Effects of three antibiotics (cefotaxime, rifampicin and gentamicin) were tested on in vitro shoots cultures of tansy (Tanacetum vulgare L.)- These antibiotics were selected because endophytic bacteria isolated from the in vitro shoot cultures of tansy showed that all bacteria were Gram-negative and their growth was reduced by these three antibiotics. Five isolates were Enterobacteriaceae, three were fluorescent Pseudomonas, and two were aerobic bacteria. Increased concentrations of antibiotics caused usually linear or quadratic changes on the initiation of shoot growth, shoot number, growth rate, and shoot height. These changes and changes in pH of the culture media were tansy genotype-dependent following treatments with gentamicin. Also, the treatment with rifampicin or cefotaxime showed a genotype-dependent effect, because they resulted in significantly higher percentage of rooted plants in one of the three tansy genotypes tested. The growth rate and length of shoots were reduced in the media containing both gentamicin and rifampicin, but less so than in media containing both gentamicin and cefotaxime.  相似文献

Changes in soil carbon, the largest terrestrial carbon pool, are critical for the global carbon cycle, atmospheric CO2 levels and climate. Climate warming is predicted to be most pronounced in the northern regions and therefore the large soil carbon pool residing in boreal forests will be subject to larger global warming impact than soil carbon pools in the temperate or the tropical forest. A major uncertainty in current estimates of the terrestrial carbon balance is related to decomposition of soil organic matter (SOM). We hypothesized that when soils are exposed to warmer climate the structure of the ground vegetation will change much more rapidly than the dominant tree species. This change will alter the quality and amount of litter input to the soil and induce changes in microbial communities, thus possibly altering the temperature sensitivity of SOM decomposition. We transferred organic surface soil sections from the northern borders of the boreal forest zone to corresponding forest sites in the southern borders of the boreal forest zone and studied the effects of warmer climate after an adaptation period of 2 years. The results showed that initially ground vegetation and soil microbial community structure and community functions were different in northern and southern forest sites and that 2 years of exposure to warmer climate was long enough to cause changes in these ecological indicators. The rate of SOM decomposition was approximately equally sensitive to temperature irrespective of changes in vegetation or microbial communities in the studied forest sites. However, as temperature sensitivity of the decomposition increases with decreasing temperature regime, the proportional increase in the decomposition rate in northern latitudes could lead to significant carbon losses from the soils.  相似文献

The area under the cultivation of perennial bioenergy crops on organic soils in the northern countries is fast increasing. To understand the impact of reed canary grass (RCG, Phalaris arundinaceae L.) cultivation on the carbon dioxide (CO2) balance of an organic soil, net ecosystem CO2 exchange (NEE) was measured for four years in a RCG cultivated cutover peatland in eastern Finland using the eddy covariance technique. There were striking differences among the years in the annual precipitation. The annual precipitation was higher during 2004 and 2007 and lower during 2005 and 2006 than the 1971–2000 regional mean. During wet growing seasons, moderate temperatures, high surface soil moisture and low evaporative demand favoured high CO2 uptake. During dry seasons, owing to soil moisture and atmospheric stress, photosynthetic activity was severely restricted. The CO2 uptake [gross primary productivity (GPP)] was positively correlated with soil moisture, air temperature and inversely with vapour pressure deficit. Total ecosystem respiration (TER) increased with increasing soil temperature but decreased with increasing soil moisture. The relative responses of GPP and TER to moisture stress were different. While changes in TER for a given change in soil moisture were moderate, variations in GPP were drastic. Also, the seasonal variations in TER were not as conspicuous as those in GPP implying that GPP is the primary regulator of the interannual variability in NEE in this ecosystem. The ecosystem accumulated a total of 398 g C m?2 from the beginning of 2004 until the end of 2007. It retained some carbon during a wet year such as 2004 even after accounting for the loss of carbon in the form of harvested biomass. Based on this CO2 balance analysis, RCG cultivation is found to be a promising after‐use option on an organic soil.  相似文献

