Mammary glands from BALB/cfC3H midpregnant (9–11 days) mice were dissociated with collagenase and pronase, separated on a Percoll gradient, and the epithelial cells were cultured inside collagen gel. The cell number increased three-to five-fold when cultured for 6–8 days in DME/F12 (1: 1) medium containing 3% swine serum, insulin (10 μg/ml), cortisol (1.0 μg/ml), prolactin (10 μg/ml), transferrin (10 μg/ml), and epidermal growth factor (0.01 μg/ml). The casein level, as determined by radioimmunoassay, at the end of this growth phase, was much lower than that present in freshly dissociated cells. In order to stimulate casein production, the gels were released from the sides of the plastic dish and allowed to float for eight days in Waymouth's medium, containing insulin (10 μg/ml), cortisol (5 μg/ml), prolactin (10 μg/ml), and 0.25% bovine serum albumin. The casein level at the end of this differentiation phase was found to be comparable to that seen in the original freshly dissociated cells. Cells grown in DME/F12 (1: 1) medium containing 3% swine serum, insulin (10 μg/ml), and transferrin (10 μg/ml) were still capable of undergoing casein production, indicating that the presence of exogenous lactogenic hormones such as cortisol and prolactin, as well as exogenous growth factors such as epidermal growth factor, is not necessary during the growth phase for subsequent casein production during the differentiation phase. Two factors that seemed more important for subsequent casein stimulation were: (1) releasing collagen gels at the beginning of the differentiation phase, and (2) switching to'differentiation' medium. This present two-step protocol has allowed primary cultures of dissociated midpregnant mouse mammary epithelial cells to undergo several rounds of division inside a collagen gel matrix and to be subsequently stimulated to produce the mammary-specific protein, casein.  相似文献

We examined the hypothesis that the rate of bone growth limits the minimum fledging time of birds. Previous observations in California gulls indicate that linear growth of wing bones may be the rate limiting factor in wing development. If bone growth is rate limiting, then birds with relatively long bones for their size could be expected to have longer fledging periods than birds with relatively short bones. We tested this by comparing the length of wing bones, relative to body mass, to the relative length of fledging periods among 25 families. The results support the hypothesis. A strong correlation exists between relative fledging period and relative bone length. Species which have relatively long bones for their body size tend to take longer to fly. In contrast, parameters that influence flight style and performance, such as size of the pectoralis muscle and wing loading, show little or no correlation with fledging time. The analysis also indicates that, when altricial and precocial species are considered together, bone length is more highly correlated with fledging time than is body mass or rate of increase in body mass during growth. These observations suggest that linear growth of bones does limit the growth of avian wings and that it is one of the factors that influences the fledging time of birds.  相似文献

ABSTRACT White-tailed deer (Odocoileus virginianus) in North Dakota, USA, have greatly increased over the past 100 years due to conversion of prairie to agriculture, planting of tree rows, regulated hunting, and extirpation of large predators. In support of management to maintain harvestable big game while minimizing depredation damage to agriculture, the North Dakota Game and Fish Department manages wildlife management areas (WMAs) where planted trees and food plots provide habitat and public hunting. These WMAs are typically surrounded by agricultural fields, and management that concentrates deer in these areas may expose surrounding agricultural producers to increased depredation. We investigated diets, habitat use, and movements of white-tailed deer at a large WMA in central North Dakota (Lonetree WMA) to understand how the animals are responding to management designed to enhance wildlife populations. We also evaluated survival of white-tailed deer for an improved understanding of the recent trend for population growth. Diets of deer varied seasonally, including a relatively high proportion of crops from food plots in winter and mostly herbaceous forbs and browse during spring and summer. Data from radiocollared animals and biweekly spotlight surveys revealed that deer are being drawn from a relatively large area to Lonetree WMA during fall and winter. Hunting was the most important cause of mortality, but annual survival of adult and fawn females exceeded 80%. The original purpose of food plots at Lonetree WMA was to alleviate depredation on adjacent private lands. Depredation has been limited, but the multiyear trend of increased deer numbers is a new concern. A possible consequence of provisioning white-tailed deer with food plots during winter when some starvation would normally occur could be for the population to exceed a threshold above which regulated hunting will be unable to prevent irruptive growth. Research on how food plots influence winter survival is needed to inform management and prevent possible rapid increase in white-tailed deer populations across the prairie-coteau region of central North Dakota.  相似文献

