The ornate shrew (Sorex ornatus) is restricted to the vanishing wetlands of California, USA and Baja California, Mexico. Several subspecies of ornate shrews are considered ‘mammal species of special concern’ in California by the Department of Fish and Game, and one (Sorex ornatus relictus) has recently been listed as endangered. Populations of shrews around Buena Vista Lake have been diminished or extirpated due to habitat deterioration and human development. In order to study the patterns of genetic variation in isolated populations of Buena Vista Lake shrews, we developed 10 polymorphic microsatellite loci. There were 6–27 alleles per locus, and the loci had heterozygosity values that ranged from 20 to 80%. In addition, we screened 20 different populations of S. ornatus, eight species within two subfamilies of shrews (Soricinae and Crocidurinae), as well as in a mole (Talpidae, Neurotrichus gibbsii), to determine if these loci could be informative in other species as well. 相似文献

Microsatellite data are widely used to test ecological and evolutionary hypotheses in wild populations. In this paper, we consider three typical sources of scoring errors capable of biasing biological conclusions: stuttering, large‐allele dropout and null alleles. We describe methods to detect errors and propose conventions to mitigate scoring errors and report error rates in studies of wild populations. Finally, we discuss potential bias in ecological or evolutionary conclusions based on data sets containing these scoring errors. 相似文献

Polyploidy (whole‐genome duplication) has contributed significantly to angiosperm evolution and diversification. To date, it has been found that most polyploids are the result of multiple formation events, which may contribute to genetic diversity and affect interfertility among polyploid lineages of independent origin. A recently discovered allotetraploid derivative of Mimulus guttatus and M. nasutus, Mimulus sookensis, is found throughout the valleys of western Oregon and Vancouver Island. Here, we analyse the patterns of nucleotide diversity at three chloroplast and six nuclear loci in M. guttatus, M. nasutus and M. sookensis, to gain insight into the formation of M. sookensis. By analysing the patterns of genetic variation seen in the diploid progenitors in comparison with the variation seen in M. sookensis, we are able to show that M. sookensis has recurrently formed. We also observed that most M. sookensis individuals are fixed heterozygotes at all of the nuclear loci examined, suggesting that duplicate gene loss is not extensive in M. sookensis. To assess the possibility that hybridization among M. sookensis has contributed to genetic diversity, we conducted crossing experiments within M. sookensis. We found that M. sookensis of independent origin are highly interfertile, suggesting that crossing barriers do not exist within M. sookensis, and that hybridization among M. sookensis may result in new recombinant genotypes. Together, the data suggest that although recurrent origins may be common, they can contribute to genetic diversity without contributing to reproductive isolation among independently arisen polyploid lineages. 相似文献

Abstract In March 2000, Canada lynx (Lynx canadensis) were listed as a federally threatened species in 14 states at the southern periphery of their range, where lynx habitat is disjunct and snowshoe hare (Lepus americanus) densities are low. Forest conditions vary across lynx range; thus, region-specific data on the habitat requirements of lynx are needed. We studied lynx in northern Maine, USA, from 1999 to 2004 to assess quality and potential for forests in Maine to sustain lynx populations. We trapped and radiocollared 43 lynx (21 M, 22 F) during this period and evaluated diurnal habitat selection by 16 resident adult lynx (9 M, 7 F) monitored in 2002. We evaluated lynx selection of 8 habitats at multiple spatial scales, and related lynx habitat selection to snowshoe hare abundance. Lynx preferred conifer-dominated sapling stands, which supported the highest hare densities on our study site (x̄ = 2.4 hares/ha), over all other habitats. The habitats where lynx placed their home ranges did not differ by sex. However, within their home ranges, males not only preferred conifer-dominated sapling stands, but also preferred mature conifer, whereas females singularly preferred conifer-dominated sapling stands. Approximately one-third of Maine's spruce-fir forest and nearly 50% of our study area was regenerating conifer or mixed-sapling forest, resulting from a disease event and intensive forest management (e.g., large clear-cuts). Our findings suggest that current habitat conditions in Maine are better than western montane regions and approach conditions in boreal forests during periods of hare abundance. We recommend that forest landowners maintain a mosaic of different-aged conifer stands to ensure a component of regenerating conifer-dominated forest on the landscape. 相似文献

