Characterization of large basking shark Cetorhinus maximus aggregations in the western North Atlantic Ocean

Cetorhinus maximus aggregations recorded during extensive aerial survey efforts off the north‐eastern United States between 1980 and 2013 included aggregations centring on sightings with group sizes of at least 30 individuals. These aggregations occurred in summer and autumn months and included aggregation sizes of up to 1398 individuals, the largest aggregation ever reported for this species. The aggregations were associated with sea surface temperatures of 13–24° C and chlorophyll‐a concentrations of 0·4–2·6 mg m?3 and during one aggregation, a high abundance of zooplankton prey was present. Photogrammetric tools allowed for the estimation of total body lengths ranging between 4 and 8 m. Characterization of these events provides new insight into the potential biological function of large aggregations in this species. 相似文献

Sugars, particularly trehalose and sucrose, are used to stabilize liposomes during hydration (freeze-drying and air-drying). As a result, dry liposomes are trapped in a sugar glass, a supersaturated and thermodynamically unstable solid solution. We investigated the effects of the glassy state on liposome fusion and solute retention in the dry state. Solute leakage from dry liposomes was extremely slow at temperatures below the glass transition temperature (Tg); however, it increased exponentially as temperature increased to near or above the Tg, indicating that the glassy state had to be maintained for dry liposomes to retain trapped solutes. The leakage of solutes from dry liposomes followed the law of first-order kinetics and was correlated linearly with liposome fusion. The kinetics of solute leakage showed an excellent fit with the Arrhenius equation at temperatures both above and below the Tg, with a transitional break near the Tg. The activation energy of solute leakage was 1320 kJ/mol at temperatures above the Tg, but increased to 1991 kJ/mol at temperatures below the Tg. The stabilization effect of sugar glass on dry liposomes may be associated with the elevated energy barrier for liposome fusion and the physical separation of dry liposomes in the glassy state. The half-life of solute retention in dry liposomes may be prolonged by storing dry liposomes at temperatures below the Tg and by increasing the Tg of the dry liposome preparation. 相似文献

A large-subunit mitochondrial ribosomal DNA sequence translocated to the nuclear genome of two stone crabs (Menippe) 总被引:1,自引:0,他引:1
Two DNA sequences that appear to be homologous to large-subunit
mitochondrial ribosomal RNA genes have been identified in the stone crabs
Menippe mercenaria and M. adina. Amplification from whole genomic DNA by
polymerase chain reaction (PCR) with oligonucleotide primers based on
conserved portions of large-subunit mitochondrial rRNA genes consistently
amplified two products of similar length (565 and 567 bp). These products
differed at 3% of their nucleotide bases, and could be distinguished by a
HindIII site. Only one of these sequences (designated the A sequence) was
detected by PCR in purified mitochondrial DNA. The other (designated the B
sequence) hybridized to total genomic DNA at a level consistent with a
nuclear genome location. It is unlikely that the type B product would have
been recognized as a nuclear copy by examination of its sequence alone.
This is the first report of a mitochondrial gene sequence translocated into
the nuclear genome of a crustacean.

The combination of passive drug permeability, affinity for uptake and efflux transporters as well as gastrointestinal metabolism defines net drug absorption. Efflux mechanisms are often overlooked when examining the absorption phase of drug bioavailability. Knowing the affinity of antimalarials for efflux transporters such as P-glycoprotein (P-gp) may assist in the determination of drug absorption and pharmacokinetic drug interactions during oral absorption in drug combination therapies. Concurrent administration of P-gp inhibitors and P-gp substrate drugs may also result in alterations in the bioavailability of some antimalarials. In-vitro Caco-2 cell monolayers were used here as a model for potential drug absorption related problems and P-gp mediated transport of drugs. Artemisone had the highest permeability at around 50 x 10−6 cm/sec, followed by amodiaquine around 20 x 10−6 cm/sec; both mefloquine and artesunate were around 10 x 10−6 cm/sec. Methylene blue was between 2 and 6 x 10−6 cm/sec depending on the direction of transport. This 3 fold difference was able to be halved by use of P-gp inhibition. MRP inhibition also assisted the consolidation of the methylene blue transport. Mefloquine was shown to be a P-gp inhibitor affecting our P-gp substrate, Rhodamine 123, although none of the other drugs impacted upon rhodamine123 transport rates. In conclusion, mefloquine is a P-gp inhibitor and methylene blue is a partial substrate; methylene blue may have increased absorption if co-administered with such P-gp inhibitors. An upregulation of P-gp was observed when artemisone and dihydroartemisinin were co-incubated with mefloquine and amodiaquine. 相似文献

