaca Rancho State Park (CRSP), California, USA. From 2001 to 2003, we studied 10 adult pumas outfitted with Global Positioning System collars. Although number of visitors to CRSP was increasing, no dangerous puma-human encounters were reported during our study. Male and female pumas typically moved short distances during the day (mean of means of individual hourly movements = 168 m and 131 m each hr, respectively) and moved the most at night (mean of means = 690 m and 390 m each hr, respectively). Of 10 pumas, 9 were least active during the day and most active during the evening or at night. In contrast, most visitor use of trails (x̄ = 85%) occurred during the day. Based on puma and human activity patterns, risk of a puma-human encounter was greatest during the evening. Puma prey caches were randomly distributed in relation to trails and park facilities; however, 8 of 33 caches were still within 100 m of a trail and 2 were within 300 m of a facility. Individual puma behavior relative to human activity areas was variable. Some pumas appeared to temporally avoid human activity areas; others used the park randomly in relation to human activity areas; none appeared to be attracted to human activity areas. Pumas that did not show detectable responses to human activity may have been exhibiting some level of habituation; if so, this level of habituation did not result in puma-human conflicts. When human activity peaked during the day, adult male and female pumas were within 100 m of a trail an average of 9% and 19% of the time they were located in the park, respectively. Thus, there were opportunities for puma-human encounters. Management personnel can take a proactive approach to deal with puma-human interactions through education and protocols that help to minimize probability of conflicts; this may provide the best chance for a continued puma presence in habitat used by pumas and people.  相似文献

Methods for the specific detection of Bacillus spores are needed in many situations such as the recognition of food poisoning. This study presents an experimental design in order to find the best combination of germination conditions leading to a rapid and detectable fluorescent in situ hybridization (FISH) signal from Bacillus cereus spores present in pure cultures and milk samples.
B. cereus ATCC 14579 and HER 1414 were incubated in 20 different growth media by using a combination of various germinants such as sugars, amino acids and dipicolinic acid. Also, three different germination factors were tested: incubation temperature, inoculum concentration and a heat shock treatment. Permeabilization procedure and hybridization time were optimized on the best germination condition found. B. cereus -specific FISH probes were validated under the optimized condition and in detection of spiked B. cereus spores in 1% ultra heat-treated milk samples. FISH-labeled cells were detected by using flow cytometry, and the results were confirmed by fluorescence microscopy. The optimal condition allows the detection of B. cereus spores in less than 2 h. Overall, a ninefold reduction in total time for detection was achieved when comparing with previous works. Therefore, the permeabilization and hybridization optimizations mentioned in this study are major improvements for the detection time of B. cereus spores.

PRACTICAL APPLICATIONS


By using the optimized conditions of germination/outgrowth, permeabilization and hybridization, the detection of 103 cfu/mL of Bacillus cereus spores using fluorescent in situ hybridization is possible within 2 h in milk sample.  相似文献

ABSTRACT We examined the effect of harvesting intensity and pattern on red squirrels (Tamiasciurus hudsonicus), northern flying squirrels (Glaucomys sabrinus), and yellow-pine chipmunks (Tamias amoenus) in mature inland Douglas-fir (Pseudotsuga menziesii glauca) forests in south-central British Columbia, Canada. We sampled squirrels 1 year before harvesting through 4 years after harvesting and estimated population parameters using open-population models. Relative to unharvested stands, each of the 3 species showed a strong response to tree removal. From 2 years to 4 years after logging, red squirrel density was 40% (SE = 7.1) lower in stands with 50% basal-area tree removal. From 1 year and up to 4 years after logging, northern flying squirrel density averaged 60% (SE = 5.2) lower in harvested treatments regardless of intensity or pattern of logging. In contrast, density of yellow-pine chipmunks increased markedly with increased logging intensity. Beginning 3 years after logging, yellow-pine chipmunk density was 734% (SE = 269) greater in stands with 50% basal-area tree removal. In the short term, harvesting intensity was a more important determinant of squirrel density than harvesting pattern. Retaining >10 m2 per ha of live residual stand structure in mature inland Douglas-fir forests maintained habitat for forest-dependent species such as red squirrels and northern flying squirrels, albeit at lower densities.  相似文献