1. A 2 × 2 factorial design was employed to look at the influence of two levels of phosphorus (P; high and low) and two levels of light (high and low) and their interactions, on phytoplankton abundance, elemental tissue composition and community structure in two seasons (April and June) of 2005. 2. A novel feature of the experiment was the creation of high and low P levels by manipulating sediment core conditions in the laboratory. Sediment cores were incubated with their associated overlaying water column from four different sites in Mona Lake, Michigan, under anaerobic or aerobic conditions, respectively. 3. After 24 days, the water overlaying the sediment cores was collected and used as growth media for phytoplankton collected from Mona Lake. Phytoplankton communities were grown in the laboratory in the high or low P water, and subjected to high (250 μmol m?2 s?1) or low (10 μmol m?2 s?1) light for 9 (April) or 14 (June) days. 4. In the April experiment, high P treatments resulted in significantly higher chlorophyll‐a concentrations, significantly lower C : P ratios from two of the four sites, and greater dominance by Scenedesmus at all sites relative to low P treatments. High light generally led to higher chlorophyll‐a concentrations, higher C : P ratios and greater Scenedesmus and Fragilaria biovolume at all sites. A significant interaction was measured between P and light for chlorophyll‐a and Scenedesmus biovolume, suggesting the influence of P was more apparent at high light than at low light levels. 5. In the June experiment, high P increased ash‐free dry mass (AFDM), lowered C : P ratios and resulted in increased Pediastrum biovolume. High light levels led to greater chlorophyll‐a concentrations, AFDM and C : P ratios, as well as increased biovolumes of Scendesmus, Pediastrum and Fragilaria. A significant interaction was found between P and light for all three taxa, as the positive influence of P was more pronounced at high light levels. 6. The results of our study demonstrate that sediment‐derived P stimulates phytoplankton growth, but that its effect on phytoplankton dynamics is modulated by other factors, such as light. 相似文献

Spatial and seasonal variation in nutrient excretion by benthic invertebrates in a eutrophic reservoir 总被引:8,自引:0,他引:8
1. Nitrogen (N) and phosphorus (P) fluxes via excretion by benthic invertebrates were quantified in a eutrophic reservoir (Acton Lake, Ohio, U.S.A.). We quantified variation in nutrient fluxes seasonally (June until November 1997), spatially (three sites) and among taxa (chironomids, tubificid oligochaetes and Chaoborus). 2. The three taxa differed in spatial distribution and contribution to nutrient fluxes. Tubificids were the most abundant taxon at two oxic sites (1.5 and 4 m depth), and were exceedingly rare at an anoxic, hypolimnetic site (8 m). Chironomids were abundant only at the shallowest oxic site. Chaoborus was the only abundant taxon at the anoxic site. Total benthic invertebrate biomass was greatest at the shallowest site and lowest at the anoxic, hypolimnetic site. 3. Mass‐specific excretion rate [μmol NH4–N or soluble reactive P (SRP) excreted mg dry mass–1 h–1] varied among experiments and was influenced by temperature. Differences among taxa were not significant. Thus, nutrient flux through benthic invertebrates was affected more by total invertebrate biomass and temperature than by species composition. 4. Fluxes of N and P via benthic invertebrate excretion (μmol NH4–N or SRP m–2 day–1) were greatest at the oxic sites, where fluxes were dominated by the excretion of tubificids and chironomids. The N and P fluxes at the anoxic site were much lower, and were dominated by excretion by Chaoborus. The ratio of N and P excreted by the benthic invertebrate assemblage varied seasonally and was lowest at the anoxic site. 5. Comparison with other measured inputs shows that excretion by benthic invertebrates could be an important source of nutrients, especially of P. In order to complement ecological information with genetic data we isolated and characterized 14 polymorphic microsatellite markers from raccoons (Procyon lotor). Three multiplexed panels comprising the loci were developed and 29 individuals from a contiguous habitat patch in northern Indiana, USA were genotyped. The number of alleles per locus ranged from four to 18, and overall heterozygosities ranged from 0.31 to 1.00. One locus was identified as possibly being X‐linked, since males appeared to be hemizygous. Data generated using these markers will be used to further our understanding of small‐scale raccoon population dynamics in a highly fragmented landscape. 相似文献
