Ullucus virus C (UVC) is a comovirus prevalent in Ullucus tuberosus grown at high altitudes in the Bolivian and Peruvian Andes. It was transmitted mechanically to U. tuberosus (Basellaceae) and to five of 26 species from three of eight other families, infecting U. tuberosus symptomlessly but inducing conspicuous systemic infection in Chenopodium amaranticolor and C. quinoa. Sap from infected C. quinoa was usually infective after 10 min at 70 but not 75 °C, after dilution to 10-7 but not 10-8, and after 8 but not 16 wk at 20 °C. UVC was not transmitted by either of two aphid species (Aphis gossypii and Myzus persicae) or through seed of C. quinoa, but it was transmitted by leaf contact between infected and healthy plants. UVC has isometric particles which, in neutral phosphotungstate, are c. 28 nm in diameter. The particles sediment as three components (T, M and B) with sedimentation coefficients (s?20, w) of 51 S (T), 95 S (M) and 116 S (B). M component particles have a buoyant density (g cm-3) in caesium chloride of 1.404, and B component particles separated into minor and major sub-components with densities of 1.409 and 1.463, respectively. T, M and B particles were serologically indistinguishable, and each contained similar relative amounts of two polypeptides of mol. wts 20 700 and 45 100. T particles contained only protein, but M particles also contained c. 30% ss-RNA of mol. wt 1–45 ×106 and B particles c. 38% ss-RNA of mol. wt 2·2 × 106. The virus is serologically distantly related to cowpea mosaic virus but, as it showed no relationship to any of 11 other similar viruses, it is probably a distinct member of the comovirus group.  相似文献

Predicting how insect crop pests will respond to global climate change is an important part of increasing crop production for future food security, and will increasingly rely on empirically based evidence. The effects of atmospheric composition, especially elevated carbon dioxide (eCO2), on insect herbivores have been well studied, but this research has focussed almost exclusively on aboveground insects. However, responses of root‐feeding insects to eCO2 are unlikely to mirror these trends because of fundamental differences between aboveground and belowground habitats. Moreover, changes in secondary metabolites and defensive responses to insect attack under eCO2 conditions are largely unexplored for root herbivore interactions. This study investigated how eCO2 (700 μmol mol?1) affected a root‐feeding herbivore via changes to plant growth and concentrations of carbon (C), nitrogen (N) and phenolics. This study used the root‐feeding vine weevil, Otiorhynchus sulcatus and the perennial crop, Ribes nigrum. Weevil populations decreased by 33% and body mass decreased by 23% (from 7.2 to 5.4 mg) in eCO2. Root biomass decreased by 16% in eCO2, which was strongly correlated with weevil performance. While root N concentrations fell by 8%, there were no significant effects of eCO2 on root C and N concentrations. Weevils caused a sink in plants, resulting in 8–12% decreases in leaf C concentration following herbivory. There was an interactive effect of CO2 and root herbivory on root phenolic concentrations, whereby weevils induced an increase at ambient CO2, suggestive of defensive response, but caused a decrease under eCO2. Contrary to predictions, there was a positive relationship between root phenolics and weevil performance. We conclude that impaired root‐growth underpinned the negative effects of eCO2 on vine weevils and speculate that the plant's failure to mount a defensive response at eCO2 may have intensified these negative effects.  相似文献

Nitrous oxide emissions from a cropped soil in a semi-arid climate   总被引:5,自引:0,他引:5  
Understanding nitrous oxide (N2O) emissions from agricultural soils in semi‐arid regions is required to better understand global terrestrial N2O losses. Nitrous oxide emissions were measured from a rain‐fed, cropped soil in a semi‐arid region of south‐western Australia for one year on a sub‐daily basis. The site included N‐fertilized (100 kg N ha?1 yr?1) and nonfertilized plots. Emissions were measured using soil chambers connected to a fully automated system that measured N2O using gas chromatography. Daily N2O emissions were low (?1.8 to 7.3 g N2O‐N ha?1 day?1) and culminated in an annual loss of 0.11 kg N2O‐N ha?1 from N‐fertilized soil and 0.09 kg N2O‐N ha?1 from nonfertilized soil. Over half (55%) the annual N2O emission occurred from both N treatments when the soil was fallow, following a series of summer rainfall events. At this time of the year, conditions were conducive for soil microbial N2O production: elevated soil water content, available N, soil temperatures generally >25 °C and no active plant growth. The proportion of N fertilizer emitted as N2O in 1 year, after correction for the ‘background’ emission (no N fertilizer applied), was 0.02%. The emission factor reported in this study was 60 times lower than the IPCC default value for the application of synthetic fertilizers to land (1.25%), suggesting that the default may not be suitable for cropped soils in semi‐arid regions. Applying N fertilizer did not significantly increase the annual N2O emission, demonstrating that a proportion of N2O emitted from agricultural soils may not be directly derived from the application of N fertilizer. ‘Background’ emissions, resulting from other agricultural practices, need to be accounted for if we are to fully assess the impact of agriculture in semi‐arid regions on global terrestrial N2O emissions.  相似文献