Savannas comprise a large area of the global land surface and are subject to frequent disturbance through fire. The role of fire as one of the primary natural carbon cycling mechanisms is a key issue in considering global change feedbacks. The savannas of Northern Australia burn regularly and we aimed to determine their annual net ecosystem productivity (NEP) and the impact of fire on productivity. We established a long‐term eddy covariance flux tower at Howard Springs, Australia and present here 5 years of data from 2001 to 2005. Fire has direct impacts through emissions but also has indirect effects through the loss of productivity due to reduced functional leaf area index and the carbon costs of rebuilding the canopy. The impact of fire on the canopy latent energy exchange was evident for 40 days while the canopy was rebuilt; however, the carbon balance took approximately 70 days to recover. The annual fire free NEP at Howard Springs was estimated at −4.3 t C ha−1 yr−1 with a range of −3.5 to −5.1 t C ha−1 yr−1 across years. We calculated the average annual indirect fire effect as +0.7 t C ha−1 yr−1 using a neural network model approach and estimated average emissions of fine and coarse fuels as +1.6 t C ha−1 yr‐1. This allowed us to calculate a net biome production of −2.0 t C ha−1 yr‐1. We then partitioned this remaining sink and suggest that most of this can be accounted for by woody increment (1.2 t C ha−1 yr‐1) and shrub encroachment (0.5 t C ha−1 yr‐1). Given the consistent sink at this site, even under an almost annual fire regime, there may be management options to increase carbon sequestration by reducing fire frequency.  相似文献

Abstract: We designed and developed a vehicle-mounted very high frequency-based telemetry system that integrated an on-board antenna, receiver, electronic compass, Global Positioning System, computer, and Geographic Information System. The system allows users to accurately and quickly obtain fixes, estimate and confirm locations of radiomarked animals, and immediately record data into an electronic spreadsheet or database. The total cost of materials to build the system was $7,349 (United States currency). Mean error angle of 2.63 ± 12.1° (SD; range = −33.7–42.2°) and mean location error distance of 128 ± 91.3 m (SD; range = 0–408 m) suggested precision and accuracy of our system were comparable to other reported systems. Mean time to record 5 bearings/test transmitter was 6.28 ± 0.24 minutes (SE), which is the most efficient system reported to locate animals in the field. Vehicle-mounted telemetry systems like ours provide additional value to studies that involve tracking highly mobile species because investigators need not take bearings from established receiving stations and because investigators can immediately recognize bounced signals and take additional bearings and optimize accuracy of location estimates.  相似文献

Abstract: Double-crested cormorant (Phalacrocorax auritus) populations on the Great Lakes expanded greatly during the past 2 decades. On Lake Erie, the number of breeding cormorants increased from 174 birds (87 nests) in 1979 to 26,542 (13,271 nests) in 2000. In 2000, 81% of the breeding population was on 2 western-basin islands (East Sister and Middle Islands). The plant communities on these islands represent some of the last remnants of Carolinian vegetation in Canada. Our study is the first to quantitatively assess the relationship between the distribution of nesting cormorants and forest health. On East Sister Island, 2 measures of forest cover were obtained using infrared aerial photographs and ground-based measurements of leaf area index. These measures of forest cover were correlated (rs = 0.70, P < 0.001), which validated the use of remotely sensed data to assess forest cover. Cormorant nest density was negatively correlated with tree cover on both East Sister and Middle Islands. Temporal comparisons of Middle Island data indicated a reduction in tree cover from 1995 to 2001, and these reductions coincided with a large increase in the island's cormorant population. Although correlational in nature, our results suggest that cormorants may be detrimentally affecting island forests.  相似文献

ABSTRACT Until large numbers of bat fatalities began to be reported at certain North American wind energy facilities, wildlife concerns regarding wind energy focused primarily on bird fatalities. Due in part to mitigation to reduce bird fatalities, bat fatalities now outnumber those of birds. To test one mitigation option aimed at reducing bat fatalities at wind energy facilities, we altered the operational parameters of 21 turbines at a site with high bat fatalities in southwestern Alberta, Canada, during the peak fatality period. By altering when turbine rotors begin turning in low winds, either by changing the wind-speed trigger at which the turbine rotors are allowed to begin turning or by altering blade angles to reduce rotor speed, blades were near motionless in low wind speeds, which resulted in a significant reduction in bat fatalities (by 60.0% or 57.5%, respectively). Although these are promising mitigation techniques, further experiments are needed to assess costs and benefits at other locations.  相似文献

Reproductive structures of Frasnian (early Late Devonian) age from Wuhan, Hubei province, South China provide evidence for the early emergence of monomegaspory and integumentation in plants of non-progymnosperm affinity. Sphinxia wuhania gen. et sp. nov. pre-dates the occurrence of other reproductive structures exhibiting a similar suite of characters and adds support to the view that evolution towards an ovule may not have been a unique event. Sphinxia possesses a single functional megaspore with associated aborted members of the meiotic tetrad, a distinct, striated sporangial wall, and a sterile envelope bearing proximal spines. The morphology of the structure precludes assignment to any known group of plants in the absence of additional information regarding the parent plant. A progymnospermous affinity is unlikely. Thus, Sphinxia demonstrates that the Devonian progymnosperms did not lack potential competition from similarly specialized reproductive strategies in other plant lineages.  相似文献
