Abstract. The ultrastructure of paraspermatogenesis is examined in the littorinid subfamily Littorininae, with special emphasis on Littoraria (Palustorina) articulata (PHILIPPI 1846). In particular the study focuses on the fate of the nucleus and origin of the rod bodies during parasperm development. Parasperm of the Littorininae are rounded or oblong cells, which undergo an abortive meiosis and eliminate part of the nucleus but often retain a nuclear remnant. The cytoplasm is filled with numerous spherical vesicles in all Littorininae, but in Littoraria (and in certain species of Nodilittorina, Tectarius and Cenchritis) dense 'rod-bodies' also occur. Littoraria (Palustorina) are unique in possessing a flagellum-like structure termed the 'pseudotrich", which lacks an axoneme but contains microtubules during its development. Paraspermatogonia differ from euspermatogonia in the structure of the nucleus and in the extensive rough endoplasmic reticulum (RER) and swollen cytoplasm. Two types of secretions develop in Littoraria : (1) numerous, spherical granules (composed of putative glycoprotein, also seen in other Littorininae) and (2) rhomboid granules (composition uncertain but reacting positively to RNA stains; these granules arising within RER cisternae close to the nucleus). As the rhomboid granules fuse to form the larger, rod-bodies (polygonal in cross section), the RER membrane enclosing the rod-bodies becomes confluent with the outer nuclear membrane, thereby forming a common compartment. Results of this study clearly show that the rod-bodies are secretions of the RER cisternae and not, as claimed in some light microscopic accounts, the product of fusion of eusperm nuclei which have entered the parasperm cytoplasm (either by active eusperm penetration or by phagocytosis). Developmental characteristics of littorinid parasperm show differences between species and may, in some cases, provide characters diagnostic of subgenera.  相似文献

The effect of dominance on the mating behaviour of both females and males was investigated for the communally breeding pukeko (Porphyrio porphyrio melanotus). Pukeko are polygynandrous with most groups consisting of three to 7 breeding males and one or two breeding females that lay eggs in a single nest. Linear dominance hierarchies are well established within breeding groups. Alpha females were involved in more copulations than beta females, but this difference did not appear to be a result of dominance status. There was also no evidence of egg tossing or egg destruction by breeding females. There was no positive correspondence between dominance rank of males and their frequency of sexual activity or copulations during the prelaying or laying periods. Dominants did interrupt matings by subordinates, but both qualitative and quantitative analyses of behaviour found no evidence of mate guarding. Furthermore dominants were no more likely to interrupt a copulation involving a subordinate male than they were to passively observe it or not react at all. This led us to suggest that males tended to behave indifferently toward mating competitors. As a result, multiple paternity is predicted to be high in pukeko groups. We also question the standard interpretation that males are behaving cooperatively in communal groups rather than coactively.  相似文献

Trends in coral cover are widely used to indicate the health of coral reefs but are costly to obtain from field survey over large areas. In situ studies of reflected spectra at the coral surface show that living and recently dead colonies can be distinguished. Here, we investigate whether such spectral differences can be detected using an airborne remote sensing instrument. The Compact Airborne Spectrographic Imager (Itres Research Ltd, Canada) was flown in two configurations: 10 spectral bands with 1-m2 pixels and 6 spectral bands with 0.25-m2 pixels. First, we show that an instrument with 10 spectral bands possesses adequate spectral resolution to distinguish living Porites, living Pocillopora spp., partially dead Porites, recently dead Porites (total colony mortality within 6 months), old dead (>6 months) Porites, Halimeda spp., and coralline red algae when there is no water column to confuse spectra. All substrata were distinguished using fourth-order spectral derivatives around 538 nm and 562 nm. Then, at a shallow site (Tivaru) at Rangiroa Atoll, Tuamotu Archipelago (French Polynesia), we show that live and dead coral can be distinguished from the air to a depth of at least 4 m using first- and fourth-order spectral derivatives between 562–580 nm. However, partially dead and recently dead Porites colonies could not be distinguished from an airborne platform. Spectral differences among substrata are then exploited to predict the cover of reef substrata in ten 25-m2 plots at nearby Motu Nuhi (max depth 8 m). The actual cover in these plots was determined in situ using quadrats with a 0.01-m2 grid. Considerable disparity occurred between field and image-based measures of substrate cover within individual 25-m2 quadrats. At this small scale, disparity, measured as the absolute difference in cover between field and remote-sensing methods, reached 25% in some substrata but was always less than 10% for living coral (99% of which consisted of Porites spp.). At the scale of the reef (all ten 25-m2 quadrats), however, disparities in percent cover between imagery and field data were less than 10% for all substrata and extremely low for some classes (e.g. <3% for living Porites, recently dead Porites and Halimeda). The least accurately estimated substrata were sand and coralline red algae, which were overestimated by absolute values 7.9% and 6.6%, respectively. The precision of sampling was similar for field and remote-sensing methods: field methods required 19 plots to detect a 10% difference in coral cover among three reefs with a statistical power of 95%. Remote-sensing methods required 21 plots. However, it took 1 h to acquire imagery over 92,500 m2 of reef, which represents 3,700 plots of 25 m2 each, compared with 3 days to survey 10 such plots underwater. There were no significant differences in accuracy between 1-m2 and 0.25-m2 image resolutions, suggesting that the advantage of using smaller pixels is offset by reduced spectral information and an increase in noise (noise was observed to be 1.6–1.8 times greater in 0.25-m2 pixels). We show that airborne remote sensing can be used to monitor coral and algal cover over large areas, providing that water is shallow and clear, and that brown fleshy macroalgae are scarce, that depth is known independently (e.g. from sonar survey).  相似文献