The unidirectional influx of cycloleucine into in vitro preparations of gill tissue of the mussel, Mytilus californianus, was determined. Influx was found to be linear for at least an hour, and the kinetics of cycloleucine influx conformed to Michaelis-Menten type kinetics. The transport mechanism(s) for cycloleucine is relatively specific for the L-enantiomorph of neutral amino acids, and is capable of accumulating cycloleucine to intracellular concentrations much higher than those of the surrounding medium. Evedence is presented that the transport of amino acids by gill tissue plays a significant role in whole animal nutrition. 相似文献

A typing scheme has recently been developed for Neisseria meningitidis serogroup A based on the clonal population structure of these bacteria. An international strain collection consisting of 423 group A strains isolated from 23 epidemics or outbreaks since 1963, as well as from older epidemics and numerous non-epidemic situations was used in the analysis. Strains were first segregated into electrophoretic types, depending on the combined score for the electrophoretic mobilities of 7 cytoplasmic isoenzymes resolved by starch gel electrophoresis and of 2 outer membrane proteins resolved by sodium dodecyl sulphate polyacrylamide gel electrophoresis. The bacteria were subsequently assigned to one of 21 clones after numerical analysis of their electrophoretic types.The epidemiological value of the typing scheme was assessed by examining case and carrier strains isolated during (1982–83) and subsequent to (1984–85) an epidemic in the Gambia, West Africa. The case isolates, all of which were serogroup A, were of a single clonal type. All serogroup A carrier isolates were also of this clone, while carrier strains of other serogroups showed greater clonal diversity. These results indicate that case strains during an epidemic show little clonal diversity and thus that the typing scheme is of value in distinguishing the etiology of epidemics.A retrospective epidemiological analysis of the strains in the international collection showed that most serogroup A epidemics were associated with a single or predominant clone, although some epidemics were of mixed etiology. The survey included 256 isolates from 15 African epidemics since 1963, a period which covers 3 major epidemic waves (1960–63; 1967–73 and 1981–83), thus permitting a detailed epidemiological analysis of serogroup A epidemics in this continent.Epidemiological records indicate that seven clones have been responsible for sets of epidemics throughout the world since 1915 and that at least two of these sets can be considered to represent mutually exclusive pandemics, linking numerous epidemics between 1967–75 and 1973–83, respectively. 相似文献

Summary Effects of forest plant species composition and physiognomy on bird and invertebrate communities were investigated in three discrete, relatively undisturbed forest types along a dry-wet soil moisture gradient. Using discriminant function analysis, a 100% floristic and a 78% vegetation structural discrimination were obtained between the three forest types. However, the bird communities of these different forest types were very similar in species composition, and had much lower densities than those normally encountered in other, superficially similar forests. Although an 81% discrimination between forest types was attained through analysis of ground surface invertebrates, measures of litter and aerial invertebrate abundance were also of limited use as discriminators. Historical and biogeographic factors, as well as the low nutritional levels in the soil and vegetation may be the causes of low bird and invertebrate density and diversity. It is concluded that floristics and vegetation structure have, at best, a minor influence on bird community structure, and possibly also on invertebrate community structure in the Knysna Forest. 相似文献

A dynamic model is developed to describe the swing phase of the hindlimb of a normally walking horse. The limb was represented by four rigid segments constrained to move in a sagittal plane only. The mathematical equations of motion of this four-element pendulum were formulated using Lagrange's theorem. The morphometric parameters from the hindlimb segments of 3 horses were determined using high-speed film analysis. Five muscle groups were incorporated in the model. Muscle activity was derived from earlier EMG measurements. Optimization of muscle moments resulted in a simulated swing movement that approximated that in the living animal. 相似文献

We have studied the calorimetric and infrared spectroscopic properties of the amino acid proline which has been implicated in the stabilization of biomacromolecules during reduced water states. It has been suggested that the ability of this molecule to protect biomacromolecules during these stress states may be related to the formation of polymeric aggregates of proline monomers in solution. The structure of this aggregate is thought to be an alternates stack, forming a hydrophilic colloid-like polymer which is thought to interact with hydrophobic moieties of biomacromolecules, reducing the exposed hydrophobic area during reduced water conditions. Calorimetric data presented in this work show that in increasing concentration of proline in solution the enthalpy associated with the melting of bulk water is greatly reduced, indicating strong hydrogen bonding character of proline in aqueous solution. Proline shows two eutectic phase separations at moderate concentrations and one of these eutectics may be the proposed intermolecular state. A partial phase diagram for proline is presented. Fourier-transform infrared spectroscopic data indicate that the COO- asymmetric stretch of proline shows marked splitting with increasing proline concentration. This suggests that the carboxylate is in different environments, with the high energy vibrations representing COO- groups which are participating in the hydrogen bonding pattern associated with the formation of the intermolecular stack. Changes in the CH2 asymmetric and symmetric stretches of the pyrrolidine rings of proline are consistent with the proposed stack structure. We also suggest a possible mechanism by which these intermolecular associations may be important in the protection of biomacromolecules during reduced water states. 相似文献