Fungi and Gram-negative bacteria were isolated from inside the roots of field-grown goat's rue (Galega orientalis). Fungi were isolated from three plants out of a total of 45 tested. Two multinuclear Rhizoctonia solani isolates were identified to the anastomosis group 5 (R. solani AG-5-Gal) using pairings with known AG test cultures. One fungal isolate was identified to Phoma chrysanthemicola. Gram-negative bacteria were isolated from three plants out of 25 tested. They were identified using classical methods, the BIOLOG identification system based on the utilisation of 95 different carbon sources, and the MIDI system for the analysis of whole cell fatty acids. The two latter systems were computer-associated and utilised an extensive reference library of isolates. One bacterial isolate was identified as Enterobacter agglomerans and two isolates as Pseudomonas marginalis. R. solani AG-5-Gal reduced the emergence of Lupinus luteus, L. polyphyllus and french bean (Phaseolus vulgaris) and the growth of broad bean (Viciafaba), L. luteus and french bean, but did not cause obvious damage in goat's rue and pea (Pisum sativum). However, R. solani AG-5-Gal was re-isolated from the roots of all the test plant species following inoculation. P. chrysanthemicola reduced the emergence of L. polyphyllus and the growth of goat's rue, french bean and broad bean, and it was re-isolated from all of the test plant species (except for french bean) following inoculation. All the bacteria reduced the emergence of french bean, but not that of goat's rue and pea, when applied to the soil. When the roots were dipped into bacterial suspension, all the bacteria damaged french bean and L. polyphyllus. Additionally, P. marginalis JV3 damaged goat's rue and red clover. The pathogenicity of the fungi and bacteria were not changed when they were double-inoculated in pairs, except for R. solani AG-5-Gal and P. marginalis JV2 which reduced the emergence of goat's rue when inoculated together but not when inoculated separately.  相似文献

Nuclear DNA content was determined in three accessions of Solanum brevidens, three accessions of S. etuberosum, and one accession of S. fernandezianum, which are diploid (2n = 2×= 24), closely related, non tuber-bearing wild potato species belonging to the series Etuberosa (Solanaceae). The plants were grown in vitro at 18°C or at 25°/22°C (day/night). S. brevidens was also grown in soil in the glasshouse at 25°/19°C (day/night), and in growth chambers at 18°C or 32°C. Leaf nuclei were isolated using a chopping method and stained with propidium iodide. Chicken red blood cells (CRBC; 2.33 pg) were added to the samples of nuclei as internal standards. The fluorescence of plant nuclei relative to CRBC was measured with an EPICS PROFILE flow cytometer. The 2C values of in vitro-grown S. brevidens and S. etuberosum were similar (1.48–1.54 pg, depending on the accession), but they were smaller than the 2C value of S. fernandezianum (1.63 pg). The 2C values of S. brevidens and S. etuberosum were generally smaller than those of the diploid species S. berthaultii (1.60–1.61 pg) and the diploid clones of S. tuberosum (1.60–1.72 pg). A similar relative difference of nuclear DNA content was found also between tetraploid S. brevidens and tetraploid S. tuberosum (2C = 3.15–3.16 pg and 3.50–3.62 pg, respectively). High (32°C) and low (18°C) growth temperatures caused abnormal changes in morphology and reduced fertility in S. brevidens in the growth chamber. The 2C values of S. brevidens grown at 25°/19°C (day/night) or at 32°C were similar, whereas the 2C values were c. 10% lower at 18°C.  相似文献

The accumulation of potato virus Y?(PVY?) and potato leaf roll virus (PLRV) was studied in plants of Solanum brevidens co-infected with each of six viruses or a viroid. Virus could not be detected by ELISA in plants of S. brevidens infected solely with PVY. However, accumulation of PVY was increased c. 1000-fold in plants doubly infected with tobacco mosaic virus or potato spindle tuber viroid (PSTVd). PVY titres in doubly infected plants of S. brevidens were between 1% and 0.1% of those found in the PVY-susceptible interspecific Solanum hybrid DTO-33. Double infections of 5. brevidens by PVY and alfalfa mosaic virus or potato viruses M, S, T or X did not significantly enhance PVY accumulation. Accumulation of PLRV was not enhanced in plants co-infected with any of the six viruses or PSTVd.  相似文献