Background  

Hematopoietic development in vertebrate embryos results from the sequential contribution of two pools of precursors independently generated. While intra-embryonic precursors harbour the features of hematopoietic stem cells (HSC), precursors formed earlier in the yolk sac (YS) display limited differentiation and self-renewal potentials. The mechanisms leading to the generation of the precursors in both sites are still largely unknown, as are the molecular basis underlying their different potential. A possible approach to assess the role of candidate genes is to transfer or modulate their expression/activity in both sites. We thus designed and compared transduction protocols to target either native extra-embryonic precursors, or hematopoietic precursors.  相似文献

Phenological shifts are the most widely reported ecological responses to climate change, but the requirements to distinguish their causes (i.e. phenotypic plasticity vs. microevolution) are rarely met. To do so, we analysed almost two decades of parturition data from a wild population of North American red squirrels (Tamiasciurus hudsonicus). Although an observed advance in parturition date during the first decade provided putative support for climate change‐driven microevolution, a closer look revealed a more complex pattern. Parturition date was heritable [h2 = 0.14 (0.07–0.21 (HPD interval)] and under phenotypic selection [β = ?0.14 ± 0.06 (SE)] across the full study duration. However, the early advance reversed in the second decade. Further, selection did not act on the genetic contribution to variation in parturition date, and observed changes in predicted breeding values did not exceed those expected due to genetic drift. Instead, individuals responded plastically to environmental variation, and high food [white spruce (Picea glauca) seed] production in the first decade appears to have produced a plastic advance. In addition, there was little evidence of climate change affecting the advance, as there was neither a significant influence of spring temperature on parturition date or evidence of a change in spring temperatures across the study duration. Heritable traits not responding to selection in accordance with quantitative genetic predictions have long presented a puzzle to evolutionary ecologists. Our results on red squirrels provide empirical support for one potential solution: phenotypic selection arising from an environmental, as opposed to genetic, covariance between the phenotypic trait and annual fitness.  相似文献

Populations delineated based on genetic data are commonly used for wildlife conservation and management. Many studies use the program structure combined with the ΔK method to identify the most probable number of populations (K). We recently found K = 2 was identified more often when studies used ΔK compared to studies that did not. We suggested two reasons for this: hierarchical population structure leads to underestimation, or the ΔK method does not evaluate K = 1 causing an overestimation. The present contribution aims to develop a better understanding of the limits of the method using one, two and three population simulations across migration scenarios. From these simulations we identified the “best K” using model likelihood and ΔK. Our findings show that mean probability plots and ΔK are unable to resolve the correct number of populations once migration rate exceeds 0.005. We also found a strong bias towards selecting K = 2 using the ΔK method. We used these data to identify the range of values where the ΔK statistic identifies a value of K that is not well supported. Finally, using the simulations and a review of empirical data, we found that the magnitude of ΔK corresponds to the level of divergence between populations. Based on our findings, we suggest researchers should use the ΔK method cautiously; they need to report all relevant data, including the magnitude of ΔK, and an estimate of connectivity for the research community to assess whether meaningful genetic structure exists within the context of management and conservation.  相似文献

Contemporary patterns of genetic structure among fragmented populations can either result from historic patterns or arise from human-induced fragmentation. Use of historic samples collected prior to fragmentation allows for the origin of genetic structure to be established and appropriate management steps to be determined. In this study, we compare historic and contemporary levels of genetic diversity and structure of an endangered passerine, the New Zealand mohua or yellowhead (Mohoua ochrocephala), using nuclear microsatellites. We found that a significant amount of allelic richness has been lost over the last 100 years. Close to half of this was due to extinction of birds from entire regions, but almost as much was due to loss of genetic diversity within extant populations. We found a pattern of isolation by distance among contemporary populations, which could have resulted from historic structure due to limited gene flow along a latitudinal cline. However, we found that minimal genetic structure existed historically. The pattern of increased structure over time was confirmed by factorial correspondence analysis. We conclude that the genetic structure apparent today resulted from anthropogenic effects of recent fragmentation and isolation. We emphasize the importance of assessing genetic structure of populations prior to their fragmentation, when determining the significance of contemporary patterns. This study highlights the growing importance of museum specimens as a tool in the conservation of threatened and endangered species.  相似文献

Two methods for the characterization of protein molecular weights from their diffusion coefficients are discussed. These measurements can be made quickly and reliably at low concentrations using quasielastic light-scattering techniques. First, an empirical calibration of the diffusion coefficient at infinite dilution of denatured random coils against molecular weight is reported. The second method combines the measurement of D0 with the intrinsic viscosity [η]. This D0–[η] relationship proves to be very insensitive to polymers structure or solvent type. The data indicate that the ratio of the hydrodynamic radius measured by viscosity to the hydrodynamic radius measured by diffusion is about 15% smaller than that predicted by theoretical models. The nature of the molecular-weight average obtained for polydisperse systems is defined for a Schulz distribution. These hydrodynamic methods have also been used to demonstrate the presence of chain branching in the glycoprotein ovomucoid. In addition, a method is proposed by which the effective segment length and an excluded volume parameter for random coils may be evaluated for diffusion measurements.  相似文献

Treatment of Saccharomyces cerevisiae cells with low concentrations of either hydrogen peroxide or menadione (a superoxide-generating agent) induces adaptive responses which protect cells from the lethal effects of subsequent challenge with higher concentrations of these oxidants. Pretreatment with menadione is protective against cell killing by hydrogen peroxide; however, pretreatment with hydrogen peroxide is unable to protect cells from subsequent challenge with menadione. This suggests that the adaptive responses to these two different oxidants may be distinct.  相似文献
