In order to complement ecological information with genetic data we isolated and characterized 14 polymorphic microsatellite markers from raccoons (Procyon lotor). Three multiplexed panels comprising the loci were developed and 29 individuals from a contiguous habitat patch in northern Indiana, USA were genotyped. The number of alleles per locus ranged from four to 18, and overall heterozygosities ranged from 0.31 to 1.00. One locus was identified as possibly being X‐linked, since males appeared to be hemizygous. Data generated using these markers will be used to further our understanding of small‐scale raccoon population dynamics in a highly fragmented landscape. 相似文献

Lystrosaurus is one of the few therapsid genera that survived the end-Permian mass extinction, and the only genus to have done so in abundance. This study identifies which species of Lystrosaurus have been recovered from Permian and Triassic strata to determine changes in the species composition across the Permo–Triassic (P–T) boundary in the Karoo Basin of South Africa. Data generated from museum collections and recent fieldwork were used to stratigraphically arrange a total of 189 Lystrosaurus specimens to determine which species survived the extinction event. Results reveal that L. curvatus and L. maccaigi lived together on the Karoo floodplains immediately before the extinction event. L. maccaigi did not survive into the Triassic in South Africa. L. curvatus survived, but did not flourish and soon became extinct. Two new species of Lystrosaurus , L. murrayi and L. declivis , appeared in the Early Triassic. It is possible that L. murrayi and L. declivis occupied different niches to L. maccaigi and L. curvatus , and had special adaptations that were advantageous in an Early Triassic environment. We suggest that L. maccaigi may be used as a biostratigraphic marker to indicate latest Permian strata in South Africa and that, in support of previous proposals, the genus Lystrosaurus should not be used as a sole indicator of Triassic-aged strata. Effects of xanthene dyes on mycelial growth and conidial germination in three species of entomopathogenic fungi, Beauveria bassiana, Metarhizium anispoliae and Paecilomyces fumosoroseus, were evaluated in a variety of assay systems. In a disk-diffusion assay, erythrosin B and phloxine B (but not eosin B) produced zones of inhibition in colonies of all three species under continuous exposure to light at disk-loadings of 100mug. None of the dyes produced zones of inhibition in the absence of light at disk loadings of 100mug. Both erythrosin B and phloxine B inhibited mycelial growth of all three species in the light in a dose-dependent manner. Weaker dose-responses for inhibition of growth in the dark were observed for some fungus/dye combinations. Erythrosin B, tested singly, completely inhibited conidial germination in the light in all eight fungal strains tested at 100mug ml-1 medium, but failed to inhibit conidial germination in any of these strains in the dark at the same concentration of dye. For single strains of each of the three fungi, erythrosin B and phloxine B inhibited conidial germination in a dose-dependent manner in the light with IC50s < 6.2mug dye ml-1 medium for all fungus/dye combinations. Phloxine B was a more potent inhibitor of germination than erythrosin B for all three fungal species. At fixed dosages of erythrosin B and phloxine B, inhibition of conidial germination in all three species increased with time of exposure to light. These results constitute the first quantitative demonstration of photodynamic inhibition of conidial germination in fungi by xanthene dyes. 相似文献

We evaluated some products and protocols designed for reliable RNA extraction from minute tissue samples and safe tissue storage at room temperature without RNA degradation. Success of RNA retrieval was compared for varying amounts of tissue (3, 5, 10 hair follicles), stored at different temperatures (room temperature, ?20 °C) for variable durations (1, 3, 6, 12 weeks). We also compared two RNA isolation kits specialized for small samples. RNA was successfully retrieved from as few as 3 hairs stored at room temperature for up to 6 weeks, suggesting the potential for gene expression analyses on minimally invasive samples from natural populations. 相似文献

Despite only limited Pleistocene glacial activity in the southern hemisphere, temperate forest species experienced complex distributional changes resulting from the combined effects of glaciation, sea level change and increased aridity. The effects of these historical processes on population genetic structure are now overlain by the effects of contemporary habitat modification. In this study, 10 microsatellites and 629 bp of the mitochondrial control region were used to assess the effects of historical forest fragmentation and recent anthropogenic habitat change on the broad-scale population genetic structuring of a southern temperate marsupial, the Tasmanian pademelon. A total of 200 individuals were sampled from seven sites across Tasmania and two islands in Bass Strait. High mitochondrial and nuclear genetic diversity indicated the maintenance of large historical population sizes. There was weak phylogeographical structuring of haplotypes, although all King Island haplotypes and three Tasmanian haplotypes formed a divergent clade implying the mid-Pleistocene isolation of a far northwestern population. Both the mitochondrial and nuclear data indicated a division of Tasmanian populations into eastern and western regions. This was consistent with a historical barrier resulting from increased aridity in the lowland 'midlands' region during glacial periods, and with a contemporary barrier resulting from recent habitat modification in that region. In Tasmania, gene flow appears to have been relatively unrestricted during glacial maxima in the west, while in the east there was evidence for historical expansion from at least one large glacial refuge and recolonization of Flinders Island. 相似文献

ABSTRACT Effects of silvicultural activities on birds are of increasing interest because of documented national declines in breeding bird populations for some species and the potential that these declines are in part due to changes in forest habitat. Silviculturally induced disturbances have been advocated as a means to achieve suitable forest conditions for priority wildlife species in bottomland hardwood forests. We evaluated how silvicultural activities on conservation lands in bottomland hardwood forests of Louisiana, USA, influenced species-specific densities of breeding birds. Our data were from independent studies, which used standardized point-count surveys for breeding birds in 124 bottomland hardwood forest stands on 12 management areas. We used Program DISTANCE 5.0, Release 2.0 (Thomas et al. 2006) to estimate density for 43 species with >50 detections. For 36 of those species we compared density estimates among harvest regimes (individual selection, group selection, extensive harvest, and no harvest). We observed 10 species with similar densities in those harvest regimes compared with densities in stands not harvested. However, we observed 10 species that were negatively impacted by harvest with greater densities in stands not harvested, 9 species with greater densities in individual selection stands, 4 species with greater densities in group selection stands, and 4 species with greater densities in stands receiving an extensive harvest (e.g., >40% canopy removal). Differences in intensity of harvest influenced densities of breeding birds. Moreover, community-wide avian conservation values of stands subjected to individual and group selection, and stands not harvested, were similar to each other and greater than that of stands subjected to extensive harvest that removed >40% canopy cover. These results have implications for managers estimating breeding bird populations, in addition to predicting changes in bird communities as a result of prescribed and future forest management practices. 相似文献