The reasons for female desertion of offspring and the evolution of predominantly male care among monogamous bird species are not clearly understood. We studied parental effort during the incubation and chick rearing periods in the Eurasian Curlew Numenius arquata in western Finland, and compared timing of brood desertion with other populations in Europe. Males and females contributed equally to incubation and showed no differences in the intensity of mobbing behaviour towards a potential nest predator (stuffed crow) shortly after hatching. However, females deserted their offspring approximately halfway through the brooding period ( c. 16 d after hatching), while males remained with chicks until independence ( c. 35 d). Females with late-laid clutches deserted their offspring sooner after hatching than those with clutches produced earlier in the season. Curlew females deserted younger chicks in northeast Europe, where laying dates were later, breeding seasons shorter and migration distances were longer, than in western and central Europe. We suggest that the most likely reasons for offspring desertion by females may be associated with increased female survivorship and maintenance of pairbond between years.  相似文献

We show that sediment respiration is one of the key factors contributing to the high CO2 supersaturation in and evasion from Finnish lakes, and evidently also over large areas in the boreal landscape, where the majority of the lakes are small and shallow. A subpopulation of 177 randomly selected lakes (<100 km2) and 32 lakes with the highest total phosphorus (Ptot) concentrations in the Nordic Lake Survey (NLS) data base were sampled during four seasons and at four depths. Patterns of CO2 concentrations plotted against depth and time demonstrate strong CO2 accumulation in hypolimnetic waters during the stratification periods. The relationship between O2 departure from the saturation and CO2 departure from the saturation was strong in the entire data set (r2=0.79, n=2 740, P<0.0001). CO2 concentrations were positively associated with lake trophic state and the proportion of agricultural land in the catchment. In contrast, CO2 concentrations negatively correlated with the peatland percentage indicating that either input of easily degraded organic matter and/or nutrient load from agricultural land enhance degradation. The average lake‐area‐weighted annual CO2 evasion based on our 177 randomly selected lakes and all Finnish lakes >100 km2 ( Rantakari & Kortelainen, 2005 ) was 42 g C m?2 LA (lake area), approximately 20% of the average annual C accumulation in Finnish forest soils and tree biomass (covering 51% of the total area of Finland) in the 1990s. Extrapolating our estimate from Finland to all lakes of the boreal region suggests a total annual CO2 evasion of about 50 TgC, a value upto 40% of current estimates for lakes of the entire globe, emphasizing the role of small boreal lakes as conduits for transferring terrestrially fixed C into the atmosphere.  相似文献

Sweet potato feathery mottle virus (SPFMV, genus Potyvirus) is globally the most common pathogen of cultivated sweet potatoes (Ipomoea batatas; Convolvulaceae). Although more than 150 SPFMV isolates have been sequence‐characterized from cultivated sweet potatos across the world, little is known about SPFMV isolates from wild hosts and the evolutionary forces shaping SPFMV population structures. In this study, 46 SPFMV isolates from 14 wild species of genera Ipomoea, Hewittia and Lepistemon (barcoded for the matK gene in this study) and 13 isolates from cultivated sweet potatoes were partially sequenced. Wild plants were infected with the EA, C or O strain, or co‐infected with the EA and C strains of SPFMV. In East Africa, SPFMV populations in wild species and sweet potato were genetically undifferentiated, suggesting inter‐host transmission of SPFMV. Globally, spatial diversification of the 178 isolates analysed was observed, strain EA being largely geographically restricted to East Africa. Recombination was frequently detected in the 6K2‐VPg‐NIaPro region of the EA strain, demonstrating a recombination ‘hotspot’. Recombination between strains EA and C was rare, despite their frequent co‐infections in wild plants, suggesting purifying selection against strain EA/C recombinants. Positive selection was predicted on 17 amino acids distributed over the entire coat protein in the globally distributed strain C, as compared to only four amino acids in the coat protein N‐terminus of the EA strain. This selection implies a more recent introduction of the C strain and a higher adaptation of the EA strain to the local ecosystem. Thus, East Africa appears as a hotspot for evolution and diversification of SPFMV.  相似文献