The rate constants for diffusion of THO across the red cell membrane of beef and dog, and the rate of entrance of water into the erythrocytes of these species under an osmotic pressure gradient have been measured. For water entrance into the erythrocyte by diffusion the rate constants are 0.10 ± 0.02 msec.-1 (beef) and 0.14 ± 0.03 msec.-1 (dog); the permeability coefficients for water entrance under a pressure gradient of 1 osmol./cm3 are 0.28 See PDF for Equation These values permit the calculation of an equivalent pore radius for the erythrocyte membrane of 4.1 A for beef and 7.4 A for dog. In the beef red cell the change in THO diffusion due to osmotically produced cell volume shifts has been studied. The resistance to THO diffusion increases as the cell volume increases. At the maximum volume, (1.06 times normal), THO diffusion is decreased to 0.84 times the normal rate. This change in diffusion is attributed to swelling of the cellular membrane.  相似文献

The fate of all eggs laid by Guillemots Uria aalge in seven study areas on the Isle of May was recorded in 12 consecutive seasons. The average success at each site was related to its physical characteristics, bird density, position on the cliff and availability of suitable habitat for ticks. The numbers of neighbours and walls, type of site, slope where the egg was incubated and distance from the top of the cliff all had a significant effect on both hatching and breeding success. Potential tick habitat had no significant effect. Much variation in hatching and breeding success remained unexplained. Sites used when the colony was less than half its current size and most frequently in the period 1984–1995 were the most successful. Since an average Guillemot pair remains together at a site for only 3–4 years. the consistently high success of many sites over a period of 10–12 years suggests that the most used sites were occupied by a succession of high-quality birds.  相似文献

Abstract Three manipulations of the male contribution to copulation were used to investigate what determines the switch-off of female receptivity that follows mating in Lucilia cuprina. Multiple matings by males led to a reduction in their effectiveness at switching-off females: the first twelve matings caused more than 50% of females to be switched-off 1 day after mating, while the first four matings caused more than 50% of females to be switched-off for 7 days after mating. Males mated up to twenty-three times and on average ten times during 10 h of continuous access to virgin females. The number of sperm transferred declined logarithmically through a series of matings as did the quantity of material transferred from the male accessory gland (measured as radiolabeled material) through the first six matings. Overnight isolation of multiply-mated males led to considerable recovery of their ability to switch-off females, and to limited renewal of their ability to transfer sperm. Castrated males transferred similar quantities of accessory gland material to females as did sham-operated males during their first five matings and in their first three matings switched-off 95% of females for 1 day but only 48% for 7 days. When normal mating pairs were separated at increasing intervals after coupling, an increasing proportion of females were switched-off at 1 or 7 days afterward. 7 days after 2 min matings no females were switched-off though 40% of the number of sperm transferred in a full mating had been transferred by this time. The proposed explanation for these data is that both the initial switch-off and its duration are determined by the quantity of a receptivity-inhibiting substance that normally enters the female haemo-lymph after being injected by the male into the wall of the bursa copulatrix. It is proposed that when castrated males mate, an absence of sperm results in most accessory gland secretion entering the empty spermathecae (rather than the wall of the bursa as usually occurs) and hence being unable to reach the haemolymph and exert its influence. The effective dose of the receptivity-inhibiting substance is measured on a logarithmic scale.  相似文献

Three pupfish (Cyprinodon) morphotypes (two endemic) occur in some of the young (6000 ypb) saline lakes on the Bahamian island of San Salvador. The ‘normal’ morph, a detritivore/omnivore, is not different in its general features from Cyprinodon variegatus from other Bahamian islands. ‘Bulldog’ is a scale‐eater/piscivore that preys upon normal pupfish, and ‘bozo’ is a specialized molluskivore. Reproductive isolation among these morphs is not predicted by the evolutionary biology of congeneric species because sympatry of even morphogically and ecologically quite divergent pupfishes has usually resulted in hybridization/introgression. Survey of variation at eight microsatellite loci reveals that sympatric normal and bulldog populations are genetically distinctive by several criteria, and are therefore likely reproductively isolated. The bulldog morph in Crescent Pond is markedly divergent from those in Little Lake and Osprey Lake, a finding consistent with, although it does not prove, separate parallel origins of this morphotype. The data also suggest that the bulldogs in the latter two lakes did not evolve by intralacustrine speciation from the current sympatric normal populations. Some of the genetic data suggest that the bozo morph may also be reproductively isolated from the other two pupfishes, but only a small, pooled sample of this rare morphotype was available, and the issue is not resolved. Isolating mechanisms between bulldog and normal morphs are of special interest because of the possibility that they arose as a consequence of a predator–prey relationship. The mature ovaries accounted for about 25% of dry weight and 30% of the protein content of wild-type (anautogenous) females of L. cuprina which had fed ad lib. on liver. The protein content of gravid females was 50% greater than at emergence. Protein ingested during the adult stage, therefore, plays an important role in ovarian maturation. The protein content of the ovaries of females fed measured amounts of liver exudate was from 37 to 52% of the amount of protein ingested.

Only limited ovarian development occurred in females fed only protein (high MW fraction of liver exudate or bovine serum albumin). The presence, in addition, of low MW components (low MW fraction of liver exudate or a salt mixture) was necessary for ovarian maturation. Quantitative feeding showed that the high and low MW fractions of liver exudate were, respectively, superior to bovine serum albumin and the salt mixture, which was based on the cation analysis of the low MW fraction of the exudate.  相似文献
